Suspension system of vehicle have enough endurance during its life time to protect passenger. Spring is one of major part of vehicle. Thus, a fatigue strength evaluation for leaf spring based on road load response was carried out. At first, strain of leaf spring is measured on the city condition and proving ground condition. And next, the damage analysis of road load response data was carried out. And fatigue test of leaf spring were also carried out. Based on ε-N life relation, fatigue life of leaf spring was evaluated at belgian mode, city mode and drawing test specification. After that, it is compared the design life of leaf spring and evaluated fatigue life by belgian mode, city mode and drawing test specification. From the above, the maximum load-fatigue life relation of leaf spring was defined by test. and new test target of belgian mode and city mode was proposed to accept design specification of leaf spring. It is expect that proposed test target can verify leaf spring fatigue endurance at specific road condition.

Nonalcoholic fatty liver disease (NAFLD) occurs due to the accumulation of fat in the liver, leading to fatal liver diseases such as nonalcoholic steatohepatitis (NASH) and cirrhosis. Elucidation of the molecular mechanisms underlying NAFLD is critical for its prevention and therapy. Here, we observed that deubiquitinase USP15 expression was upregulated in the livers of mice fed a high-fat diet (HFD) and liver biopsies of patients with NAFLD or NASH. USP15 interacts with lipid-accumulating proteins such as FABPs and perilipins to reduce ubiquitination and increase their protein stability. Furthermore, the severity of NAFLD induced by an HFD and NASH induced by a fructose/palmitate/cholesterol/trans-fat (FPC) diet was significantly ameliorated in hepatocyte-specific USP15 knockout mice. Thus, our findings reveal an unrecognized function of USP15 in the lipid accumulation of livers, which exacerbates NAFLD to NASH by overriding nutrients and inducing inflammation. Therefore, targeting USP15 can be used in the prevention and treatment of NAFLD and NASH.

Suspension system of vehicle is very important part because it acts on ride comport and safety. And the spring is one of major part vehicle. Therefore it has to be designed to operate under severe conditions to ensure enough endurance. In general, the most typical and traditional method for fatigue design methods need many additional fatigue tests according to its geometry, materials, and fatigue loading condition. This indicates that a lot of cost and time for it have to be consumed. In this paper. the ΔP-N_f curve were obtained by fatigue tests. Using these results, ΔP-N_f curves were rearranged in to the Δδ_f relation with the maximum stress. By using the maximum stress and the fatigue data previously obtained as the fatigue test results, the fatigue design criterion could be predicted without any additional fatigue tests.

Recently, a new issue in designing spot welded structures such as automobile and train car bodies is to predict an economical fatigue design criterion. One of the most typical and traditional methods is to use a ΔP-N$\sub$f/ curve. However, since the fatigue data on the ΔP-N$\sub$f/ curve vary according to the welding conditions, materials, geometry of joint and fatigue loading conditions, it is necessary to perform the additional fatigue tests for determining a new fatigue design criterion of spot-welded lap joint having specific dimension and geometry. In this study, the stress distributions around spot welds of various spot welded lap joints such as in-plane bending type (IB type), tension shea. type (TS type) and cross tension type (CT type) were numerically analyzed. Using these results, the ΔP-N$\sub$f/ curves Previously obtained from the fatigue tests for each type were rearranged into the Δ$\sigma$-N$\sub$f/ relations with the maximum stresses at the nugget edge of the spot weld.

<P>The giant freshwater prawn, <I>Macrobrachium rosenbergii</I>, a sexually dimorphic decapod crustacean is currently the world’s most economically important cultured freshwater crustacean species. Despite its economic importance, there is currently a lack of genomic resources available for this species, and this has limited exploration of the molecular mechanisms that control the <I>M. rosenbergii</I> sex-differentiation system more widely in freshwater prawns. Here, we present the first hybrid transcriptome from <I>M. rosenbergii</I> applying RNA-Seq technologies directed at identifying genes that have potential functional roles in reproductive-related traits. A total of 13,733,210 combined raw reads (1720 Mbp) were obtained from Ion-Torrent PGM and 454 FLX. Bioinformatic analyses based on three state-of-the-art assemblers, the CLC Genomic Workbench, Trans-ABySS, and Trinity, that use single and multiple <I>k</I>-mer methods respectively, were used to analyse the data. The influence of multiple <I>k</I>-mers on assembly performance was assessed to gain insight into transcriptome assembly from short reads. After optimisation, <I>de novo</I> assembly resulted in 44,407 contigs with a mean length of 437 bp, and the assembled transcripts were further functionally annotated to detect single nucleotide polymorphisms and simple sequence repeat motifs. Gene expression analysis was also used to compare expression patterns from ovary and testis tissue libraries to identify genes with potential roles in reproduction and sex differentiation. The large transcript set assembled here represents the most comprehensive set of transcriptomic resources ever developed for reproduction traits in <I>M. rosenbergii</I>, and the large number of genetic markers predicted should constitute an invaluable resource for future genetic research studies on <I>M</I>. <I>rosenbergii</I> and can be applied more widely on other freshwater prawn species in the genus <I>Macrobrachium</I>.</P>

The vehicle suspension system is directly influenced to ride and handling. Therefore, the major components of the vehicle suspension system should have enough fatigue strength during its lifetime to protect passenger from the traffic accident. Spring is one of major suspension part of vehicle. Thus, in this paper, a fatigue design method for leaf spring based on geometric parameter was proposed. At first, it makes the fatigue test of leaf spring. On the base of these data, evaluation of numerical stress analysis for leaf spring assembly were performed. And next, after studying numerically on geometrical parameters of leaf spring assembly, investigated on the development of a fatigue life prediction method using the theory of neural network. An economical prediction method of fatigue design criterion for leaf spring assembly using the theory of artificial neural network was developed and certified its ability. Without performing a lot of additional fatigue test for a long time, fatigue design criterion for a new leaf spring assembly having different geometry can be predicted on the base of the already obtained fatigue data.

Although nanometric dead Lactobacillus plantarum has emerged as a potentially important modulator of immune responses, its underlying mechanism of action has not been fully understood. This study aimed to identify the detailed biochemical mechanism of immune modulation by micronized and heat-treated L. plantarum LM1004 (MHT-LM1004, <1 μm in size). MHT-LM1004 was prepared from L. plantarum LM1004 via culture in a specifically designed membrane bioreactor and heat treatment. MHT-LM1004 was shown to effectively induce the secretion of TNF-α and IL-6 and the mRNA expression of inducible nitric oxide synthase (iNOS). MHT-LM1004 enhanced the expression of TLR-2, phosphorylation of MAPKs (ERK), and nuclear translocation of NF-κB in a dose-dependent manner. Oral administration of MHT-LM1004 (4 × 10<sup>9</sup> or 4× 10<sup>11</sup> cells/kg mouse body weight) increased the splenocyte proliferation and serum cytokine levels. These results suggested that MHT-LM1004 effectively enhances early innate immunity by activating macrophages via the TLR-2/MAPK/NF-κB signalling pathway and that this pathway is one of the major routes in immune modulation by the Lactobacillus species.

<P><B>Background</B></P><P>Stanniocalcin-1 (STC1) is involved in fundamental biological processes such as angiogenesis, inflammation and wound healing, but little is known about its expression in human coronary atherosclerotic plaques or its relationship to plaque instability.</P><P><B>Objective</B></P><P>STC1 expression was examined in the culprit coronary plaques of 70 patients with acute myocardial infarction (AMI; n=49) or stable angina (n=21) who underwent directional coronary atherectomy.</P><P><B>Methods</B></P><P>The specimens were stained with H&E, STC1-specific antibodies, and endothelial cells, macrophages and smooth muscle cell markers.</P><P><B>Results</B></P><P>The baseline characteristics of the two groups of patients were largely similar. CD31-immunopositive and CD68-immunopositive areas, indicative of the presence of endothelial cells and macrophages, respectively, were proportionately larger while areas immunopositive for α-actin, as a smooth muscle cell marker, were proportionately smaller in the AMI group than in the stable angina group. The proportion of STC1-immunopositive areas was significantly greater in the AMI group than in the stable angina group (20.0% (8.2–29.0%) vs 8.8% (3.9–19.4%), p=0.022). Areas positive for STC1 were independently correlated with those immunopositive for CD31 (r=0.42, p<0.001) and CD68 (r=0.40, p<0.001). STC1 immunoreactivity co-localised with CD31-immunopositive and CD68-immunopositive cells.</P><P><B>Conclusions</B></P><P>STC1 is differentially expressed in the culprit coronary plaques of patients with AMI versus those with stable angina. STC1 may play a role in plaque instability.</P>

The recent tendency in the automobile industries is toward light weighting vehicle body to improve the problems by environmental pollution as well as improving fuel cost. The effective way to reduce the weight of vehicle body seems to be application of new materials for body structure, and such trend is remarkable. Among the various materials for vehicle body, stainless steel sheets and cold rolled steel sheets are under the interests. However, in order to guarantee reliability of new material and to establish the long life fatigue design criteria of body structure, it is necessary to assess spot weldability and fatigue strength of spot welded lap joints fabricated under optimized spot welding condition. In this paper, spot weldability of the stainless steel sheets, STS301L and STS304L, and the cold rolled steel sheets, SPCC and SPCD. Fatigue strength of lap joints spot -welded between similar and dissimilar materials were also assessed.
