        필자는 2004년 3월~2006년 12월 주그리스 대사로 재임하는 동안 세 가지 monumental events와 함께 하는 영광을 가졌다. 우선, 2004년 8월 아테네 올림픽과 우리나라 참가가 먼저 떠오른다. 남ㆍ북한 선수단이 한반도기를 앞세우고 아테네 올림픽 개ㆍ폐회식에 공동 입장한 것은 아테네 올림픽의 백미였다. 두 번째는 2006년 9월 3일~9월 5일 한국 대통령으로서는 최초로 노무현 대통령의 그리스 국빈 방문이 실현되어 한ㆍ그리스 관계에 새로운 장을 열었다는 점이다. 세 번째는 50여 년 만에 성사된 그리스의 유엔안보리 비상임 이사국 진출과 반기문 유엔 사무총장 선출이 절묘하게 시기적으로 연결된 점이다. 노 대통령의 2006년 9월 초 그리스 국빈 방문은 그리스의 반기문 후보 지지를 사실상 확약 받는 계기가 되었다. 대사 재임 중 역사의 현장에 참여할 수 있었던 것은 여간 축복이 아닐 수 없다. 이와 같은 행운을 운동 경기에 비유하여 아테네 생활의 트리플 크라운(triple crown)이라고 명명하고 싶다. My ambassadorial posting to Greece coincided with three monumental events with consequences for mankind’s civilization as well as the relations among nations ; namely, Athen’s hosting of the 28th Summer Olympics in August 2004, President Roh Moo-hyun’s state visit to Greece at the beginning of September 2006 and Greece’s assumption of the non-permanent membership of the UN Security Council for the terms of 2005-2006. In parallel, the campaign by Foreign Minister Ban Ki-moon to seek the office of the top diplomat at the UN was in full swing. I was quite privileged to be present at the creation of history. It did represent the highlights of the Athen’s sports extravaganza that the athletes both from South and North Korea marched together at the opening and closing ceremonies of the Athen’s Olympics. President Roh Moo-hyun’s state visit, the first ever by a sitting Korean President, to Greece, opened a new chapter in the growing partnership between the two countries. Greece’s support for candidate Ban Ki-moon, meanwhile, proved instrumental in his eventual winning the appointment to the position of the UNSG. In a nutshell, I would like to hold these extraordinary experiences dear as triple crown, as is often referred to in sports achievements.

        6-[(N-3,4-디플루오로페닐)아미노]-7-클로로-5,8-퀴놀린디온의 항진균작용 및 안전성 평가

        6-[(N-3,4-Difluorophenyl)amino]-7-chloro-5,8-quinolinedione(RCK4) was tested for antifungal activities, against systemic infections with Candida albicans in normal mice. The therapeutic potential of RCK4 had been assessed in comparison with ketoconazole and fluconazole. RCK4 had ED50, 0.30 +/- 0.14mg/kg but ketoconazole and fluconazole had ED50, 8.00 +/- 0.73, 10.00 +/- 0.43mg/kg respectively. Intraperitoneally administered RCK3 at the ED50 for 7 days and 14 days reduced Candida albicans colony count in the kidneys and liver as well as ketoconazole and fluconazole at these ED50. And administered RCK4 at the ED50 for 14 days improved survival rates as well as ketoconazole. Acute oral toxicity studies of RCK4 were carried out in ICR mice of both sexes. These acute oral toxicities of RCK4 were low and LD50 values were over 2,850mg/kg in ICR mice. The genotoxicities of RCK4 had been evaluated. RCK4 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. The clastogenicity was tested on the RCK4 with in vivo mouse micronucleus assay. RCK4 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. These results indicate that RCK4 has no genotoxic potential under these experimental conditions.

        Xenopus 수정란에 미세주입된 ${\lambda}-DNA$의 배발생에 미치는 영향 및 미세 구조에 관한 연구

        In an attempt to produce transgenic amphibia, bacteriophage ${\lambda}-DNA$ was microinjected into fertilized eggs of Xenopus laevis, and the effect on early embryogenesis and the ultrastructural behavior of exogenous DNA were investigated. The effect of microinjected gene on embryonic development showed differences according to the concentration of injected DNA and the incubation temperature. Various concentrations of ${\lambda}-DNA$ were tested. Among those, microinjection of 1-2 ng DNA dissolved in 20 nl TE buffer was not shown to disturb normal embryonic development and was recorded the highest survivability to the late tadpole stage (Stage 43); however, injection of increased concentrations of DNA than above provoked irregular cleavages or abnormal appearances, which resulted in reduced survivability. When the injected embryos were incubated at low temperatures (e.g., $12^{\circ}C$), 54.5% of the embryos developed to Stage 43, whereas 42.4% survived when incubated at room temperature. The survivability showed also differences according to the injection site. 58.0% of the embryos developed to Stage 43 when microinjected into the vegetal pole, whereas 44.9% survived when microinjected into the animal pole. To understand the structural fate or behavior of injected DNA a combined light and electron microscopical study was applied. The nucleus-like structure was observed in the ${\lambda}$ DNA-injected embryos, which was quite a similar to the interphase nuclei of normal Xenopus laevis. The nucleus-like structure showed the typical double-layered nuclear membrane and nuclear complexes; however, it consisted of unusual structures such as furrows of nuclear envelope into the nucleoplasm.

        Xenopus laevis 초기 발생과정에서 Ubiquitin 의 발현양상과 Ubiquitin Conjugates 의 분포

        N/A By RT-PCR analysis, we confirmed that Xenopus ubiquitin (Xub) is constitutively expressed at a high basal level during all stages of embryogenesis. In situ hybridization was performed to study in more detail the spatial expression pattern on Xub. The first attractable signals were observed in the neural fold and in the anteriormost region of early neurula. At the tailbud stage, the signals concentrated in cells of the developing brain, otic vesicle, branchial arches, pronephros and somites. As development progressed Xub transcript gradually localized to abdominal region. We also have examined immunohistochemically the distribution of ubiquitin-conjugated proteins using a monoclonal antibody raised against polyubiquitin of Amoeba proteus. The immunostaining patterns coincided largely with the expression patterns of Xub. Futhermore, we compared the expression patterns of Xub with TUNEL staining to identify a relationship between ubiquitin and programmed cell death during the development of Xenopus.

        멕시코산 Axolotl , Ambystoma mexicanum 의 FGF - 8 cDNA 절편 클로닝

        FGF-8 (fibroblast growth factor-8) has been known to play crucial roles in various developmental processes such as initiation, outgrowth and patterning of vertebrate limbs, especially in amniotes. However, it is not known whether FGF-8 plays similar roles in the developmental processes of amphibians. Here, we report a partial fragment cloning of an FGF-8 cDNA from embryo and limb blastema cDNA libraries of a salamander, Mexican axolotl (Ambystoma mexicanum). The 1.27 kb cDNA fragment cloned in the present study contains a coding region covering 192 amino acid residues with conserved consensus amino acid residues of FGF family and shows 85% amino acid identity with that of mouse, and 91% amino acid identity with those of chick and Xenopus. Therefore, the cDNA clone obtained in the present study is believed to be a partial fragment of a putative FGF-8 cDNA of Mexican axolotl.

        Partial Cloning of Xhox3.1 Gene in Xenopus laevis

        VNTR(variable number tandem repeat) loci in the genome are characterized-as their copy number of the same repeat unit and are extremely polymorphic having potentially hundreds of alleles at a single locus. In this study, allele distribution on the hTPO VNTR marker was analyzed by PCR(polymerase chain reaction) in Korean population(100 individuals). Twenty two alleles were identified in the hTPO VNTR locus and alleles 7(8.5%), 10(11.0%), and 15(15.0%) showed three highest peaks. The allele frequency of hTPO VNTR locus is in the 95% confidence interval. PIC(polymorphism information content) value calculated on hTPO VNTR loci was 0.92. POE(power of exclusion) value used in paternity cases was 0.80. MP(matching probability) used in forensic identification was 0.018. In conclusion, statistical data from hTPO VNTR locus showed this locus is a very useful genetic marker for the linkage study, evolutionary genetics and forensic application.

        Xenopus laevis 의 초기 발생에서의 Xc - myc protooncogene 의 발현 양상

        Temporal and spatial expression pattern of Xc-myc protoocogene was studied in the early development of Xenopus laevis. The myc mRNA is localized in animal hemisphere of early stage embryos. From the gastrula stage Xc-myc gene is expressed, mainly in dorsal area and the expression of the gene is maintained persistantly in neuroectoderm and epidermis. After tailbud stage, strong expression of Xc-myc gene is observed in several developing tissues and organs originated from ectoderm and mesoderm, such as central nervous system, optic cup, lens placode, otic placode, gill bud, cement gland as well as in the somite and epidermis. However no expression was observed in the endoderm at every developmental stage. Animal cap and equatorial zone of blastrulae, in additiort to ectoderm and mesoderm of exogastrulae showed that the expression was occurred these areas regardless of the presence of inductive activity. These observations suggest that the expression of Xc-myc protooncogene plays important roles in the development of ectodermal and mesodermal derivatives in a specific way.
