당근 hairy root 배양에 의한 callus유기 및 anthocyanin생성

The experiments were conducted to optimize culture conditions for callus growth and anthocyanin formation by in vitro culture of Daucus carola callus derived from hairy root, which is induced by inoculation of Agrobaderium rhizogenes ATCC 15834. Callus induction from hairy root showed the best result in treatment with 0.2mg/l 2,4-D, and callus growth was suppressed by increasing concentrations of 2,4-D. For the selection of optimum medium, Nitsch & Nitsch medium supplemented with 0.2mg/l 2,4-D and 0. lmg /1 BA promoted callus growth, but severely suppressed anthocyanin formation. Hormone free MS medium was effective for both callus and anthocyanin formation. In treatments of plant growth regulators, 0.05 mg/1 2, 4-D hastened callus growth while 0.05mg/l NAA decreased anthocyanin formation. However, BA and zeatin were effective for callus growth compared with kinetin. High concentrations of cytokinins promoted anthocyanin formation, especially l.0mg/l BA gave the highest anthocyanin formation. On the other hand, cytokinins showed higher capacity for anthocyanin formation than auxins.
