Luteolin inhibits H2O2-induced cellular senescence via modulation of SIRT1 and p53

Luteolin, a sort of flavonoid, has been reported to be involved in neu-roprotective function via suppression of neuroinflammation. In this study, we in-vestigated the protective effect of luteolin against oxidative stress-induced cellular senescence and its molecular mechanism using hydrogen peroxide (H2O2)-induced cellular senescence model in House Ear Institute-Organ of Corti 1 cells (HEI-OC1). Our results showed that luteolin attenuated senescent phenotypes including alterations of morphology, cell proliferation, senescence-associated β-galactosidase expression, DNA damage, as well as related molecules expression such as p53 and p21 in the oxidant challenged model. Interestingly, we found that luteolin induces expression of sirtuin 1 in dose- and time-dependent manners and it has protective role against H2O2-induced cellular senescence by upregulation of sirtuin 1 (SIRT1). In contrast, the inhibitory effect of luteolin on cellular senescence under oxidative stress was abolished by silencing of SIRT1. This study indicates that luteolin effectively protects against oxidative stress-induced cellular senescence through p53 and SIRT1. These results suggest that luteolin possesses therapeutic potentials against age-related hearing loss that are induced by oxidative stress.

Purpose: The -1237T/C polymorphism of the Toll-like receptor 9 (TLR9) gene has been implicated in the susceptibility of inflammatorybowel diseases (IBDs), but the results remain conflicting. We further investigated this association via meta-analysis. Materials and Methods: Multiple electronic databases were extensively searched until February, 2015. The strength of associationwas evaluated by calculating the pooled odds ratios (ORs) and 95% confidence intervals (CIs). Results: A total of 2987 cases and 2388 controls from eight studies were analyzed. Overall, association was found between TLR9 -1237T/C polymorphism and the risk of IBDs when all the studies were pooled (recessive model, OR: 1.59, 95% CI: 1.02–2.47, p=0.04; homozygote comparison, OR: 1.62, 95% CI: 1.04–2.52, p=0.03; allele model, OR: 1.13, 95% CI: 1.00–1.27, p=0.05). Stratificationby ethnicity indicated an association between TLR9 -1237T/C polymorphism and IBDs risk in Caucasians (recessive model,OR: 1.59, 95% CI: 1.02–2.47, p=0.04; homozygote comparison, OR: 1.62, 95% CI: 1.04–2.52, p=0.03; allele model, OR: 1.12, 95% CI: 1.00–1.27, p=0.05). When stratified by disease type, significant correlation were only found in the Crohn’s disease subgroup (recessive model, OR: 1.69, 95% CI: 1.05–2.73, p=0.03; homozygote model, OR: 1.74, 95% CI: 1.07–2.82, p=0.02; allele model, OR: 1.15, 95% CI: 1.01–1.32, p=0.04). Conclusion: The present study suggested that the TLR9 -1237T/C polymorphism might act as a risk factor in the development of IBDs, particularly in Caucasians.
Upregulation of MicroRNA-34a Sensitizes Ovarian Cancer Cells to Resveratrol by Targeting Bcl-2

Purpose: Resveratrol (REV), a natural compound found in red wine, exhibits antitumor activity in various cancers, includingovarian cancer (OC). However, its potential anti-tumor mechanisms in OC are not well characterized. Here, we tried to elucidatethe underlying mechanisms of REV in OC cells. Materials and Methods: The anti-proliferative effects of REV against OC cells were measured using CCK-8 assay. Apoptosis wasmeasured using an Annexin V-FITC/PI apoptosis detection kit. The anti-metastasis effects of REV were evaluated by invasion assayand wound healing assay. The miRNA profiles in REV-treated cells were determined by microarray assay. Results: Our results showed that REV treatment suppresses the proliferation, induces the apoptosis, and inhibits the invasion andmigration of OV-90 and SKOV-3 cells. miR-34a was selected for further study due to its tumor suppressive roles in various humancancers. We found miR-34a overexpression enhanced the inhibitory effects of REV on OC cells, whereas miR-34a inhibition hadthe opposite effect in OC cells. In addition, we verified that BCL2, an anti-apoptotic gene, was found directly targeted by miR-34a. We also found that REV reduced the expression of Bcl-2 in OC cells. Further investigations revealed that overexpression of Bcl-2significantly abolished the anti-tumor effects of REV on OC cells. Conclusion: Overall, these results demonstrated that REV exerts anti-cancer effects on OC cells through an miR-34a/Bcl-2 axis,highlighting the therapeutic potential of REV for treatment of OC.

Four species of the genus Stibadocerella Brunetti, 1918 were previously known from the world. Here one new species, Stibadocerella shennongensis Zhang & Yang, sp. nov. is reported from Hubei, China. New record Stiba docerella pristina Brunetti, 1918 from Hainan, China is re-described and illustrated. A Hg2+-related quenching photoluminescence (PL) sensor was fabricated based on nitrogen-doped graphene quantum dots (N-GQDs) as the luminescent agent and glutathione as the masking agent to detect Hg2+ in tap water. The addition of Hg2+ significantly reduced the PL intensity of N-GQDs, which was attributed to coordination reaction inducing the aggregation of N-GQDs. The Hg2+-related quenching PL sensor with glutathione as the masking agent has good selectivity and accuracy. The sensor showed a linear relationship ranging from 0.5 to 110?nM with the detection limit 0.08?nM (S/N = 3). The proposed method was applied to the determination of Hg2+ in tap water, and the results were consistent with atomic absorption spectroscopy (AAS).

The paper focuses on investigating secure transmission in wireless powered communication networks (WPCN) that involve multiple energy-constrained relays and one energy-constrained source. The energy is harvested from a power beacon (PB) while operating in the presence of a passive eavesdropper. The study primarily aims to achieve energy-efficient secure communications by examining the impact of channel estimation on the secrecy performance of WPCN under both perfect and imperfect CSI scenarios. To obtain practical insights on improving security and energy efficiency, we propose closed-form expressions for secrecy outage probability (SOP) under the linear energy harvesting (LEH) model of WPCN. Furthermore, we suggest a search method to optimize the secure energy efficiency (SEE) with limited power from PB. The research emphasizes the significance of channel estimation in maintaining the desired performance levels in WPCN in real-world applications. The theoretical results are validated through simulations to ensure their accuracy and reliability.

1-Deoxynojirimycin (DNJ) as a kind of alkaloid has been confirmed that could modulate glycometabolism andhas toxicity for the eri-silkworm in our previous research. On the contrary, what is the potentially defensivemechanism when the DNJ enters the eri-silkworm. Based on comparative transcriptome sequencing, we foundthat ATP-binding cassette (ABC) transporter genes could be induced significantly by DNJ. In this study, a total of16 putative ABC transporter genes were identified, which can be classified into seven subfamilies, namely oneABCA, four ABCBs, three ABCCs, two ABCDs, one ABCE, three ABCFs, and two ABCGs. Phylogenetic analysisrevealed that ScABCs had strong conservation with Bombyx mori. Reverse transcription quantitative PCR (RTqPCR)suggested that 6 ABC transporters had a strong positive correlation between RT-qPCR and transcriptomedata. Additionally, S. c. ricini ABC transporter C-subfamily 4 (ScABCC4), S. c. ricini ABC transporter G-subfamily4 (ScABCG4), S. c. ricini ABC transporter A-subfamily 3 (ScABCA3) and S. c. ricini ABC transporter C-subfamily10 (ScABCC10) showed different expression pattern in two feed dose (1% and 2% DNJ) and three time points(6h, 12 h, 48 h). This study provides the first study on identification, characterization and expression patterns ofABC transporter genes in S. c. ricini response to DNJ, and lays a foundation for further understanding of theirphysiological roles response to the alkaloid.
Microstructure and cutting performance of CrTiAlN coating for high-speed dry milling

<P>Using a closed field unbalanced magnetron sputtering system, the cemented carbide end mills were coated with a CrTiAIN hard coating, which consisted of a Cr adhesive layer, a CrN interlayer and a CrTiAIN top layer. The microstructure and mechanical properties of the coating were investigated by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), micro indentation and scratch test. The cutting performance of the coated end mills were conducted by high-speed dry milling hardened steel (P20, HRC 45). The results indicates that the coating is composed of (Cr, Ti, Al)N columnar grains with nanolayers. The coating exhibits good adhesion to cemented carbide substrate and high microhardness of around 30 GPa. The coated end mills show significant improvement on tool life and much lower cutting force as compared to the uncoated ones. And the related mechanisms were discussed.</P>
