7Mis study was conducted to analyze genetic characteristics and to develop the breed-specific DNA marker for breed identification of Korean native goat(KNG) as an indigenous animal genetic resource using random amplified polymorphic DNA(RAPD) and DNA amplification fingerprinting(DAF) techniques. A total of 30 KNG wide the high phenotypic purity and five exotic breeds including imported black goat(1BG) as a meat type breed and Saancn(SAA). Alpine(ALP), Nubian(NUB) and Toggenburg(TOG) as dairy type breeds were used in this study. Ten individuals were sampled from each exotic breed. Genomic DNA derived from blood or milk samples was amplified by PCR using random primers. Thirty different l0-base oligonucleotide primers were used in the initial screening. Of the 30 arbitrary selected primers, twelve primers showing highly polymorphic and reproducible amplification patterns were chosen for further analysis of RAPD and DAF. A total of 75 RAPD markers were produced, 36(48.0%) bands of which were polymorphic, compared to a total of 184 DAF markers with 117(64.0%) polymorphic bands. Therefore, the amount of polymorphism detected by DAF is much greater than that of RAPD. The band sharing value (62%) in the KNG was the highest among goat breeds, indicating that genetic similarity within KNG population was higher than those of other goat breeds. Two of the primers. GEN-01(GGTGACGCAG) and GEN-08(ACCCCCGAAG), produced breed-specific DNA bands showing differences between breeds. The 1.80kb band of GEN-O1 primer and 1.0kb band of GEN-08 primer were present in all of the dairy goat breeds examined including SAA, but not present in the meat type breeds of KNG and IBG. On the contrary, the 0.70kb band of GEN-Ol and 0.60kb band of GFN-08 were found in all of the KNG, but not in the IBG nor any of the dairy goat breeds. Therefore, these DNA bands specific for KNG may be useful putative breed-diagnostic markers to discriminate KNG from other improved goat breeds. In comparison of genetic distance and dendrogram, the KNG was the most closely related to the IBG, followed by the SAA. In conclusion. the RAPD and DAF markers will be useful for estimating genetic variation and relationships in goat breeds and for the breed identification of KNG.
