한국에서 전통적으로 사용되고 있는 생약제제 중 베르베린, 계피산 및 글리시리진의 정량을 위한 간단하고, 정확하고, 재현성 있는 모세관 전기이동법을 개발하였다. 이 성분들의 분리는 25$^{\circ}C$, 20 mM 인산완충액(pH 7.5)에서 실리카 캐필러리($57 cm{\times}75 {\mu}m$ i.d.)를 사용하여 수행하였다. 350 V/cm의 전기장으로 베르베린, 계피산 및 글리시리진의 동시 정량을 13분내에 할 수 있었다. 검정곡선은 베르베린에 대해서는 1~100 ${\mu}g/mL$, 계피산은 0.3~100 ${\mu}g/mL$, 글리시리진은 2.5~100 ${\mu}g/mL$의 농도 범위 안에서 좋은 직선성을 보여 주었다. 이 성분들에 대한 상대 표준편차(n=5)의 범위는 0.96∼2.35%이었다. 베르베린, 계피산 및 글리시리진에 대한 검출한계(S/N=3)는 각각 0.5, 0.1 및 2.0 ${\mu}g/mL$이었다. 이론단수는 181,000(베르베린), 88,000(계피산) 및 169,000(글리시리진)이었다. HPLC에서는 3,100~4,800이었다. A simple, accurate and reproducible capillary electrophoresis(CE) assay has been developed for the determination of berberine, cinnamic acid, and glycyrrhizin which are used in traditional Korean medicinal preparations. Separation of these compounds was performed in 20 mM phosphate buffer(pH 7.5) and acetonitrile(75:25, v/v) using a bare fused silica capillary($57 cm{\times}75 {\mu}m$ i.d.) at 25$^{\circ}C$. With the electric field of 350 V/cm, the time needed for the separation of berberine, cinnamic acid and glycyrrhizin was within 13 min. Calibration curves were linear for 1∼100 ${\mu}g/mL$ berberine, 0.3∼100 ${\mu}g/mL$ cinnamic acid and 2.5∼100 ${\mu}g/mL$ glycyrrhizin. The ranges of relative standard deviations(n=5) for those samples were between 0.96∼2.35%. The limits of detection(S/N=3) for berberine, cinnamic acid and glycyrrhizin were 0.5, 0.1 and 2.0 ${\mu}g/mL$, respectively. The numbers of theoretical plates were 181,000(berberine), 88,000(cinnamic acid) and 169,000(glycyrrhizin), while they were 3,100∼4,800 in HPLC.

Single subcutaneous injection to SD rats of both sexes was performed to investigate the acute toxicity of new skin allergy-remedy ointment, Banaron. Banaron is composed of lidocaine hydrochloride, chloropheniramine maleate, prednisolone acetate, chlorohexidine hydrochloride, methyl salicylate, 1-menthol and dcamphor. The results were as fellows. LD_(50) values of Banaron were 8373.6 mg/kg for male and 8260.1 mg/kg for females. Death occurred within 24 hours after administration at doses up to 6600 mg/kg. The main cause of deaths seemed to be respiratory disturbance. General symptoms decreased of activity and respiratory rate, salivation, tremor and loss of consciousness which were commonly observed by some survived animals and all dead animals. No significant gross findings of internal organs and body weight changes in treatment groups in comparison with these of control group were observed at the maximum dose levels in Banaron.

Single oral administration to SD rats of both sexes were performed to investigate the acute toxicity of two new cough and cold remedies, WHS-1 and WHS-2. WHS-1 is composed of acetaminophen, chlorpheniramine maleate, cloperastine hydrochloride, dl-methylephedrine hydrochloride, caffein anhydrous, thiamine hydrochloride, riboflavin and serratiopeptidase. WHS-2 is composed of similar formula except that thiamine hydrochloride and riboflavin is not added. The results were as follows. LD_(50) values of WHS-1 were 4295.5 mg/kg for males and 4606.3 mg/kg for females, and LD_(50) values of WHS-2 were 3236.7 mg/kg for males and 4360.5 mg/kg for females. Death occurred within 2∼3 hours after administration at doses up to 2900 mg/kg in WHS-1 and 2500 mg/kg in WHS-2, the main cause of deaths seemed to be respiratory disturbance. General symptoms included decreased motor activity, salivation and loss of consciousness which were commonly observed in all dead animals treated with WHS-1 and WHS-2. No significant gross finding and body weight changes were observed at any dose level in the groups treated with WHS-1 and WHS-2.
