        Populus alba × P . grandidentata 조직배양묘의 (組織培養苗) 토양에서의 활착과 생장

        This study was undertaken to find out the effects of three kinds of potting media and two sources of explants on the survival and early growth of new plantlets of Poputus alba × P. grandidentata in the greenhouse. The results obtained can be summarized as follows; 1) Among three potting media, Terralite was best for early growth and survival of plantlets. 2) Like humidifier, an intermittent misting system can be effective in keeping relative humidity high for the plantlets. 3) Survival rates over 80% could be obtained if humidity was kept high during the hardening period. 4) During hardening period, the plantlets showed the juvenile characteristics such as smaller leaves, thinner stems, and shorter internodes. 5) There were no differences on morphological characteristics between the plantlets originating from axillary buds and the plantlets originating from multiple shoots while they were growing at the greenhouse. 6) The plantlets originating from bud culture grew normally comparing to regular cuttings.

        Organ Specific Expression of the nos-NPT II Gene in Transgenic Hybrid Poplar

        임목을 대상으로 삽입된 외래 유전자의 공간적, 시기별 발현 특성을 이해하기 위한 기초연구로서 온실에서 생육 중인 형질전환된 2년생 잡종 포플러 (Populus alba X P. grandidentata) Hansen 클론을 대상으로 삽입된 외래 유전자의 발현정도를 각 기관별로 조사하였다. Agrobacterium binary vector pRT45, pRT102 및 pRT104에 의해서 형질전환된 3계통의 형질전환체 Tr15, Tr345, Tr665 모두는 선발가능한 표식 유전자로서 nos promoter-NPT II 유전자가 대상 식물체의 genome에 삽입되어 있으며, 그외에, pin2 promoter-CAT 유전자(pRT45), nos promoter-PIN2 유전자(pRT102), Cauliflower Mosaic Virus 35s promoter-PIN2 유전자(pRT104)가 3계통의 형질전환체에 제각각 삽입되어 있는 잡종 포플러이다. 이들 3계통의 형질전환 포플러 식물체의 DNA를 PCR 검정 기법을 이용하여 분석해 본 결과 선발 가능한 표식 유전자인 NPT-II가 삽입되어 있음이 입증되었다 발현 정도를 비교 분석하기 위해서 NPT-ELISA 검정을 실시하였다. 삽입된 NPT II 유전자는 형질전환된 포플러의 잎, 엽병, 형성층 조직, 줄기의 목질부, 뿌리에서 발현되었으며, 발현 정도는 형질전환된 식물체의 계통에 따라서, 그리고 형진전환된 식물체의 부위에 따라서 다양하게 나타났다. pRT45에 의해서 형질전환된 Tr15 형질전환체의 경우, 늙은 잎과 엽병에서 NPT II 유전자가 가장 높은 수준으로 발현되었으며, 어린 잎과 뿌리 조직에서 가장 낮게 발현되었다. 삽입된 외래 유전자가 각 식물체간에, 각 기관에 따라서 각각 상이한 발현 정도를 나타내는 이와 같은 결과는 형질전환된 식물체에 대한 효과적인 선발과정이 요구됨을 의미함은 물론이고, 형질전환 식물체의 발달 과정에 따라서 삽입된 외래 유전자가 공간적, 시기적으로 각각 다르게 발현할 수 있다는 것을 나타낸다. To effectively modify tree function with genetic engineering, transgenes must be expressed at the proper level in the appropriate tissues at suitable developmental stages. Toward understanding the spatial and temporal expression of transgenes in woody plants, transgene expression was evaluated in three greenhouse-grown, transgenic lines of Populus alba ${\times}$ P. grandidentata hybrid clone 'Hansen'. All transgenic poplar lines possess constructs containing the bacterial nopaline synthase(nos) promoter linked to a neomycin phosphotransferase II(NPT II) selectable marker gene. In addition, each transgenic poplar line contains one of the following gene constructs : 1) a wound-inducible potato proteinase inhibitor II (pin2) promoter linked to a chloramphenicol acetyltransferase(CAT) reporter gene. 2) a nos promoter linked to a PIN2 structural gene : or 3) a Cauliflower Mosaic Virus 35s promoter linked to a PIN2 structural gene. Polymerase chain reaction(PCR) was used to verify the presence of foreign genes in the poplar genome. Enzyme-linked immunosorbent assays(ELISAs) were used to evaluate organ specific expression of the nos-NPT II construct. NPT II expression was detected in leaves, petioles, stems, and roots of transgenic poplar, thereby indicating that the nos promoter is potentially effective for general constitutive expression of transgenes. NPT expression varied among transgenic poplar lines and among organs for one transgenic line, Tr15. With Tr15, NPT II levels were highest in older leaves and petioles. These results indicate that screening of several transgenic lines may be required to identify lines with optimal transgene expression.

        Agrobacterium Binary Vector 에 의한 포플러 형질전환을 위한 기초연구

        Three clones of Populus alba × P. grandidemtata have been tested for susceptibility to Agrobacterium tumefaciens strains A281 and A348. We determined the optimum concentration of kanamycin sulfate for effective selection of leaf disc-derived, transgenic tissues transformed using Agrobacterium binary vector pGA472 containing a neomycin phosphotransferase gene (NPT-II) which confers kanamycin resistance. Of the wild type Ti plasmids contained by the two Agrobacterium strains, pTiBo542 of strain A281 appears Co be best suited to serve as a helper plasmid for binary vector systems. A relatively low concentration (10㎎/ℓ) of kanamycin sulfate inhibited adventitious shoot initiation from leaf discs on regeneration medium. Transformed kanamycin-resistant calli were obtained by culturing Agrobacterium inoculated leaf discs on selective regeneration medium. The transformed kanamycin-resistant calli continued to grow on regeneration media supplemented with kanamycin sulfate to levels of 50 and 204㎎/ℓ. The growth of non-cultivated control calli was severely inhibited on regeneration medium containing 50㎎/ℓ kanamycin sulfate.

        형질 전환된 포플러에 대한 nos - NPT Ⅱ 유전자의 기관별 발현 특성

        Polyploidy induction in the crandon clone of Populus alba × P. grandidentata by colchicine treatment

        Crandon clone of Populus alba×P. grandidentata was treated with various colchicine concentrations and treatment durations to determine the best method for inducing polyploidy through in vitro treatment of axillary buds. Colchiploid shoots were only produced from in vivo, but not from in vitro colchicine treatments. Most colchiploid shoots originated from buds that were treated 24 h. duration with 1% colchicine solution and 48 h. duration with 2% colchicine solution. Twelve colchiploid shoots produced from in vivo colchicine treatment continued to exhibit the chimera leaf shape after cuttings and have mixed chromosome numbers, 2n (38) and 4n (76). The adventitious shoots produced from chimeric leaf discs through in vitro culture appeared either normal or colchiploid leaf shape with thicker stem diameter. The colchiploid adventitious shoots had the mixed 2n-4n chromosome numbers.

        Populus alba × P . grandidentata 조직배양 식물체 원형질의 분리와 배양

        Protoplast-source meterial and enzyme strength had a significant influence on protoplast yield from hybrid poplar, Populus alba × P. grandidentata. The yield of protoplasts from in vitro culture of 1 month-old plantlets was more than that from greenhouse grown 4 month-old stock plant. In vitro cultured plantlets regulary produced more viable protoplasts with E-I enzyme solution (0.5% cellulase and 0.1% macerase) than those with E-II enzyme solution (1.0% cellulase and 0.2% macerase) after overnight incubation. The mean yield of protoplasts from in vitro cultured plantlets was 4 × 10^6 with E-I enzyme solution. Cell division was observed in these protoplast cultures after 7-10 days. Protoplast-derived hybrid poplar cells survived over 3 weeks in culture and some continuous cell divisions were evident. Other aspects associated with protoplasts from in vitro cultured plantlet are also discussed.

        한국 숲 운동에서의 다영역간 파트너십 사례연구

        한국 숲 운동의 대표적인 사례인 생명의 숲 가꾸기 국민운동은 IMF 사태로 파생된 국가적 위기 상황을 극복하기 위한 하나의 수단으로 시민사 회단체, 산림전문가, 기업, 산림청 등이 다영역간 파트너십을 구축하여 숲 가꾸기 사업을 제안하였고, 정부에서는 그러한 제안을 하나의 정책으로 반영하여 시행함으로써 지난 4년여 동안 하루 2만여 명을 고용할 수 있는 새로운 일자리를 창출하는 한편 압축고도성장기에 방치해 두었던 40여만 ha의 산림도 가꾸는 사업을 펼 수 있었다 숲 가꾸기 사업의 지속과 전국적 확산은 시민단체, 학계, 기업 및 산림청의 조화로운 파트너십의 결과였다 이런 다영역간 파트너십은 북한의 산림녹화를 위한 평화의 숲 운동, 학교 녹화를 위한 학교 숲 운동, 중국과 몽골 및 극동러시아의 산림문제와 사막화 방지를 위한 동북아 산림포럼 등의 모태가 되었으며, 국민적 관심을 산림에 모울 수 있는 계기를 제공하기도 하였다 한편 본고에서는 숲 운동의 전개를 위한 각 영역별 역할과 기능, 숲 운동이 우리 사회에 끼친 영향, 숲 운동에서의 다영역간 파트너십에 대한 평가, 숲 운동에서의 다영역간 파트너십이 극복해야 할 과제 등도 함께 논의되었다 Citizen groups, corporations, forestry experts, and Forest Service were united under the common goals for Jobs and enhancing the quality of forests when Asian financial crisis hit Korean economy hard and massive number of workers were driven out to streets without any job This is how Forest of Life Movement was born and how a typical example of multi sectoral partnership for common benefit was established. It was the Forest Service that actually implemented forest tending projects a cross the country which have created 20,000 job a day fot the last 4 years and 400,000 ha of forests became rejuvenated However, multi sectoral partnership in forest movement should get credit for the successful implementation of the projects because they had laid the foundation for such movement through their interests and participations in forestry issues. Forest of Life Movement ignited citizen groups aggressive involvement in forestry issues and gave birth of other citizens groups such as Northeast Asia Forest Forum, Forests of Peace, and Greening School Yard Campaign Forest of Life also pursued a goal that can realize nature conservation, sense of community, harmony with nature, and respect for life. In this paper, the function and role of the each sector for partnership, social impacts of forest movement, assessment of the multi sectoral partnership, and the problems to overcome the barriers of forest movement were also discussed
