      • DNA 추출에 의한 Feulgen 반응효과에 관한 연구

        By the Reports, the residual DNA amount after acid hydrolysis of Feulgen's method. a absorption assay for DAN was effectively applied. At an optimal time length of acid hydrolysis for Feulgen reaction, the ratio of the residual DNA of non-hydrolysis to total DAN is 10% or more in hepatocyte or lymphocyte nuclei. In this paper, a Ultraviolet diode array spectrophotometer(HEWLETT 8452A^TM) has been used to investigate the yield of DNA from each purification methods;phenol-chloroform-isoamyl alcohol method, proteinase K method, alkaline lysis method, boiling method according to polymerase chain rcaction procedure, and the Feulgen reaction is able to detect absorption comparison to normal cell vs abnormal cell DNA. this samples be readily extracted from bovine liver tissue and human stomach tissue. as a purification for Feulgen reactions, total genomic DNA is used. Among the new DNA technological methods for Feulgen reaction, The sensitivity and specificity of the phenol-chloroform-isoamyl alcohol method is approximately 60% depending on the availability of other methods for comparison. and nucleic acid amount after purification, but a absorption assay for only DNA was effectively increased when approximatcly 60 minutes lcngth of acid hydrolysis and Feulgen reaction, and the absorption of abnormal tissue absorption comparison with normal cell. It is proposed as a first approach for the definition of a purification standard for DNA mcasurcments. Although not entirely free of technical limitations. DNA purification holds advantages over other diagnostic tests. including exquisite sensitivity, specificity, interchangeability and spced. It is expected that, in the immcdiate future DNA detection will be a prominent part of the methods used in clinical pathology.

      • 생체 전기의 임상적 이용실태에 대한 고찰

        To know the frequency of the clinical physiologic test (:bioedectrometrics) which is regarded as an indication of the importance of bioedectric sign and of the increase of medical benefit to man, and to prove the necessity of the eledtric sign in the step of the first medical treatment, I examined the diagnostic results of the physical electric sign made in the bioelectrometric laboratory of Chun-nam university hospital. A slight electric current is produced in the body surface because of the excitement resulted from the stimulation when cardiac miscle, cerebral cell and skeletal muscle is impulsed. This electric current is called physical electric sign. We can apply the informations obtained from this physical electric sign to various clinicobiochemical tests. Bioelectrometric which does not give any pain to person gives us a lot of indirect medical informations is widely used in study, therapy and diagnosis owing to the development of science such as edectronics, mechanics, physiology etc. We can see that the frequency of this test increases by 15% every year, and the abnormal condition can be looked for easily in comparson with other usual tests and that edectrocardiogram (: ECG) is useful in adult and electroencephalogram (;EEG) in infant. We should pay attention to the abnormal percentage ECG of 68.85% in the forties and EEG positive rate 65.38% in the teens. Both ECG and EEG tend to be recorded as comparatively high positive rate in the forties regardless of sex Though bioelectrometrics is indirect medical information, it should be widely used to discover diseases without delay and prevent adolescent diseases.

        It is important that we should do our best to keep the function of the blood cell originally in extracorporeal circulation for open heart surgery. In cryo-surgery, to control the temperature of circulatory blood is hypothermia. It is generally known that the pathological and physiological changes in the blood cell are one of the important causes of postperfusion syndrome. So in this report, the statistical differences between the various physical changes in circulatory blood cell(n=20) and pre0P values are compared through the clinical tests. As a result, on account of the long-term perfusion the increase of red blood cell distribution width(RDW)(p<0.05) and the decrease of plateletcrit(PCT)(p<0.01) are shown, but white blood cell(WBC) parameters are not changes. It is suggested that blood cell damage can be prevented by means of the effective perfusion and pheresis instead of whole blood transfusion is more proper.

        To investigate the fetal bovine serum(FBS) effect for growth and living cells on mouse foreskin cell that this report experimented to primary cluture with serum contain(group Ⅰ) versus serum free(group Ⅱ) culture conditions in the Hank's balanced salt solution(HBSS). Those foreskin cells that were maintained in culture media under air conditions equilibrated with 5% CO, 95% air and humidifyed to the water tray. The inverted microscopic distribution of culture cells at 48 hours after postincubation is that of Group Ⅰ is showed to clear in backgroung at the focus and increased to attachment of unknown cluster cells at culture well bottom. But, growth and living cells appears to increase at group Ⅱ. Fibroblasts are appdared that some of the spindle-shaped cells derived from dermal lesions to group Ⅰ and Ⅱ at 28 hours after postincubation by the silver stain. The FBS was known to growth inhibitors of fibroblast during cell culture, but these serum contained culture system was not inhibited to growth of fibroblasts to compare with serum free media. The results of growth cells have been differented under two culture conditions; the number of epithelial cells of group Ⅰwas showed to increase at only early stage(24 hours), but that of group Ⅱ was increased to compare with group Ⅰ after 48 hours. By these results, It is that of the serum free culture is effected to primary culture for establish of cell line and purification of special constitution of cell.

        개섬술 체외순환시 ACD 가 칼숨 농도 변화에 미치는 영향

        Because heart function arrest for a moment during open heart surgery, the safe prevation of myocardial condition in both metabolic and physiologic aspect by reducing energy consumption of ischemic heart to a minimum by means of extracorporeal circulation makes circulation function restore to normal. Among the various factors which have an infl uence on the contraction of ischemic heart, serum ionized calcium plays an important role. Bank citrated blood is supplemented because a lot of bleeding takes place during open heart surgery. Calium chelating agent, ACD solution included in the bank citrated blood plays a role in decreasing calcium density in cell. In observing 20 congenital heart disease patients are changed serum total calcium and ionized cacium concentration in response to the used quantity of bank citrated blood during cardio pulmonary by pass. Calcium concentration is decreased more in B group in which whole blood is more used than in A group in which whole blood is less used (P < 0. 01) . On the contrary, calcium concentration is much more increased in two TOF patients who receive the prime volume in which whole blood is little used than other patients(P<0.03). This change is remarkably appeared in the patient who has the small total blood volume, as the operation time is prolonged, calcium concentration is steadily increased.

        This report experimented that to investigate the σypsin treatment for inhibition of fibroblastic contamination and the fetal bovine serum(FBS) effect for trypsin inactivity toprimary culture of mouse foreskin. 0.25 % cold trypsin incubation could removed to dermallayer and subcutaneous materials, and 2.5% FBS σeatment could stopped to trypsin activity. πlose foreskin cells that were maintained in Dulbecco’s Modified Eagle Medium(DMEM) with nonessential amino acid(NEAA) under air conditions equilibrated with 5 % C02, 95 % air and humidified to the water tray. πle inverted microscopic distribution of culture cells at 24 hours after postincubation is showed to c1ear in background at the focus and increased to attachment of c1uster cells at culture well bottom. and then, growth cells and living cell ratio are increased to cold trypsin and FBS treatmented group at 48 hours. Fibroblasts are appeared that some of the spindle-shaped cells derived from dennal lesions to control group, but experimental group is showed to epitheloid cells compare with control group by the silver stain. The trypsin pretreatmented samples showed to epitheloid colonies after postinoculation without other cellul따 contamination to primary culture. πns culture system might provide to a procedure of epidennal cell pure culture.

      • 화상분석기를 이용한 정도별 이형성증에 대한 연구

        The purpose of this study was to develop discriminant analysis models for predicting cervical normal/dysplasia case diagnoses using cytometric features derived from the digital image analysis of cell monolayers. The database consisted of 19 cases diagnosed either as normal (n=5), moderate dysplasia (n=7), severe dysplasia (n=7) on monolayer preparations. We studied the nuclear and cytoplasmic characteristics of cells in the normal, moderate dysplasia and severe dysplasia on cervical samples. The morphometric parameters selected for the analysis were nuclear/cytoplasmic ratio and the nuclear variations measured by image analysis on normal and precancerous lesions of cervical smears; several shape factors; area; perimeter; maximal, minimal and equivalent circle diameters. The results showed that the dysplasia samples exhibited changes in both cellular and nuclear form and size but lacked substantial differences in the tumor grades. The coefficient of nuclear variation is as follows to normal cell 21.8±3.2%, moderate dysplasia 33.5±6.1%, severe dysplasia 27.7±5.8 of cervical smears.

      • 홍어 장점막 점액의 조직화학적 성상에 관한 연구

        This study is performed in order to clarify the histochemical structure, the distribution of mucous cell and goblet cell, and the histochemical properties of the mucosubstances in the middle region of intestinal mucosa and rectum of Raja kenojei. In the H&E stain, distribution of the mucous cells and acidophilic cells were a more compacted middle portion than other regions of intestine, but the former was more than the latter in the number of mucous cells to rectum. The mucosubstances were stained with aldehyde fuchsin pH 1.7-alcian blue(pH 2.5) stain and then compared to distribution of the mucosubstances used in image and microscope technology(IMT-Size5). The middle intestine of Raja kenojei was composed of mucous cells having only large amounts of mucosubstances in the distal region was much more than that of proximal region. It was two types of mucous cells to rectum, one type was the same as proximal intestine while the other had small amounts of weakly sulfated and large amounts of carboxylated mucins.

        Unicryl 표본에서 광학/전자 현미경 In Situ Hybridization -Rabbit hemorrhagic disease viral RNA 증명-

        Rabbit hemorrhagic disease<RHD) which was first recognized in China in 1984 spread via Eastern Europe to many countries of Western Europe and other parts of the world. This study is purposed. to detection of RHD viral RNA by In situ hybidization(ISH) as the immunohistochemical technique on base of the hemagglutinin inhibition titer and histopathologically changes after inoculation of rabbits with isolates of RHDV obtained from RHD infected cell. After experimental intramuscular infections with isolates of the virus, Infection were leaded acute RHD and showed convulsions, peresis and somtimes bloody or bloody-foamy nasal excretions. The body temperature of all cases show SOIre fever shortly before death. In order to detect antibodies to RlIDV, the hemagglutinin inhibition test. animal which survived an infection with RHDV developed antibodies to RlIDV, which were highly dectable at 12 hours after post infection these antibody titers reached the highest on 48 hours after infection Histopatholgically, in the liver early after infection were detected of hermrrhages. degenerative perilobular alterations in hepatocytes strongly homogenous or pale vacuolized cytoplasm, noclear pyknosis, karyorrhexis of karyolysis were observed Antibodies to RHDV were found in sera that indicate the existence of an pathogenic virus which is related to RHDV Simple stain and light microscopic ISH make possible using semi-thick sections to the post-unicryl embedded block for electron mcroscopic ISH therefore, it was established compared to viral focal location at same region. Hybridization for detection of viral RNA in unicryl embedded tissues using complementary 84 bases oligonucleotide probes labelled by biotin cyanoethyl-phosphoramidite compared with 4717-4ro) sequences of RHOV eDNA The best results were obtained to retention and accessibility of RHOV in O.Q20/o glutaraldehyde, depression of aldehyde group by ammonium hydroxide, unicryl resin block and block stain, biotinylated oligonucleotide probes cocktail, acetylation, hybridization during 2 hours at 3TC and signal marked by antibiotin antibody-10nm gold ISH microparticles signal on light microscopic in situ hybridization on semi-thick sections of normal cells and RHOV infected liver using biotinylated probe. It ~ as if indicate the presence of RHO viral nudeic acids by showed gold dense signal, appearing in the cytoplasm of degenerated hepatocytes of sudden death cases. Infected cells are easily recognized to condensed nucleus and central vacuoles, But, The light microscopic ISH were not showed to positive reaction because of limits of resolving power of light microscope and immuno-gold size. At the electron microscopic ISH, RHO viral RNA was distributed widely in the cytoplasm of liver cell. Gold particles were separated of inner and outer nudeanrembrane with destruction of the membrane, and dilatation of rough endoplasmic reticulum in the cell. Many viral RNA gold particles are present in the juxtanuclear cytoplasm and in the vicinity of rER of natural cases of sudden death and dusters of virus particles are seen in the cytoplasm.
