Bacillus species isolated from soil produced alkaline protease in alkaline media. This bacteria was especially good growth in alkaline media but no growth was observed in neutral media such as nutrient broth. Alkaline protease produced in various growth phases on the isolated strain were measured as following that enzyme was sligth produced at log phase but increased at stationary phase formed spore. Suupernantants prepared in 0.22 M TCA mixture (pH 10.5) show activity against both casein and hemoglobin. But this protease was higher activated against casein than hemoglobin. The enzyme was most active at pH 10.5and was most stable at pH 12.5 for 22hr incubation at 25℃. The optimum temperature for enzyme activity of enzyme was increased by addition of ?? at higher temperatures. The enzyme activity was affected by detergent contained cation(benzalkonium chloride) but not affected by detergent contained anion(sodium lauryl sulfate) and non-ion(Brij 35, Tween 80, Tritonx-100). The enzyme activity was not affected by metal ions such as ??, ??, ??, ??, ??, ??, ??, and ?? but was completely inhibitid by phenymethyl sulfonylfluoride (inhibitors of serine protease). These results indicate that thaes alkaline enzyme is a type of serine protease.
