Human GH (hGH) has been available over 40 years for the treatment of children with GH deficiency. Human growth hormone (hGH) is mainly produced in the somatotrophic cells of the pituitary in brain and is the product of the GH-N gene. Among the edible mushrooms, the king oyster mushroom (Pleurotus eryngii ) is one of the most popular mushrooms in Asia, Europe and North America. The increasing popularity of P. eryngii among consumers is due to its flavor, texture and shelf life. We report a modified Agrobacteriummediated method for the efficient transformation of hGH2 in Pleurotus eryngii . The binary vector pCAMBIA1304 was used for the initial transformation and detected by GUS and GFP. Infiltrated samples transformed with pCAMBIA1304 showed a wider GUS response than the co-cultivated in the 50㎍/㎖ hygromycin selection medium. This transformation technique offers new prospects for the production of useful protein in the genetically improved mushroom.

Freesia is one of the most popular flowers over the world including Korea, due to the fragrance and beauty of the plant flower. The first domestic freesia cultivar ‘Shiny Gold’ was developed by NIHHS, RDA, in 2003, which has yellow double and large petals and strong fragrance. Ten years have passed since ‘Shiny Gold’ was cultivated at floral farms, and the deterioration of cut flower quality and yield are reported from the farms. Virus infection causes a reduction in the quantity and quality of the cut freesia flowers and is one of the most serious problems in Korea. Virus detection was carried by reverse transcription polymer chain reaction (RT-PCR) for FreMV, FreSV, BYMV, CMV, and TRV, as known to infect freesia. FreMV, FreSV, BYMV, and TRV were detected single or multiply, and CMV was not found in the freesia leaves collected from the farms. To produce virus-free freesia, meristem culture of ‘Shiny Gold’ was conducted in MS medium added ribavirin at different concentration. As the increased of ribavirin concentration, the growth of ‘Shiny Gold’ plantlets was inhibited in freesia’Shiny Gold’. The plantlets produced by meristem culture in ‘Shiny Gold’ were virus free at the enzyme-linked immunosorbent assay (ELISA) level.

In case of high-levelled platform in the urban railway system, a certain width of gap is inevitably formed between the train and the platform, and the gap gets wider when it is concerned with curved portion of the tracks at platform. This gap greatly contributes to decrease of passengers' safety while boarding and alighting a train. In particular, it endangers children's safety. Gap filler system is often fitted out to minimize gap and to overcome this safety problem at platform. However, as existing gap fillers are in most case structurally complex and organized to be powered from separate supply unit, there are a range of non-favorable points from the viewpoint of cost, installation and maintenance. In addition, existing gap fillers are due to cause disruptions in operation of train, with failures such as malfunctioning of control system. In this study, the authors will review on the gap filler applicable to the platforms of various height types, which neither requires any modifications of the train nor causes any operational disruptions. The proposed gap filler system here is a by-stage retractile type and based on the idea of interlocking it with train doors open-close mechanism, without any additional power supply unit or control system.

The bulb scales and shoot sections ($7\;\cal{mm}\;\times\;15\;\cal{mm}$) of Lilium oriental hybrid 'Casablanca' were cultured to compare bulblet growth in vitro. Shoots were induced from in vitro grown bulbscales on MS medium with $1.0\;\cal{mg/L}\;BA,\;0.5\;\cal{mg/L}$ IAA, and 30 g/L sucrose. The regenerated shoots were cut into shoot sections, and cultured on MS medium with $2.0\;\cal{mg/L}\;BA,\;0.5\;\cal{mg/L}$ IAA and 30 g/L sucrose for shoot proliferation. Culture of shoot sections stimulated bulblet growth significantly than the bulb scales on MS medium with 60 g/L sucrose. However, the bulblets from shoot sections did not reach ideal size to produce stems with several leaves. Therefore, liquid medium was added into the same vessels to stimulate bulblet growth further. After shoot sections were cultured on MS medium with 60 g/L sucrose and 2 g/L activated charcoal for two months in dark, $20\;\cal{ml}$ liquid media containing various concentrations of sucrose and MS salts were added. Two months later, the added liquid medium stimulated bulblet growth remarkably as compared to bulblets grown without added liquid medium. The added $25\;\cal{ml}$ liquid medium containing 120 g/L sucrose and double strength of MS salts were the most effective for growth of in vitro bulblets. More than $94\%$ bulblets produced by this method sprouted stems with several leaves after cold treatment at $5^{\circ}C$ for three months.
