        3,4,5-Trihydroxybenzoic Acid Methylester와 관련 화합물의 피부암 및 구강암 세포주에 대한 세포독성

        The cytotoxic activity of 33,4,5-trihydroxybenzoic acid methylester and related compounds on the growth of normal cell lines, human skin melanoma cells and human oral epithelioid cell line were evaluated by the 3-[4,5-dimethylthiazol- 2-yl]-2,5-diphenyltetrazolium bromide (MTT) and 2,3-bis-[2-methoxy-4-nitro-5-sulfo-phenyl]-2-H-tetrazolium-5-caboxanilide (XTT) methods. 3,4,5-Trihydroxybenzoic acid methylester decreased the cell viability of human skin melanoma cells and human oral epithelioid cells shown by the MTT method and the cell adhesion activity of human skin melanoma cells and human oral epithelioid cells shown by the XTT method. In light microscopy, 100 μM 3,4,5-trihydroxybenzoic acid methylester showed the highest cytotoxic activity. These results suggest that 3,4,5-trihydroxybenzoic acid methylester has a potential anticancer activity.

        3,4,5-Trihydroxybenzoic Acid와 관련 화합물의 구강암 세포주에 대한 세포독성효과

        Objectives : In order to evaluate the cytotoxicity of 3,4,5-trihydroxybenzoic acid and related compounds on the growth of normal cell lines and human oral epithelioid cell line, cell viability, cell adhesion ability, and morphological changes of cells were examined. Methods : We measured the cytotoxicity of 3,4,5-trihydroxybenzoic acid and related compounds with 3-[4,5-dimethyl-thiazol-2-yl]-2,5-diphenyltetrazolium bromide-[MTT), and 2,3-bis-[2-methoxy-4-nitro-5-sulfophenyl]-2H-tetrazolium -5-caboxanilide-[XTT) methods. Results : The cytotoxicity of 3,4,5-trihydroxybenzoic acid($IC_{50}$, $2,552.40\;{\mu}M$) was low according to the toxic criteria. Cytotoxic effect of 3,4,5-trihydroxybenzoic acid and related compounds against $IC_{50}$ value in cell morphology increased in a concentration-dependent manner. In light microscopy, $100\;{\mu}M$ 3,4,5-trihydroxy-benzoic acid showed th highest cytotoxic activity. Conclusions : These results suggest that 3,4,5-trihydroxybenzoic acid may have a potential anticancer activity.

        벤조산 유도체의 Chromium Trioxide 독성에 대한 세포 재생효과

        In order to evaluate the cytotoxicity of chromium trioxide, and the cell regenerative effect of phenolic acid against chromium trioxide-induced cytotoxicity, cell viability, cell adhesion activity, lactate dehydrogenase (LDH) activity, and morphological changes of cells were performed in these cultures. The toxicity of chromium trioxide (${IC}_{50}$, 44.0 ${\mu}M$) was high according to the toxic criteria. Cell regeneration of benzoic acid derivatives against ${IC}_{50}$ value of chromium trioxide in cell morphology was increased in concentration-dependent manner. These results suggest that benzoic acid derivatives may be used as a cell regenerative agent against chromium-mediated cytotoxicity.

        온열 발반사 요법이 비만여성의 우울지수 및 혈청 성분에 미치는 영향

        This study investigated obese women's general environment, mental depression changes by thermo foot-reflexology (50 minutes/one time and three times/week during 6 weeks) through a questionnaire survey of 33 obese women possessing over 23 body mass index (BMI) in Iksan, they divided into two groups and checked their triglyceride (TG), immunoglobulin G (IgG) and stress changes of blood by thermo foot-reflexology (TFR). As the results, depression changes of experimental group presented meaningful decline of about 7 value after TFR. Cortisol concentration of blood and stress index showed a decline in experimental group. TG and IgG concentration of experimental group showed meaning decrease by TFR. Therefore, we suggest that TFR did decline obese women's mental depression, stress and TG concentration without fall of immune system.

        측백잎 추출물의 사람정상상피세포에 대한 세포독성 및 항산화 효과

        많은 식물들은 오랫동안 질병치료나 식용을 위해 사용되어 왔다. 이러한 식물들은 가격이 저렴하면서도 부작용이 적을 뿐만 아니라 다양한 생리활성 성분을 보유하기 때문에 미용소재로써 활용되고 있다. 따라서 본 연구에서는 여섯 가지 식물 [측백, Thuja orientalis L.; 세이지, Salvia officinalis L.; 마르타로즈마리, Rosmarinus officinals L. (Marta Rosemary); 안나로즈마리, Rosmarinus officinals L. (Anna Rosemary); 벨로가민트, Citrus berhamia L., and 루, Ruta graveolens L.] 잎 추출물의 사람 정상상피세포(Beas-2B cells)에 대한 세포독성 및 현미경에 의한 형상학적 관찰과 항산화 활성을 측정하였다. 세포독성은 Sulforhodamine B(SRB)에 의해 측정하였으며, 현미경을 통해 세포모양을 관찰하였고, 1, 1-diphenyl-2-picrylhydrazyl(DPPH)방법에 의해 항산화활성을 측정하였다. 세포독성의 결과 측백과 벨로가민트 잎 추출물이 100 ㎍/mL의 농도에서 세포독성을 나타내지 않았으며, 세포모양도 정상적인 모양이었다. 항산화 활성 측정 결과 측백과 루 잎 추출물이 강한 항산화 활성을 나타내었다. 따라서 여섯가지 식물 추출물 중에서 측백잎 추출물이 사람정상상피세포에 세포독성을 나타내지 않으면서 강한 항산화 활성을 나타내었다. 그러므로, 본 연구를 통해 미용소재로서 측백잎 추출물의 가능성을 시사하였다.

        비듬균(Malassezia furfur)에 대한 식물 오일들의 항균활성 및 활성오일의 성분 분석

        비듬균(Malassezia furfur) 는 두피 질환을 일으키는 중요한 요소로써 오늘날 이러한 균들을 치료하는 치료제는 거의 없다. 몇몇의 전의 연구에서 오일들의 다양한 생물학적 활성이 보고되어졌지만 그것들의 항비듬균 활성은 거의 연구되지 않았고, 특히 Malassezia furfur에 대한 저해 활성은 보고되어 있지 않다. 따라서, 이 연구에서는 Malassezia furfur에 대한 식물 오일들의 저해활성을 평가하고, 이러한 오일 중에서 가장 활성이 좋은 오일을 가스 크로마토그래피(Gas chromatography-mass spectrometry)에 의해 성분을 분석하였다. 1차적으로 108개 오일의 스크리닝 과정에서 17개 오일[Allium sativum L., Carum carvi L., Coriandrum sativum L., Myroxylon balsamum var. pereirae Royle, Nepeta cataria L., Origanum vulgare L., Mentha pulegium L., Satureja hortensis L., Thymus vulgaris L. Thymus vulgaris L., Cinnamomum zeylanicum Blume, Pimenta dioica(L.) Merr., Eugenia caryophyllata Thumb.(Clove bud), Eugenia caryophyllata Thumb.(Clove leaf), Cymbopogon nardus L., Cymbopogon martinii Stapf., Citrus aurantifolia Swing. and Pimenta racemosa(Mill.) J.W.Moore] 이 2 mg/mL 의 농도에서 대조구인 이크라코나졸(itraconazole)과 비슷한 활성을 나타내었다. 이러한 17개 오일 중에서 Citrus auranifolia 오일이 가장 활성이 좋았으며, 처리농도 (1.5 mg/mL, 1.0 mg/mL, 0.5 mg/mL와 0.1 mg/mL) 에 따라 활성이 감소하는 경향을 보였다. 또한 Citrus auranifoli 오일의 가스크로마토그래피(GC-MS) 성분분석 결과 limonene, γ-terpinene and terpinolene의 함유율이 높은 것으로 나타났다. 그러므로 Citrus auranifolia 오일은 곰팡이 유래 질병들을 치료하기 위한 활성성분을 함유하고 있는 것으로 보인다. Malassezia furfur is an important causal factor for seborrheic dermatitis. Nowadays, the drugs available to treat this fungal infection are few. Several studies have documented the biological activity of essential oils. However, its antifungal properties are not completely understood, especially its anti-Malassezia activity. The aim of this study were to evaluate the effect of the plant essential oils on the growth of M. furfur using disk diffusion method and analyze by Gas chromatography-mass spectrometry (GC-MS) most active essential oils. In first screening, the 17 plant essential oils have possesses inhibitory activity against M. furfur at 2 mg/mL. Among the plant essential oils, oil of Citrus auranifoli was most active against M. furfur and its activity showed dose dependency. This anti-malassezial activity was high than that of itraconazole at 2 mg/mL. Oil of Citrus auranifolia also was phytochemically examined by GC-MS analysis, its main constituents were identified as limonene, γ-terpinene and terpinolene. It can be concluded that essential oils of Citrus auranifolia may have interesting applications to control fungal-derived diseases.

        단보 : Limonene의 세포독성과 항Malassezia 활성

        우리의 이전의 연구에서 Citrus auranifoli 오일은 Malassezia furfur와 Malassezia pachydermatis에 대해 항곰팡이 활성을 나타낸다는 것을 보여주었다. 본 연구에서 Citrus auranifoli 오일의 주요 성분인 리모넨(limonene)의 M. furfur와 M. pachydermatis에 대한 저해효과를 디크스 확산 방법(disk diffusion method)에 의해 측정하였고, Beas-2B 세포에 대한 세포독성을 cytopathic effect reduction 방법에 의해 측정하였다. Limonene의 최소곰팡이저해농도(minimum fungicidal concentration)는 양성대조구인 itraconazole보다 더 낮았고, 100 μg/mL의 농도에서 Beas-2B cells에 대한 세포독성을 나타내지 않았다. 그러나 itraconazole은 같은 농도에서 약한 세포독성을 나타내었다. 그러므로, 우리의 결과에서 limonene는 비세포독성과 함께 M. furfur and M. pachydermatis의 성장저해를 하는데 효과적이라는 것을 보여준다. A previous study of ours indicated that Citrus auranifoli oil possesses antifungal activity against Malassezia furfur and Malassezia pachydermatis. In this study, we evaluated the anti-M. furfur and M. pachydermatis activities of limonene, which is a major component of C. aurantifolia oil, using the disk diffusion method. We also examined cytotoxicity against human normal epithelial (Beas-2B) cells using the cytopathic effect reduction (CPE) method. The results revealed that the minimum fungicidal concentration (MFC) value of limonene is lower than the value for itraconazole. The MFC value of limonene was seen to be 7.81 μg/mL against M. furfur and 3.90 μg/mL against M. pachydermatis. MFC values of itraconazole against M. furfur and M. pachydermatis were 62.50 μg/mL and 31.25 μg/mL, respectively. In addition, it was noted that limonene was not toxic to Beas-2B cells with normal morphology at a concentration of 100 μg/mL. However, itraconazole exhibited weak toxicity at the same concentration. Therefore, our results indicate that limonene could potentially be effective at controlling M. furfur and M. pachydermatis infections with no cytotoxicity.

        발효, 세포배양, 생물공정 백하수오 발효액의 모발성장 효과

        The bacterial growth and pH of Pleuropterus cilinervis extracts and animal milk, fermented with Lactobacillus rhamnosus during the fermentation process, were evaluated. The results indicated that the bacterial count after fermentation always remained higher than 6 log CFU/mL, with a constant pH of approximately 4. In order to evaluate the effects of Pleuropterus cilinervis extracts and animal milk, fermented with Lactobacillus rhamnosus, on hair growth promotion, C57BL/6 mice were chosen as experimental subjects. Six week old males with similar body weights were divided into four groups; a normal group (saline), a negative control group (essence base), a positive control group and an experimental group (Pleuropterus cilinervis extracts and animal milk, fermented with Lactobacillus rhamnosus, mixed with negative control). The substances and test materials were applied topically on the back skin of the mice for 8days. Other external conditions and variables such as food intake were kept at the same as the four groups. At the end of the experiment it was noted that hair re-growth in the experimental group, using gross and histological examinations, was higher than that in the positive control group. This study therefore provides an empirical evidence that Pleuropterus cilinervis extracts and animal milk fermented with Lactobacillus rhamnosus promotes hair growth, and suggests that applications could be found in hair loss treatments.
