국내 미기록종 선충포식성 곰팡이 Drechslerella brochopaga Kan-23의 형태 및 계통분류

참외 경작지 토양으로부터 순수 분리된 선충포식성 곰팡이 Kan-23 균주는 배양 30일 이후에 분생포자가 형성되는 증식이 매우 더딘 특성을 나타내었다. 주사전자현미경(SEM)을 이용하여 Kan-23 균주의 미세형태 구조를 관찰한 결과 분생포자병의 각 가지마다 $5{\sim}10$개의 분생포자를 형성하였으며, 분생포자는 3개의 격막을 형성하고 타원형을 나타내었고, 수축성 고리형태(constricting ring)의 포식 구조체를 형성하는 특징을 나타내었다. 다양한 배지를 이용하여 포식 구조체의 형태학적 특성을 비교 검토한 결과 GPA 배지에서 2배 이상 크기의 대형 수축성 고리 형태의 포식 구조체를 형성하는 특징을 나타내었다. Kan-23 균주의 rDNA의 ITS 영역의 염기서열을 분석한 결과, Drechslerella 속의 계통군에 속하였으며, 특히 Drechslerella brochopaga (U51950)와 99%의 높은 염기서열 유사도를 나타내어 Drechslerella brochopaga로 동정되어 국내 미기록종임이 확인되었다. Strain Kan-23 was extracted from nematophagous fungi, which were isolated from the soil sample of oriental melon field. The strain exhibited the slow-growing characteristic forming conidia after prolonged incubation for 30 days. Morphological features of strain Kan-23 were observed under scanning electron microscope (SEM). It possesses erect conidiophores which contain $2{\sim}3$ side branches, with each branch producing $5{\sim}10$ conidia. The size of conidiophores were between $160{\sim}450\;{\mu}m$. Conidia were ellipsoidal with three septa[septum] in each conidium. Strain Kan-23 captured nematodes by means of giant constricting rings, which were observed in the glucose peptone agar medium. ITS region of rDNA sequence was analyzed. On the basis of the high sequence similarity of ITS region (99%), the Kan-23 strain was closely related to Drechslerella brochopaga (U51950). This is the first report on Drechslerella brochopaga as a nematophagous fungus in Korea.
난분해성 케라틴 단백질을 함유하는 닭 우모 분해세균의 분리 및 특성

양계장 부산물 시료로부터 우모 분해세균 31균주를 분리하였다. 수집된 우모 분해세균에 대해 16S rRNA 염기서열을 해석하여 계통학적 특성을 검토한 결과, Firmicutes (21균주), ${\gamma}$-proteobacteria (4균주), Actinobacteria (4균주) 그리고 Bacteroidetes (2균주) 계통군에 속하는 다양한 세균이 확보되었다. 우모 분해세균 중 우모 분해율이 75-90% 이상인 우수균주 Chryseobacterium sp. FBF-7, Stenotrophomonas maltophilia FBS-4 그리고 Lysinibacillus sp. FBW-3를 최종 선발하였다. 이들 균주를 이용한 생물학적 분해법과 Ca(OH)2를 이용한 화학적 분해법에 의해 추출된 아미노산의 특성을 비교한 결과, Chryseobacterium sp. FBF-7에 의해 추출된 총 아미노산 함량이 1661.6 ${\mu}mol$/ml로 선발 균주 중 가장 높게 나타났으며, 화학적 분해법에 의해 추출된 총 아미노산 함량과 유사하였다. Chryseobacterium sp. FBF-7에 의해 생성된 필수 아미노산 함유량은 619.3 ${\mu}mol$/ml로 총 아미노산의 37%를 함유하였으며, 화학적 분해법에 의한 경우, 596.9 ${\mu}mol$/ml의 필수 아미노산을 생산하여 총 아미노산 함유량의 32%를 차지하는 것으로 나타났다. Chryseobacterium sp. FBF-7에 의해 추출된 아미노산의 조성을 검토한 결과, valine, glutamic acid, aspartic acid, glycine 그리고 proline이 주요 아미노산이었으며, 특히 aspartic acid, threonine, serine, cysteine 그리고 tyrosine이 화학적 추출법보다 더 높게 추출되는 특징을 나타내었다. Thirty-one chicken feather-degrading bacteria were isolated from wasted feather, compost and wastewater in a chicken farm. These isolates were categorized as Firmicutes (21 strains), ${\gamma}$-proteobacteria (4 strains), Actinobacteria (4 strains), and Bacteroidetes (2 strains) by 16S rRNA gene sequence analysis. We examined the feather-degrading isolates for degradation in the 2% of chicken feather meal. The strain Chryseobacterium sp. FBF-7, Stenotrophomonas maltophilia FBS-4, and Lysinibacillus sp. FBW-3 were selected as a keratinolytic protein degrading bacteria which showed the highest feather degradation of 75-90%. The characteristics of amino acids extracted from chicken feather meal by using keratinolytic protein degrading isolates and chemical method with $Ca(OH)_2$ were analyzed. Total amino acid content of strain Chryseobacterium sp. FBF-7 was 1,661.6 ${\mu}mol$/ml, which was the highest and it was similar with chemical method. And essential amino acid content of total amino acid was thirty-seven percent (619.3 ${\mu}mol$/ml) and 596.9 ${\mu}mol$/ml for keratinolytic protein degrading isolates and chemical method, respectively. The major amino acids were valine, glutamic acid, aspartic acid, glycine, and proline by the strain Chryseobacterium sp. FBF-7 and especially, higher contents of aspartic acid, threonine, serine, cysteine, and tyrosine were detected compared with chemical method.
피틴산(Phytic acid)이 비브리오(Vibrio vulnificus) 패혈증에 미치는 영향

Phytic acid chelates excellently the metallic ions and the positive ions, especially has high affinity with Fe^(2+) and Ca^(2+). Merits of phytic acid can be taked in easily, edibile and harmless to body, so it was investigated that phytic acid can be substituted for EDTA in this study. 1. The Intensificative effect of chelating agent and disinfective osmotic shock of Vibrio vulnificus The number of initial existent fungi measured 1.7x10^(6). The percentages of the survival fungi against the osmotic shock by distillated water were calculated at 1 minute, 3 minute and 5 minute after inoculation. The percentages of the survival fungi in Mg^(2+) were 92.5%, 91.8% and 79.8% at each time, the average percentage was 88%. Also the sudden extinction was observed around 1 minute after inoculation and the survival fungi were not observed from 3 through 5 minute in spite of repeated experimentation. 2. Influence of Vibrio vulnificus on the survival of the mice. The first mouse started to die in 180 minute after inoculation in case that the inoculating number was 2.3x10^(7) cfu/ml. All died within 4.5 hour. The average of survival time was 226 minute. The first mouse started to die in 228 minute after inoculation in case that the inoculating number was 0.8x10^(6) cfu/ml. All died within 5 hour. The average of survival time was 300 minute and the survival time was 1.3 times high. The tendencies of death in two cases were similar, but the fatal rate were largely dependent on inoculating number.
