Paraffin 포매 조직 표본 제 작에 서 Microwave를 이 용한 간장과 신장조직의 고정에 관한 연구

This study was undertaken to evaluate the ef- feet of rapid heat fixation of specimens using microwave irradiation. Small portions of liver and the kidney specimen fixed by microwave ir- radiation and subsequently processed by paraffin embedding for light microscopical study. The control specimens were fixed by 10% neutral buffered formalin and processed by same method. Both were stained with hematoxylin and eosin, periodic acid-Schiff, reticulum and Masson``s trichrome. The results were as follows: 1. The tissue structures were destroyed and staining property of cells was poor by micro wave irradiation at low temperature. 2. There was a variable optimal conditions of fixation with different tissues. Optimal condition in this experiment were 135 seconds irradiation, 60 ± 2 oC, for the liver and 175 seconds irradiation, 70±2 °C, for the kidney. In these conditions, the preserving if the cellular structures and the stainability were better than those of the control. 3. There were no differences between the microwave fixation and the formalin fixation in the stainability of all special stains. Finally, the advantage of the microwave fixation is able to shorten the time of tissue preparation and get the good morphology and stainability.
