The liver expresses a considerable amount of nitric oxide (NO) upon induction with cytokines or/and endotoxin. The NO synthesized by inducible NO synthase (NOS) of the liver seems to play a role in various hepatic physiological processes. Here we investigate the effects of NO on acetaminophen (AA)-induced liver injury. The treatment of S-nitros-N-acetyl penicillamine (SNAP, exogenous NO donor) at the dose of 0.1mM decreased AA-induced hepatotoxicity suggesting the possibility of NO to play a role in protection from the hepatotoxicity induced by AA. On the other hand, the excessive NO produced by NO donor (SNAP: 0.5, 2.5, 6.25mM) has been shown to cause a concentration dependent hepatotoxicity, and such damages was decreased by Superoxide and increased by superoxide dismutase, indicating that the hepatotoxicity induced by excessive NO depends on balancing between NO and superoxide. Taken together, the results indicate that NO has biphasic effects on hepatotoxicity.

Single subcutaneous administration to S.D rats of both sexes was performed to investigate the acute toxicity of Syndella gel, a new topical drug containing deproteinised dialysate of calfs blood and micronomicin sulfate. LD_(50) values for S.D rats were 23,047 ㎎/㎏ for male and 23,725 ㎎/㎏ for female. The death occurred within 24 hours after administration at doses over 19,200 ㎎/㎏. The main cause of death seemed to be respiratory disturbance by acute shock. Major general symptoms induced by injection subcutaneously with Syndella gel were underactivity, decreased respiratory rate, salivation, tremor and loss of consciousness. No significant body weight changes and gross findings of internal organs in treatment groups in comparison with those of control groups was observed at any dose levels in Syndella gel.

This study was conducted to investigate the skin irritation toxicity of Syndella gel, a combination topical drug containing a deproteinised dialysate of calfs blood and micronomicin sulfate in the ratio of 20 to 1, in New Zealand White rabbits. In the primary skin irritation test with male New Zealand White rabbits, there was no treatment-related effect on clinical sign, and body weight was not significantly changed. The Primary Irritation Index (PII) was 0.33, indicating that Syndella gel was a mildly irritating formulation.

Antigenicity of DA-3285, human recombinant erythropoietin which was produced from mammalian cells, was examined in guinea pigs and mice. In active systemic anaphylactic test, mild anaphylactic signs were observed in guinea pigs sensitized subcutaneously with DA-3285 or DA-3285 incorporated with complete Freund`s adjuvant(CFA) when challenged with high dose(1000 IU/Kg) of DA-3285. Other groups showed negative responses. In mouse-rat passive cutaneous anaphylaxis test, 20% sera of mice immunized with DA-3285 or DA-3285 mixed with aluminum hydroxide(alum) showed mild positive responses. In the case of indirect haemagglutination reaction(IHA) test, when sheep red blood cells coated with DA-3285 was incubated with mouse serum, all the serum samples were showed negative responses. These results suggest that DA-3285 has a very weak antigenic potential and probably would not induce systemic allergic reactions in clinical uses.

This study was conducted to investigate the subacute transdermal toxicity of Syndella gel, a new topical drug containing deproteinized dialysate of calf''s blood and micronomicin sulfate in Sprague-Dawley rats. Three doses (1.97, 3.94, 7.88 g/kg) of Syndella gel was daily treated transdermally to male and female rats for 30 days. No death was occurred in either control or treated rats. No significant toxic clinical signs and body weight change were not observed at any doses in the male or female rats treated. There were no significant alterations in hematologic and biochemical parameters in both sexes, however slight increase of potassium concentration was observed in 3.94g/kg and 7.88 g/kg female groups. No significant necrotic changes were not observed in examined organs. This study showed that up to 7.88g/kg Syndella gel did not induce subacute transdermal toxicity.

ABSTRACT: Activation of cell growth leads to the multiple phosphorylation of 40S ribosomal protein S6. The kinase responsible for controlling this event is termed p70^s6k/p85^s6k. Both isoforms of the kinase are derived from a common gene activated by a complex set of phosphorylation events; each residues in a unique cellular compartment: the p70^s6k in the cytoplasm and the p85^s6k in the nucleus. Although p70^s6k/p85^s6k represent the first mitogen-activated serine/threonine kinase described, the signaling pathway leading to activation of both isoforms remains obscure. Recent studies have shown that this pathway is distinct from that of p21^ras and the p42^mapk/p44^mapk, and that bifurcation of these pathways takes place at the level of the receptor. The p70^s6k signaling pathway can also be ablated by the immunosuppressant rapamycin, which blocks p70^s6k activation and S6 phosphorylation without affecting the other kinases whose activation is triggered by mitogen treatment. In parallel, rapamycin suppresses the translation of a family of mRNAs that contain a polypyrimidine tract at their 5' transcriptional start site. The implication is that this event is mediated by the phosphorylation form of S6 that may either directly interact with the polypyrimidine tract or alter the affinity of the 40S ribosome mRNA binding site for polypyrimidine tract mRNAs, or recognize proteins that directly bind to the polypyrimidine tract.
數種動物의 膵組織內 Protein Methylases의 活性度 分布에 관한 硏究

Post- translational modification of amino acid residue side-chains in protein has come to be a well known biochemical phenomenon. These reactions include methylation, carboxylation, acetylation, phosphorylation, and hydroxylation. One such side-chain modification of protein, protein methylation is being investigated actively with regard to bacterial chemotaxis, cytochrome c, gene regulation, and carnitine biosynthesis. The methylated amino acids occur in nature in highly specialized proteins. These methylated amino acids are formed by reactions catalyzed by protein methylase Ⅰ, Ⅱ and Ⅲ. In recent studies it was reported that protein carboxymethylation is involved in amylase secretion of parotid gland by isoproterenol. It was also suggested that a small part of the total cellular protein carboxymethylation is directly involved in pancreatic enzyme secretion. On the contrary, other authors reported that there is no relationship between protein carboxymethylation and secretion in pancreas and parotid. In this study we first examined the activities of protein methylase I, II, and III in pancreatic tissues of various animals, such as rat, mouse, guinea pig, rabbit, pig, cow, cat, chicken, and frog. The following results were obtained; 1. The activities of protein methylases were generally high in pancreatic tissues of cat, pig, and rabbit. Specifically, the activity of protein methylase I was very high in pancreatic tissue of cat, but low in the tissue of rat. Bovine pancreas had been shown to be the richest source for protein methylase Ⅱ, while the activity of protein methylase Ⅲ was relatively high in pancreatic tissue of cat. 2. The activities of protein methylase Ⅰ, Ⅱ, and Ⅲ in pancreatic tissue of chicken were generally high. 3. Interestingly, the activities of protein methylase Ⅰ, Ⅱ, and Ⅲ were not detected in pancreatic tissue of frog which is amphibian.
