        해동피추출물이 실험적으로 유발된 가토의 간장기능장애에 미치는 영향

        해동피 methanol 추출물을 시료로 mouse의 급성독성실험, 실험적으로 유발시킨 가토의 간장기능 장애에 미치는 영향을 실험하여 다음과 같은 결과를 얻었다.<br/> 실험결과 해동피 methanol 추출물의 급성독성 (LD_(50))=7.3g/㎏으로 무독하였고 CCl₄로 손상시킨 가토의 간장기능회복에 있어서는 실험동물체중 ㎏당 시료 800㎎투여군에서 14일째 대조군에 비해 SGPT가 87.28±23.03 unit로 서서히 유의성있게 회복되었다. 시료 800㎎/㎏투여군에서 alkaline phosphatase 활성도는 2일째부터, 그리고 total bilirubin 측정치는 6일째부터 현저히 회복되는 경향을 보였다.<br/> 특히 total cholesterol 측정치는 200㎎/㎏투여군에서 14일째 99.36 12.42㎎/㎗과 800㎎/㎏투여군에서 10일째 96.69 18.40㎎/㎗, 14일째 80.68 8.77㎎/㎗로 대조군에 비해 현저한 감소를 보였다. This study was carried out to investigate effects of Kalopanaxii Cortex extract(K. C. E,) on CCl₄-induced liver damage in rabbits and acute toxicity in mice.<br/> 1. LD_(50)(㎎/㎏g) of K. C. E. was 7.3g/㎏ by intraperitoneal administration in mice.<br/> 2. K. C. E. groups showed more rapid recovery than the control group in CCl₄-intoxicated rabbits and 800㎎/㎏ was the most effective.<br/> 3. SGPT and alkaline phosphatase activity showed and apparent decreasing effect within 14 days and 2 days respectively in 800㎎/㎏.<br/> 4. The levels of total cholesterol and total bilirubin showed and apparent decreasing effect within 10 days and 6 days respectively in 800㎎/㎏.<br/> It is suggested that K. C. E. can be administered not only as a therapeutic agent but also a healthy food to shorten the recovery time of hepatic function in liver diseases.

        토사자 추출물이 실험적으로 유발된 가토의 간장기능장애에 미치는 영향

        This study was carried out to investigate effects of Cuscutae semen extract(C. S. E) on CCl₄-induced liver damage in rabbits and acute toxicity in mice.<br/> LD_(50)(㎎/㎏) of C. S. E. was 7.83g/㎏ by intraperitoneal administration in mice.<br/> C. S. E. groups were showed more rapid recuperation than the control group intoxicated by CCl₄ only.<br/> SGPT activity was lowered significantly in 800㎎/㎏ after 6 days.<br/> The levels of total cholesterol and total bilirubin and alkaline phosphatase were decreased apparently in 800㎎/㎏ from 6 days.<br/> It is suggested that C. S. E. can be administered not only as a therapeuticagent(such as liver tonics) but also a healthy food to shorten the recovery time in liver diseases.

        枸杞子 抽出物이 家兎의 實驗的 肝臟障碍 및 alloxan糖尿病에 미치는 影響

        枸杞子抽出物을 試料로 mouse의 毒性實驗, 實驗的으로 誘發시킨 家兎의 肝臟機能障碍 및 alloxan負荷糖尿病에 미치는 影響을 實驗하여 다음과 같은 結果를 얻었다.<br/> 1. Mouse의 急性毒性實驗에서 腹腔投與의 경우 平均致死量은 12.7g/㎏이었다.<br/> 2. CCl₄로 肝臟機能障碍가 誘發된 家兎에 枸杞子抽出物投與는 對照群에 비하여 有意性있는 回復度를 보였는데 特히 大量投與群에서 더욱 빨랐다.<br/> 1) GPT活性度 및 total bilirubin은 試料投與群에서 아주 優秀한 低下效果를 나타냈다. 特히 GPT活性度에서 200㎎/㎏은 第8日째부터, 800㎎/㎏은 第6日째부터 有意性이 있었다.<br/> 2) Alkaline phosphatase値는 對照群에 비하여 커다란 차이를 보이지 않았다.<br/> 3) Alloxan으로 誘發한 家兎의 高血糖에 대하여 800㎎/㎏는 아주 탁월한 血糖降下效果를 나타냈다. 卽 第4日째부터 對照群에 비하여 有意性이 있었으며 第12日째는 正常値에 거의 接近하였다. 또한 GPT活性度는 차츰 低下되어 第6日째부터 低下效果가 뚜렷하였다. These studies were carried out to investigate effects of Lycii fructus extract on experimentally CCl4-induced liver damage and alloxan-induced diabetes in rabbits and acute toxicity using mice.<br/> LD_(50)(㎎/㎏) of Lycii fructus extract(L.F.E.) was 12.17g/㎏ by intraperitoneal administration in mice.<br/> L.F.E. showed more rapid recuperation compared to the control group in CCl4-intoxicated rabbits and 800㎎/㎏ was the most effective.<br/> Especially GPT activity and total bilirubin level showed an apparant decreasing effect within 6 days and 8 days, respectively in 800㎎/㎏.<br/> But any differences were not observed in alkaline phosphatase and total cholesterol between sample and control group.<br/> Large amount administered group exhibited more excellent hypoglycemic effect in alloxan-induced diabetes of rabbits, that is, it was significant to the control group after 4 days and adjacent to the normal level on 12th day. And GPT activity was gradually decreased and showed clear decreasing effect after 6 days.<br/> It is suggested that L.F.E. can be administered not only as therapeutic agents (such as liver tonics or antidiabetetics) but also a natural food to shorten the recovery time of hepatic function in liver diseases and decrease the abnormally elevated blood glucose such as Diabetes Mellitus.

      • Chitosan Matrix에 Pepsin의 固定化

        For the effective immobilization of pepsin on chitosan matrix with glutaraldehyde, optimal activation methods were studied, and its enzymatic properties were investigated. In immobilizing pepsin activity experiments where it was immobilized on to chitosan matrix with glutaraldehyde, we found that the immobilized pepsin retained 65% activity. The optimum pH of the soluble pepsin was 2.0. while immobilized pepsin was 1.5. The effect of temperature on the activity of the immobilized pepsin was compared with that of the native pepsin. The result indicated that the native pepsin was 35℃, while immobilized pepsin was most active over a temperature range from 35℃ to 65℃. From Line Weaver-Burk polts, the following kinetic data were abtained: for the immbolized pepsin, Km=0.417% and Vmax=0.769㎛/L:for the native pepsin, Km=0.476% and Vmax=1.333μM/L.

        폴리우레탄 - 폴리에칠렌옥사이드 - 폴리프로필렌옥사이드 디바이스로부터의 헤파린 방출

        The release of heparin from monolithic devices composed of different ratios of polyethylene oxide-polypropylene oxide (PEO-PPO) and hydrophobic polyurethane was investigated. The release rate of heparin could be controlled by varying the PEO-PPO content. The heparin release rate from the devices increased as the content of PEO-PPO in the devices increased. The release mechanism may be associated with creation of micro-channels and pores through the devices following the change in the physical structure of the polymer network. Hydrophobic polyurethane containing PEO-PPO can provide an antithrombogenic material for prolonged release of heparin from a heparin blended system.
