본 연구는 여러가지 유기용매로 추출한 마늘의 항암성 성분을 in vitro에서 흰 생쥐의 백혈병성 임파모 세포인 L1210과 P388, 인체 직장암 세포인 HRT-18과 인체 결장암 세포인 HCT-48과 HT-29 그리고 흰쥐 복수육종암 세포인 S-180세포를 대상으로 항암효과 및 in vivo에서의 면역증강효과에 관하여 수행하였다. 실험 결과를 종합해 보면 다음과 같다. 첫째, 실험에 사용된 4가지 추출물중에서 석유에텔 추출물의 활성이 가장 높았고, 알콜 추출물의 활성도 석유에텔 추출물의 활성과 비슷하였으며 회수율은 가장 높았다. 둘째, 암세포의 종류에 따라서 증식억제율에 차이가 났다. 세째, 배양중 마늘 추출물의 농도가 높을수록 세포의 증식이 억제될 뿐만 아니라 암세포가 사멸되는 현상을 보였다. 넷째, 마늘 알콜 추출물을 포함한 배양액에서 증식된 L1210 및 HCT-48세포의 크기 분포도는 배양시간에 따라 세포크기가 작아짐을 보였고 현미경으로 관찰한 결과 본래의 형태에서 위축되거나 세포주위가 흐트러지면서 사멸하였다. 다섯째, 마늘 알콜 추출물이 S-180으로 접종되어 생긴 mass의 성장을 억제시켰다. 여섯째, S-180에 접종된 생쥐의 흉선, 간 및 비장의 무게는 접종되지 않은 생쥐의 그것보다 무거웠으며 특히 S-180에 접종되고 마늘 추출물로 처리된 생쥐의 흉선은 S-180만 접종된 생쥐의 흉선보다 무거웠다. 일곱째, 생쥐(C57BL/6J)의 복강으로 마늘 추출물을 투여했을때 대조군에 비해 NK세포의 활성이 증가하였으며 마늘 추출물과 S-180세포를 함께 투여했을 때 보다 큰 NK세포활성의 증가를 가져왔다. 여덟째, 복강으로 마늘 알콜 추출물이 투여된 생쥐 비장세포의 용혈반형성 세포수는 아무것도 처리하지 않은 생쥐의 그것보다 많았다. 그러나 개체차이를 감안할 때 좀 더 추구해야 할 것으로 사료된다. This study was devised to observe the immunopotentiating activity and the cytotoxic activities of garlic extract against various cancer cells, that is, murine leukemic lymphocyte(L1210 and P388) and human rectal(HRT-18), colon cancer cells(HCT-48 and Ht-29) and murine ascitic tumor cell(S-180) in vitro. Each extract was prepared by extraction with water, ethyl alcohol, chloroform and petroleum ether, and then cytotoxic activity of each extract was compared with control group and each other. The experimental results obtained are summarized as follows; The inhibitory effects on growth rate of cancer cells were strongest in the petroleum ether extract and next in the ethyl alcohol extract. The growth rates of the cancer cells in medium containing garlic alcohol extract were inhibited gradually to a significant degree in proportion, to the increase of the extract concentration. The patterns of size distribution of L1210 and HCT-48, maintained, for 48 hours, in medium containing garlic alcohol extract were shifted to direction of the small size region. The microscopic growth patterns of human cancer cell lines in medium containing garlic alcohol extract were shrinked and distracted. The weights of immune organs of mice innoculated with S-180 and treated with garlic are increased more than only innoculated with S-180. The growth of S-180 mass of mice is inhibited by garlic alcohol extract. The activity of NK cells from C57Bl/6J mice pretreated with garlic extract, especially innoculated with S-180 is increased.
