        This study was conducted to evaluate effect of ion exchange resins on growth of angelica (Angelica keiskei), and additionally investigated growth response of angelica according to germanium concentration in rhizosphere using the ion exchange resins containing germanium. As a result, the addition of the ion exchange resins induced significant increase of fresh weight and dry weight at 55 days of transplanting, and plant height and leaf area were also showed the similar pattern. Stem diameter and number of petioles were significantly higher in AS-62 and AS-64 treatment than in control. Inorganic elements concentration of angelica plants substantially didn’t show significant differences between control and treatments, but nitrogen, phosphorus, and potassium concentration were higher in treatments than in control. These results showed that the growth of angelica appeared to be promoted due to increased absorption rate of these elements caused by treatment of the ion exchange resins. There were no significant differences between control and treatments in the concentration of calcium, iron and boron, except for magnesium concentration of shoots. Germanium was not detected in the plants. In conclusion, the addition of ion exchange resin containing essential elements to soil showed positive effect on angelica growth.

      • <i>Vigna angularis</i> inhibits IL-6-induced cellular signalling and ameliorates collagen-induced arthritis

        <P><B>Objectives.</B> The present study was conducted in order to assess whether extracts or isolated compounds from <I>Vigna angularis</I> were able to suppress IL-6 signalling and to show the therapeutic effect on collagen-induced arthritis (CIA) in mice.</P><P><B>Methods.</B> The effect of <I>V. angularis</I> on IL-6 signalling was studied by measuring Stat3-dependent luciferase activity, expression of inflammation-related genes, and phosphorylation of Janus kinase 2 (JAK2), signal transducer and activator of transcription 3 (STAT3) and extracellular signal-regulated kinase (ERK) induced by IL-6. CIA was induced by immunizing with bovine type II collagen. <I>V. angularis</I> extract (VAE) was administrated orally at 50 and 100 mg/kg from day 1 to day 28. Induction of arthritis was evaluated with a visual scoring system and histological analysis.</P><P><B>Results.</B> Extracts or two triterpenoid compounds from <I>V. angularis</I> showed potent inhibitory effects on pSTAT3-inducible luciferase activity, STAT3 tyrosine phosphorylation and the expression of inflammation-related genes induced by IL-6. Administration of VAE significantly suppressed the progression of CIA, accompanied by a reduced antibody response to type II collagen and protection from tissue damage in knee joints.</P><P><B>Conclusion.</B> Administration of VAE has a therapeutic effect on CIA and this effect is associated with the inhibitory activity on IL-6/STAT3 signalling. These results suggest that extracts or compounds from <I>V. angularis</I> could be a useful treatment for diseases related to IL-6, including RA.</P>

      • A study on the molecular change of uterine myometrial cells in mice by endocrine disrupting chemicals

        Objective: This study examined the hypothesis that endocrine disrupting chemicals (EDC) are associated with premature birth (PTB). Di-2-ethylhexyl phthalate (DEHP) and bisphenol A (BPA) are best known as endocrine disruptors. Although they are known to be linked to the pathogenesis of PTB, the molecular and cellular mechanisms have yet to be fully understood. Methods: Five-week-old, virgin female ICR mice were administered a bolus of 100ul sunflower seed oil (100%), DEHP, and BPA by intraperitoneal injection every three days for three weeks. Formalin-fixed paraffin uterine sections were stained with H&E. Protein and RNA were extracted from uterus using protein extraction solution and Tri-RNA reagent, respectively. Data analysis was conducted by using western blot and quantitative real-time polymerase chain reaction (qRT-PCR). Results: In the group that treated with DEHP and BPA respectively, the expression of cyclooxygenase-2 (COX-2), oxytocin receptor (OTR), and connexin 43 (CX-43) related to the onset of labor pain and delivery were obviously increased. These group also showed a significant increase in the mRNA levels of the pro-inflammatory cytokines tumor necrosis factor (TNF)-, interleukin (IL)-1, and IL-6. Conclusion: These results support that DEHP and BPA activate inflammatory signals and induce the expression of the uterine activation proteins. Together, this findings may provide insight into the association between EDC exposure and obstetrical complications.

        Saturated fatty acids enhance osteoclast survival

        <P>Hyperlipidemia and marrow fat are associated with lowering bone density in vivo, suggesting that lipid contributes to bone loss. Using bone marrow-derived macrophages, we investigated the effect of saturated fatty acids (SFA) on osteoclastogenesis. The level of free fatty acids and adiposity in bone marrow was significantly elevated in obese mice. SFA increased osteoclast (OC) survival by preventing apoptosis. SFA caused the production of MIP-1alpha and led to activation of nuclear factor (NF)-kappaB in the OC. The absence of Toll-like receptor 4 (TLR4) or myeloid differentiation factor 88 (MyD88) abolished the survival effect of SFA on OC.</P>

        Case Report : Uterine Lipoleiomyoma in Peri or Postmenopausal Women

        Lipoleiomyoma is an uncommon neoplasm of the uterus, composed of smooth muscles intermixed with mature adipocytes. These tumors are considered a benign variant of uterine leiomyomas. Herein, we report six cases of lipoleiomyoma experienced in our institution from January 2005 to March 2015. The patients ranged in age from 45 to 70 years; the etiology may be related to estrogen deficiency occurring after menopausal transition. Except for one lipoleiomyoma in the broad ligament, all others were found in the uterine corpus. The presenting symptoms were nonspecific, and most cases were incidentally diagnosed during surgery for other reasons. We performed preoperative imaging studies, including abdominal and pelvic computed tomography and magnetic resonance imaging. Preoperatively, four patients were diagnosed as having a pelvic mass and one patient was diagnosed as having a right ovarian mature teratoma. In one case, we found a gynecologic malignancy (cervical cancer 1A1). Histologically, there was no gross or microscopic contiguity between the lipoleiomyoma and the malignancy. Lipoleiomyomas seem to have a benign clinical course. In our study, there were no recurrences of or deaths attributed to the lipoleiomyomas during a mean follow-up period of 16.17 ± 23.80 months.       • Increased expressions of oxytocin receptor and connexin 43 in the mouse uterus treated with endocrine disrupting chemicals : Implication of preterm labor

        Objective: Di-2-ethylhexyl phthalate (DEHP) and bisphenol A (BPA) are common endocrine-disrupting chemicals (EDCs) that are associated with preterm birth (PTB). However, the molecular and cellular mechanisms of these chemicals are still unclear. Accordingly, in this study, we evaluated whether EDCs were associated with PTB. Methods: Five-week-old virgin female ICR mice were administered a bolus of 100 L sunflower seed oil (100%), DEHP, or BPA by intraperitoneal injection every 3 days for 3 weeks. Formalin-fixed paraffin-embedded uterine sections were stained with hematoxylin and eosin and immunohistochemical staining. Human myometrial tissues were obtained from premenopausal women (n = 5) with uterine fibroids at the time of elective hysterectomy. Proteins and RNA were extracted from mouse uterine tissues and human myometrial cells using protein extraction solution and Tri-RNA reagent, respectively. Data analysis was conducted by using western blotting and quantitative real-time polymerase chain reaction. Results: Notably, in human myometrial cells treated with DEHP and BPA, the expression levels of cyclooxygenase-2, oxytocin receptor, and connexin 43, which are related to the onset of labor pain and delivery, were obviously increased. Also, they showed significant increases in mRNA levels of pro-inflammatory cytokines, including tumor necrosis factor-, interleukin-1, and interleukin-6. Meanwhile, progesterone receptor-A expression was specifically increased in samples obtained from mice treated with DEHP. Conclusion: Overall, these findings showed that maternal exposure to DEHP and BPA during pregnancy activated inflammatory signaling and induced the expression of uterine activation proteins through the decreased uterine responsiveness to the relaxatory effects of progesterone, providing insights into the association between EDCs exposure and obstetric complications.

        BRCA1 -mutated ovarian cancer with skin metastasis: a case report

        The major risk factor for ovarian cancer (OC) is mutation of the BRCA1 or BRCA2 DNA mismatch repair genes, which occurs in approximately 10% of OC cases. Most previous studies have demonstrated that BRCA1 - and BRCA2 -mutated OCs are associated with better prognosis than sporadic OCs. However, information about the patterns and clinical course of the metastatic spread of BRCA -mutated OCs is limited. Herein, we describe a case of OC with a BRCA1 mutation and skin metastases in a 49-year-old patient, which to the best of our knowledge has not been reported previously.

        Clinical application of serum anti-Müllerian hormone in women

        Anti-$M{\ddot{u}}llerian$ hormone (AMH), a peptide growth factor of the transforming growth $factor-{\beta}$ family, is a reliable marker of ovarian reserve. Regarding assisted reproductive technology, AMH has been efficiently used as a marker to predict ovarian response to stimulation. The clinical use of AMH has recently been extended and emphasized. The uses of AMH as a predictive marker of menopause onset, diagnostic tool for polycystic ovary syndrome, and assessment of ovarian function before and after gynecologic surgeries or gonadotoxic agents such as chemotherapy have been investigated. Serum AMH levels can also be affected by environmental and genetic factors; thus, the effects of factors that may alter AMH test results should be considered. This review summarizes the findings of recent studies focusing on the clinical application of AMH and factors that influence the AMH level and opinions on the use of the AMH level to assess the probability of conception before reproductive life planning as a "fertility test."

      • Comparison of four malignancy risk indices in the preoperative evaluation of patients with borderline ovarian tumor

        목적: The purpose of this study is to find an appropriate risk of malignancy index (RMI) cutoff-value by comparative analysis of the four malignancy risk indices (RMI 1, RMI 2, RMI 3, and RMI 4) in patients with borderline ovarian tumor (BOT) as a separate group. 방법: Seventy-six women with BOT and 434 individuals having benign adnexal mass were enrolled in this retrospective study. The sensitivity, specificity, positive predictive values, negative predictive values and diagnostic accuracy of the four malignancy risk indices, and RMI were calculated for prediction of discrimination between BOTs and benign adnexal masses and the results were compared. 결과: The area under the curves for RMI 1, RMI 2, RMI 3, and RMI 4 were 0.742, 0.755, 0.765 and 0.787, respectively. The accuracy of RMI 4 method was better than RMI 1, RMI 2 and RMI 3. A direct comparison of the four indices showed that RMI 4 at a cutoff level of 190 was significantly better at predicting malignancy than RMI 1 (p=0.0048) at a cutoff level of 80, RMI 2 (p=0.0000) at a cutoff level of 110, and RMI 3 (p=0.0021) at a cutoff level of 80. The RMI 4 at a cutoff level of 190 had a sensitivity of 71.1%, a specificity of 70.7%, a PPV of 29.8%, a NPV of 93.3%, and a DA of 70.8%. 결론: This study is first in the published studies comparing the four malignancy risk indices in patients with BOTs as a separate group. The best RMI 4 cut-off was found to be 190 for preoperative differential diagnosis or patient selection for further referral in patients suspected of BOTs.
