The diagnosis of latent Mycobacterium tuberculosis infection (LTBI) is recommended in hematological malignancy patients and before hematopoietic stem cell transplantation (Guidelines for the prevention and management of infectious complications of solid organ transplantation, 2004). Compared to traditional methods such as tuberculin skin test (TST), T-SPOT.TB has been shown to be more specific. In the present study we enrolled 536 patients for whom T-SPOT.TB was performed, among which 295 patients also received the TST test. The agreement (79%) between T-SPOT.TB and TST was poor (x=0.274, P<0.001). The patients with positive T-SPOT.TB results numbered 62 (11.6%), in which only 20 (48.8%) of the 41 receiving the TST test had positive results. A majority of the patients with T-SPOT.TB positive results had some other evidence ofTB, such as TB history, clinical symptoms and an abnormal chest CT scan. Active TB was found in 9 patients, in which 2 had negative TST results. We followed up the patients and no one developed active TB. Our study suggested that the T-SPOT.TB may be more useful for screening LTBI and active TB in hematological malignancy patients and hematopoietic stem cell transplant recipients than the TST test.

An ILV5 (acetohydroxyacid reductoisomerase) gene expression vector (designated pILV5) was constructed by cloning the yeast ILV5 gene into a yeast expression vector (pLG669Z)containing yeast CYC1 promoter. S. cerevisiae cells harboring the pILV5 plasmid showed ~3.7-fold higher level of the diacetyl reductase activity compared with the control yeast cells. When they were applied to wine fermentation using Campbell Early and Muscat Baily A grapes, the diacetyl content level was reduced to ~35.0–39.0% of those obtained with the control yeasts.

Ribosome-associated membrane protein 4 (RAMP4) is a membrane protein that exposes its N-terminal hydrophilic portion on the cytoplasmic side and spans the membrane close to the C-terminal end. RAMP4 has previously been reported to belong to the set of proteins that remains associated with membrane-bound ribosomes, and controls the glycosylation of major histocompatbility complex class II-associated invariant chain. RAMP4 also may be relative to the stabilization of membrane proteins in response to stress, with other components of translocon, and molecular chaperons in ER. Application of 5`-RACE technique with specially designed primer, we cloned a 715 bp cDNA fragment which contains a 195 bp ORF, termed RAMP4. The deduced protein has 64 amino acid residues and contains a putative transmembrane-spanning domain at the COOH terminus.

Trypsin inhibitors occur in a wide range of foods, they have been studied extensively in plants such as soybean (Glycinemax) as the majority of species are considered as important food sources. Selected soybeans were subjected to various treatments including microwave heating, supercritical fluid CO2 de-fatting, ultrasound incubating by different treat time (5min-120min) to determine the effect on soybean trypsin inhibitor (STI) activities compared to the non-treatment. The results indicated that microwave reduced trypsin inhibitors ranged from 20.57±1.40 TIU/mg to 14.60±1.48 TIU/mg compared to the control (21.44±1.04 TIU/mg) respectively (p<0.05). The inactivation of STI activities by ultrasound ranged from 28.47±0.16 TIU/mg to 19.86±0.78 TIU/mg compared to the control (28.87±0.18 TIU/mg) respectively (p<0.05). While the observed STI activities by supercritical CO2 de-fatting was not significant compared to the control. This study signifies the effective potential of inactivation of trypsin inhibitor by several applicable treatments which might be benefit in the food manufacture and livestock feeding industry.

The present study examined, for the first time, the in vitro wound healing potential of chitosan green tea polyphenols (CGP) complex based on the activation of transglutaminase (TGM) genes in epidermal morphogenesis. Response surface methodology was applied to determine the optimal processing condition that gave maximum extraction of green tea polyphenols. The antioxidant activity, scavenging ability, and chelating ability were studied and expressed as average EC50 values of CGP and other treatments. In silico analysis and gene coexpression network was subjected to the TGM sequences analysis. The temporal expressions of TGMs were profiled by semi-quantitative reverse transcription (RT)-PCR technology within 10 days after wounding and 2 days postwounding. CGP showed the effectiveness of antioxidant properties, and the observations of histopathological photography showed advanced tissue granulation and epithelialization formation by CGP treatment. In silico and coexpression analysis confirmed the regulation via TGM gene family in dermatological tissues. RTPCR demonstrated increased levels of TGM1–3 expression induced by CGP treatment. The efficacy of CGP in wound healing based on these results may be ascribed to its antioxidant properties and activation of the expression of TGMs, and is, thus, essential for the facilitated repair of skin injury.

T Bayesian network methodology taking place of fault tree analysis used for reliability assessment has gotten lots of attention in recent years. On basis of the current Bayesian network method used for calculating structural importance degree, a new Bayesian network method is raised. This new method can avoid repeat modification of the parameters in the conditional probability tables, and hence the intermediate results computed during inference process can be shared to decrease calculating complexity. The new method is proved to be correct in a mathematic way and a corresponding algorithm named SID_Jointree for realizing this new method is designed, which guarantees the new method can be realized in computer. Finally, the correctness and efficiency of the new method is validated by using two fault tree cases.
