        자외선을 쪼인 DNA의 알카리에 의한 상해 반응 메카니즘을 연구하기 위해 DNA의 모델 화합물로서 thymidine dinucleotide monophosphate, d(TpTp)를 thymidine으로부터 출발하여 'phosphotriester' 방법에 따라 합성하고 이것에 자외선을 쪼여 광부가물을 분리하였다. 분리된 d(TpTp) 광부가물의 분광학적 성질은 d(TpT) 광부가물과 완전히 일치하였다. d(TpTp) 광부가물을 산 또는 알카리로 처리하여 스펙트럼을 비교한 결과 생성물이 서로 달랐으며 thymine 광부가물의 경우에도 같은 결과를 얻었다. 이 광부가물을 알카리로 처리하여 전기영동에 대한 상대적인 이동도를 비교하면 처리한 후의 이동도가 처리하기 전에 비해 현저히 감소하였다. 이러한 결과로부터 d(TpTp) 광부가물을 알카리로 처리하면 인산결합 및 당-피리미돈의 C-N결합이 끊어진 화합물이 생성될 뿐만 아니라 광부가물의 피리마딘 잔기에도 반응이 일어나 산으로 처리하였을 때와는 달리 탈수반응이 아난 다른 종류의 반응이 일어남을 알 수 있었다. Thymidine dinucleotide monophosphate was synthesized by 'phosphotriester' method starting from thymidine and (6-4) photoadduct of d(TpTp) was isolated after irradiation of UV light in order to understand the nature of alkali labile lesion of UV irradiated DNA. Spectroscopic properties of the isolated d(TpTp) (6-4) photo adduct were quite similar to that of d(TpT) photoadduct. Spectroscopic properties of the alkali hydrolyzed product of d(TpTp) photoadduct were different from that of the acid hydrolyzed photoadduct. Similar result was obtained when the reaction was run by use of thymine (6-4) photoadduct. The relative mobility of d(TpTp) photoadduct on electrophoresis was considerably decreased after treatment with hot alkali. These results show that not only the phosphate and C-N bond between sugar-pyrimidone moiety were cleaved but also transformation of pyrimidine moiety of the photoadduct took place by treatment with hot alkali. This transformation of the photo adduct was different from that of acid hydrolysis which gives dehydration product.
