        Silver Nitrate 용액의 정소내 주입에 의한 가토의 거세효과에 관한 연구

        This experiment was carried out to develop a chemical castration with a single intratesticular injection of silver nitrate solution in domestic rabbits. A total of 44 bucks of 4 to 5 months old were alloted at random into the following 4 groups: 9 bucks for controls, 6 bucks for 0.3㎖ injection of 1% AgNO₃, 14 bucks for 0.5㎖ injection of 1% AgNO₃ and 15 bucks for 0.5㎖ injection of 5% AgNO₃. Changes in body weight, testicular width in scrotum. weight and histology of testis at necropsy, blood cell counts and plasma levels of testosterone of the buck were measured for 4 weeks after treatment. The results obtained were summarized as follows: Compared with the controls, the body weight gain of bucks treated with AgNO₃ was not significantly (P$lt;0.05) different during the experimental period for 4 weeks. The white blood cell counts were tended to be increased slightly but insignificantly 1 day after injection of AgNO₃ and then decreased to normal level soon. There was no significant change in red blood cell counts in the bucks treated with AgNO₃. Most of the testis of bucks were swollen significantly (P$lt;0.05) by 6 to 12㎜ in the width of testis in scrotum 1 day after injection of AgNO₃, but began to decrease in sue from 3 days after treatment and to their normal size in a week. The AgNO₃ injection was not sufficient to castrate some bucks successfully. Only 60.0 to 66.7% of the bucks treated with 0.0㎖ of 1 or 5% AgNO₃ solution showed the plasma levels of testosterone of !sower than 0.1 ng/㎖ by 28 days after treatment. This result was considered to be due to the fact that not a whole but a limited part of testis was injured by the present method of injection. The weight of testis of the bucks of which plasma testosterone were less than 0.1 ng/㎖ by 7 to 28 days after treatment was not significantly(P$lt;0.05) different between the treatments or periods after treatment. This result indicates that the period of 4 weeks following treatment might be insufficient for the injured testicular tissue to be necrotized and absorbed fully. The extent of damage of testis by injection of AgNO₃ was much different by the local effect in the parts of distance from injection. The proximal portion of testis injected with AgNO₃ was fully damaged and necrotized, but a distal part of testis was not severely damaged.

        완만동결법에 의한 생쥐의 정상배 및 분할배의 생존성에 관한 연구

        The effects of cryoprotectant, equilibration time and cell stage on the survival of intact and bisected mouse embryos were determined following slow freezing. The number of blastomeres during in vitro development was counted by nuclei staining with Hoechst 33342. The post-thaw survival rates of intact 8-cell embryos, morula and blastocysts after slow freezing were 62.8, 76.8 and 60.0% in 1.5 M glycerol, and were 64.4, 75.6 and 57.6% in 1.5 M DMSO, respectively. The survival rates of ZP-free or bisected morula after slow freezing were 60.5 and 48.6% in 1.5 M glycerol, respectively. The number of blastomeres following in vitro culture for 24 to 28 hours of fresh and frozen morula was significantly (p$lt;0.05) different as 53.6±1.3 and 40.0±1.0, respectively. The mean number of blastomeres following in vitro culture for 24 to 28 hours of bisected and bisected frozen morula was 25.0±1.0 and 21.8±0.8, respectively. When 531 fresh, 282 frozen, 130 bisected and 66 bisected frozen embryos were transferred to 46, 23, 15 and 12 recipient mice, 26.9, 21.7, 14.6 and 7.6% of them developed to normal offspring, respectively.

        Vitrification 방법에 의한 생쥐의 정상배 및 분할배의 생존성에 관한 연구

        This study was carried out to investigate the effects of cell stage, equilibration time, precoolmg, straw loading and dilution method on the post-thaw survival of intact, ZP-free or bisected mouse embryos. When embryos were vitrificated at two-step dilution in double columns following equilibration for 10 minutes. the significantly(P$lt;0.01) higher post-thaw survival rates were obtained with 8-cell embryos(91.3%) or morula (89.3%) than blastocysts(74.6%). Loading of morula in straws with double Medium-2 columns improved significantly(P$lt;0.01) their post-thaw survival rates(81.30%), compared with single Medium-2 columns(70.0%). Any significant effects on the post-thaw survival rate of embryos were not observed between the dilution methods and temperatures of Medium-2. The post-thaw survival rate of ZP-free or bisected embryos cryopreserved by the present vitrification was resulted in 73.5 and 68.8%, respectively.

        This experiment was carried out to investigate the developmental potential of bisected morulae and blastocysts, and the isolated blastomeres from 4-and 8-cell embryos in mouse. The results obtained were summarized as follows: l. The rates of blastomeres survived manipulation were 83.4 and 76.7%, in 4-and 8-cell embryos, respectively. 2. The rates of half embryos survived bisection were 77.7 and 75.3%. in morulae and blastocysts, respectively. 3. The rates of development of isolated blastorneres from 4-and 8-cell embryos were 24.7 and 10,4% respectively. 4. The rates of development of the bisected morulae and blastocysts were 67.7 and 71.9% respectively.
