      • The ERCC1 C118T Polymorphism Predicts Clinical Outcomes of Colorectal Cancer Patients Receiving Oxaliplatin-Based Chemotherapy: a Meta-analysis Based on 22 Studies

        Background: Although the predictive value of the excision repair cross-complementing group 1 (ERCC1) C118T polymorphism in clinical outcomes of patients with colorectal cancer (CRC) receiving oxaliplatin-based chemotherapy has been evaluated in numerous published studies, the conclusions are conflicting. Therefore, we performed the present meta-analysis to determine the precise role of the ERCC1 C118T polymorphism in this clinical situation and help optimize individual chemotherapy. Materials and Methods: A multiple search strategy was used to identify eligible studies. Pooled odds ratios (ORs) and their 95% confidence intervals (CIs) were used to estimate objective response and oxaliplatin-induced toxicity, with hazard ratios (HRs) with 95%CIs for progression-free survival (PFS) and overall survival (OS). Results: A total of 22 studies including 2,846 CRC patients were eligible in the analysis. Overall, no significant correlation was found between the ERCC1 C118T polymorphism and objective response to oxaliplatin-based chemotherapy, in all patients or in the Asian and Caucasian subgroups. However, the pooled analysis showed that the PFS and OS were significantly shorter in patients who carried T/T or T/C genotypes of ERCC1 C118T as compared to the C/C genotype. On stratified analysis by ethnicity, the ERCC1 118T allele was associated with a favorable prognosis in Caucasians (PFS, HR=0.58, 95%CI: 0.24-1.44; OS, HR=0.38, 95%CI: 0.22-0.64) but an unfavorable prognosis in Asians (PFS, HR=2.49, 95%CI: 1.87-3.33; OS, HR=2.63, 95%CI: 1.87-3.69) based on a dominant model. In addition, we failed to find a statistically significant impact of ERCC1 C118T polymorphism on oxaliplatin-induced toxicity. Conclusions: The ERCC1 C118T polymorphism may have prognostic value in patients with CRC undergoing oxaliplatin-based chemotherapy.

        Construction and Characterization of a Full-length cDNA Library and Identification of Genes Involved in Salinity Stress in Wild Eggplant (Solanum torvum Swartz)

        The objectives of this paper were to construct a full-length cDNA library and to isolate genes that confer salt tolerance from the leaves of salinity-tolerant wild eggplant variety, ‘Torvum Vigor’ (Solanum torvum Swartz). A full-length cDNA library from the leaves was successfully constructed by a switching mechanism at 5’-end of RNA transcript (SMART) approach and a long-distance PCR (LD-PCR) technique. The titer of the primary cDNA library was 3.6 ×106 cfu・mL-1 and that of the amplified library was 1.2 × 1010 cfu・mL-1. Gel electrophoresis results showed that most of the cDNA inserts ranged from 0.40 to 2.5 kb, with a recombination rate of 99%. A total of 427 randomly selected positive clones were sequenced. After removing the unsuccessful reads, 364 datasets were obtained and have been submitted to the NCBI Nucleotide Sequence Database under GenBank accession numbers JK265131-JK265494. Among the 364 submitted sequences, 74.45% of them contained full-length coding regions. BLASTX analysis revealed that 62.36% of the ‘Torvum Vigor’ expressed sequence tags (ESTs) possessed homology to known or putative proteins of other organisms. Seven genes that might be responsible for the encoding of known proteins in other organisms were identified to confer salt tolerance. This evidence demonstrated that the cDNA library constructed was a full-length library of high quality. It could be a useful resource for further research in the cloning of stress-related genes, which could be utilized in the genetic improvement of vegetable crops using transgenic technology.

        Multi-Objective Optimization of Multistage Pump Balance Drum System Based on BP Neural Network and Genetic Algorithm

        The axial force balancing capacity of a balance drum is a key factor affecting the life of multi-stage centrifugal pumps. In this paper, a double shell segmental multistage pump is taken as the research object. The hydraulic performance and axial force performance are set as the optimization objectives, and the performance data are obtained by numerical simulation with FLUENT software. The BP neural network is used to establish the prediction model of structural parameters of the balance system, hydraulic performance and residual axial force performance, and it is used as the adaptive value evaluation model of genetic algorithm to solve the optimal value in the sample space. The results show that the radial clearance of the balance drum and the balance tube orifice flowmeter, the axial width of the balance cavity are the significant factors affecting the hydraulic performance and axial force performance of the multistage pump. When the radial clearance of the balance drum is 0.1mm, the clearance of the orifice flowmeter is 1.95mm, and the axial width of the balance cavity is 55mm, the multi-stage pump has the best hydraulic performance and the smallest residual axial force. The vortex band in the balance cavity can increase the amount of the fluid spin and enhance the axial force balancing capacity of the balance drum. The greater the area occupied by the negative high-rotation fluid in the balance cavity, the stronger the ability of the balance drum to balance the axial force. The test results show that compared with the prototype multistage pump, at nominal flow rate, the head and efficiency of the optimized model are increased by 0.71% and 1.63% respectively, and the bearing temperature and vibration speed of the multi-stage pump are significantly reduced.

        Identification and genetic mapping for rht-DM, a dominant dwarfing gene in mutant semi-dwarf maize using QTL-seq approach

        Semi-dwarfism is an agronomically important trait in breeding for stable high yields and for resistance to damage by wind and rain (lodging resistance). Many QTLs and genes causing dwarf phenotype have been found in maize. However, because of the yield loss associated with these QTLs and genes, they have been difficult to use in breeding for dwarf stature in maize. Therefore, it is important to find the new dwarfing genes or materials without undesirable characters. The objectives of this study were: (1) to figure out the inheritance of semi-dwarfism in mutants; (2) mapping dwarfing gene or QTL. Maize inbred lines ‘18599’ and ‘DM173’, which is the dwarf mutant derived from the maize inbred line ‘173’ through 60Co-γ ray irradiation. F2 and BC1F1 population were used for genetic analysis. Whole genome resequencing-based technology (QTL-seq) were performed to map dwarfing gene and figured out the SNP markers in predicted region using dwarf bulk and tall bulk from F2 population. Based on the polymorphic SNP markers from QTL-seq, we were fine-mapping the dwarfing gene using F2 population. In F2 population, 398 were dwarf plants and 135 were tall plants. Results of χ2 tests indicated that the ratio of dwarf plants to tall plants was fitted to 3:1 ratio. Furthermore, the χ2 tests of BC1F1 population showed that the ratio was fitted to 1:1 ratio. Based on QTL-seq, the dwarfing gene was located at the region from 111.07 to 124.56 Mb of chromosome 9, and we named it rht-DM. Using traditional QTL mapping with SNP markers, the rht-DM was narrowed down to 400 kb region between SNP-21 and SNP-24. The two SNPs were located at 0.43 and 0.11 cM. Segregation analysis of F2 and BC1F1 indicated that the dwarfing gene was likely a dominant gene. This dwarfing gene was located in the region between 115.02 and 115.42 Mb on chromosome 9.

        Effect of Balance Drum Clearance on the Flow in the last stage Impeller Back Sidewall gap

        In order to study the effect of balance drum clearance on flow distribution in back sidewall gap of last-stage impeller, numerical calculation of a multistage pump was carried out. The accuracy of the numerical calculation method was verified with the external characteristic test. The axial distribution of the radial velocity at the inlet and the bottom of the back sidewall gap at different angles was plotted. The results showed that the radial velocity near the impeller flows radially to the outlet of the impeller, and the radial velocity near the pump shell flows radially to the hub. Influenced by the high-turbulent energy fluid at the outlet of the impeller, there are obvious eddy current movement at the inlet of the back sidewall gap, and the radial velocity increases with the increase of the balance drum clearance. When the flow rate increases from 0.5Q to 1.5Q, the radial velocity decreases gradually at the same angle. The results can provide a reference for the pump design and optimization.

        Enantioselective Liquid–Liquid Extraction of Underivatized Amino Acids with Simple Chiral Aminophenyl-Aldehyde

        The derivative of aminophenyl-aldehyde with an asymmetric carbon and an uryl group, (S)-2, was synthesized. The combination of (S)-2 and aliquat-336 in CDCl3 extracted underivatized amino acids in water layer by imine formation with enantioselectivities of 12/1 for Phe, 13/1 for Val, and 12/1 for Leu, which are comparable with those of previously reported binaphthol-based extractor (S)-1. The enantioselectivities of (S)-2 is remarkable considering the low molecular weight compared to (S)-1. Density functional theory computations and experimental data demonstrate that imine bond is strengthened by resonance-assisted hydrogen bond with the nearby ?NH? group.

        Algorithm of Gaussian Sum Filter Based on SGQF for Nonlinear Non-Gaussian Models

        To improve the filtering effect of the sparse grid quadrature filter (SGQF) under non-Gaussian conditions, the Gaussian sum technique is introduced, and the Gaussian sum sparse grid quadrature filter (GSSGQF) is developed. We present a systematic formulation of the SGQF and extend it to the discrete-time nonlinear system with the non-Gaussian noise. The proposed algorithm approximates the non-Gaussian probability densities by a finite number of weighted sums of Gaussian densities, and takes the SGQF as the Gaussian sub-filter to conduct the timeand measurement update for each Gaussian component. An application in the discrete-time nonlinear system with the non-Gaussian noise has been shown to demonstrate the accuracy of the GSSGQF. It outperforms the unscented Kalman filter (UKF), the cubature Kalman filter (CKF) and the SGQF. Theoretical analysis and simulation results prove that the GSSGQF provides significant performance improvement in the calculation accuracy for nonlinear non-Gaussian filtering problems.

        The Effect of Transformer Leakage Inductance on the Steady State Performance of Push-pull based Converter with Continuous Current

        As a result of the advantages such as high efficiency, continuous current and high stability margin, push-pull converter with continuous current (PPCWCC) is competitive for battery discharge regulator (BDR) which plays an important role in power conditioning unit (PCU).Leakage inductance yields current spike in low-ripple current of PPCWCCs. The operating modes are added due to leakage inductance. Therefore the steady state performance is affected, which is embodied in the spike of low-ripple current. PPCWCCs which are suitable for BDR can be separated into three types by current spike characteristics. Three representative topologies IIs1, IIcb2 and Is3 are analyzed in order to investigate the factors on the magnitude and duration of spike. Equivalent current sampling method (ECSM) which eliminates the sampling time delay and achieves excellent dynamic performance is adopted to prevent the spike disturbance on current sampling. However, ECSM reduces the sampling accuracy and telemetry accuracy due to neglecting the spike. In this paper, ECSM used in PPCWCCs is summarized. The current sampling error is analyzed in quality and quantity, which provides the foundation for offsetting and enhancing the telemetry accuracy. Finally, current sampling error rate of three topologies is compared by experiment results, which verify the theoretical analysis.

        Digital Transformation, Manipulation of Asset Evaluation and M&A Performance: Discussion on the Intermediary Effect of Internal Control

        Purpose - The purpose of this study was to examine the internal relevance between digital transformation, manipulation of asset evaluation and corporate M&A performance and further explores the impact path of manipulation of asset valuation on corporate M&A performance. Design/methodology/approach - This study based on the financial data of A-share listed companies in Shanghai and Shenzhen Stock Exchanges from 2013 to 2021. Findings - First, manipulation of asset evaluation is negatively correlated with M&A performance. Second, Digital transformation significantly weakens the negative correlation between manipulation of asset evaluation and corporate M&A performance. Third, The effectiveness of internal control plays a partially intermediary role in the process of manipulation of asset evaluation affecting M&A performance. Research implications or Originality - Enriching the existing literature on the subject, the study can also provide useful reference for improving the performance of corporate mergers and acquisitions, regulating asset valuation, promoting the digital transformation of enterprises and improving internal control mechanisms, with both theoretical and practical implications.
