        고정화 효소를 이용한 명태고기풀 수세액으로부터 Peptide 생산에 관한 연구

        Peptides separated from fish paste washing liquid of an Alaska pollak(Theragria chalcogramma) were purified and characterized. The fish paste washing liquid (supernatant) was separated by centrifugation of fish paste homogenate. The fish paste washing liquid of 0.5% concentration was hydrolyzed for 24 hour at 50℃ by immobilized protease in bioreactor and decomposing liquid of protein having 50% decomposing rate (OPA method) was obtained. The crude peptide fractions were obtained from this liquid by Dowex 50w(H^+) column chromatography. Purified peptides(SP-fraction peptides) were fractionated by using SP-Sepadex C-25(H^+) column chromatography. Molecular weights and amino acid compositions of these peptides were estimated by Sephadex G-50 column chromatography and HPLC, respectively. When the washed peptides was eluated with 0.6-0.9% and 1.2-2.0% of NaCl, peptides composed of weakly basic amino acids and strongly basic amino acid were respectively eluted. Molecular weights of each peptide fractions showed the broad distribution from 1,000 Da to 3,000 Da in the order of SP-4>SP-3>SP-2>SP-1. Peptides contained a large quantity of glycine, arginine, glutamic acid, and alanine in the washed peptide and its SP-fractions, respectively.

      • 駕莫灣內 細菌 Flora의 季節的 變動에 關한 硏究: 2. 大腸菌群의 分布에 關하여

        1989年 11月부터 1990年 10月까지 駕漠灣 海水의 水質評價를 위해 糞便汚染指標微生物인 大腸菌群을 測定한 結果는 다음과 같다. 1. 大腸菌群의 菌數測定結果는 A海域에서 4.0×⁻¹MPN/100ml 以下로 淸淨海域임을 알 수 있었고 D海域 8.1×10³MPN/100ml로 調査 海域中 가장 汚染되어 있었다. 2. 季節的 分布는 冬季인 11月과 12月, 다음해 1月과 2月에 거의 出現하지 않았고 5月에서 10月 사이 높은 分布를 나타내었다. 3. MacConky agar에 依한 大腸菌群數는 A, B, C海域에서는 夏季에, D海域에서는 年中 分布하는 傾向을 보였다. 4. 썰물과 밀물에 따른 大腸菌群의 菌數差는 밀물에 비해 썰물때 훨씬 높은 것으로 나타났다. 5. 生化學的 性狀에 따라 18類型으로 詳別되었다. 6. 分離菌 77菌株에 對한 同定結果는 E. coli Ⅰ,Ⅱ,Ⅲ가 7菌株, K. aerogenes Ⅰ,Ⅱ가 10菌株, C. freundii Ⅰ,Ⅱ가 15菌株였고 이들 菌種이 全體分離菌中 42%를 차지하였다. Kamak bay located under the both city of Yeochun and Yeosu is contaminated by domestic sewage and industrial waste water. On that ground, this study was conducted to evaluate and to obtain basic data on the sea quality of kamak bay through the investigation on coliform bacteria group being index microorganism of feacal contamination. The results examinated with sample divided into two groups of the clean and the contaminated sea area were as follow. 1. The viable cell counts of coliform bacteria in the clean and the contaminated sea area being 4.0×1⁻¹ and 8.1×10³ MPN/100ml respectively. we knew sea area that the former is clean and the latter to contaminate. 2. Seasonal distribution of these bacterials was not detected in January, February, November, and December during winter season, but it was appeared high count from May to October. 3. Coliform bacterial count measured by MacConky agar appeared tendency that A, B, and C sea areas were high distribution in the summer season and D sea area in all seasons. 4. Grouping according to bacteriological characteristics was divided into 18groups. 5. E. coli Ⅰ, Ⅱ, Ⅲ, K. aerogens Ⅰ, Ⅱ and C.ferundii Ⅰ, Ⅱ classified 7, 10 and 15 strains among 77 strains isolated from samples, respectively.

      • 여수연안 및 동중국해의 세균상: 3. 분리균중 protease 생산균의 동정 및 효소생산조건

        During 6th~14th August 1992, 23 strains having protease activity were isolated from East China sea. Two strains(F₄-188 and F₄-193) that protease activity is the most strong were seleted by second screen. Results investigated for enzyme producing condiction with identification of these two strains were as follow: F₄-188 and F₄-193 having the strong protease activity were classified with pseudomonas sp. and protease activity 65.5 unit/㎖, 74.6 unit/㎖, respectively. The optimum pH for enzyme activity was alkaline protease with pH 8.0 in F₄-188 strain and acid protease with pH 6.0 in F₄-193 strain. The highest protease activity for nitrogen source and carbon source was ammonium sulfate(1.7%) and starch(2.1%) in F₄-188 strain, albumin(1.5%) and α-cellulose(1.0%) in F₄-193 strain, respectivly. Effect of metal salt on the enzyme production in F₄-188 and F₄-193 strains was calcium chloride and magnecium sulfate, respectively. The optimum temperature for growth of two strains was 30℃ and heat-resisting property enzyme not to effect in 60 minutes at 40℃ but more that 90% were disappeared with heat treatment for 10 minutes at 80℃.

        여수연안 및 동중국해의 세균상: 분리균의 수평분포

        1992년 8월 6일부터 8월 14일 까지에 걸쳐 조사한 동중국해와 여수 연안에서 분리한 세균상은 174균주, uibrio spp. 132균주, 대장균군 183균주에 대한 분리동정 결과는 다음과 같다. 1. 동중국해 세균상은 분리균 74균주중 40균주가 Pseudomonas spp. (54%)였고, 여수 연안에서는 분리균 100균주중 60균주가 Enterobacteriaceae (60%)였다. 2. Vibrio spp.는 동중국해에서 분리된 5균주중 V. alginolyticus가 4균주, V. parahaemolyticus가 1균주 밖에 분리되지 않았고, 여수 연안에서는 127균주중 V alginolyticus가 54균주로 42.5%를 차지 하였으며,V. parahaemolyticus는 21균주로 16.5%를 차지하여 이들 두 균종이 Vibrio의 우점종이였다. 3. 대장균군은 동중국해에서 분리한 27균주중 E. coli I이 3균주, C. freundii I이 1균주였고 여수연안에서는 분리균 156균주중 E. coli I이 79균주 (50.6%)였고 C. freundii I, II가 각각 1균주였다. 4. 대장균군의 IMViC system, 44℃의 발육, gelatin 액화 실험에 의해 16균형으로 분별 되었고 비동정균은 8~16균형으로 분리균 183균주중 99균주로 54%를 차지하였으며 5. 5℃에서 발육 가능한 저온성 대장균군은 15균주였다. The results identified for bacterial flora 174, Vibrio spp.132, and coliform group 183 strains isolated from the areas of East China Sea and Vosu coastal sea during from August 6th. to 14th. 1992 were as follow: 40 strains among the 74 strains of bacteria flora isolated from East China sea area were Pseudomonas spp.(54%) and 60 strains among the 100 strains isolated from Vosu sea area were Enterobacteriaceae (60%). Four strains were Vibrio alginoliticus and one strain of V. parahaemolyticus among 5 strains of genus Vibrio isolated from East China Sea. While 54 strains were V. alginolyticus(43%) and V. parahaemolyticus (17%) among 127 strains genus Vibrio isolated from Yosu coastal sea area. Seventy nine strains among the 156 strains of coliform group isolated from Yosu sea area were Escherichia coli I (51%) and each one strain Citrobacter freundii I and II. 3 strains among 27 strains isolated from East China sea area were E. coli (11%) and 1 strain of C. freundii I. Coliform group was grouped into 16 types by IMViC system, 44℃, gelatin liquefaction test.

        駕莫灣內의 淸淨海域과 汚染海域間에 있어서 一般細菌 flora의 量的 分布를 調査하여 汚染階級에 對한 基礎資料를 얻고 分離菌의 生化學的 性狀을 통해 이들 海域間의 細菌相에 對한 差異点을 알아보기 위해 調査한 結果는 다음과 같다. 駕莫灣內 썰물과 밀물에 對한 年中 Microflora의 分布는 汚染海域에서 썰물때, 淸淨海域에서는 밀물때 優勢한 傾向을 보여주었다. 汚染階級으로 B, C, D海域에서는 밀물시 4月에서 7月, 썰물시 4月에서 10月사이 10³~10⁵/ml로 過營養域 이었고 淸淨海域인 A海域은 10²~10⁴/ml로 富營養域으로 나타났다. 細菌 flora는 分離菌 214 菌株中 Gram-positive 菌과 Gram-negative 菌으로 Pseudomonas, E-nterobacteriaceaeㆍMoraxella가 分離菌의 69%로 優勢하였다. 食鹽濃度는 3% Nacl과 75% ASW에서 生育可能한 菌株가 84.4%로 大部分 好鹽性 菌이었다. 至適溫度는 25℃에서 46.9%, 37℃에서 51%로 나타 났다. This study carried out to get a basic data on the contamination grade of the ocean according to the quantitative distribution of marine bacteria and to know a point of difference on bacterial genus in the clean and contaminated sea area of Kamk bay. The results are as follows : The quantitative distribution in this bay shown predominent tendency when it was ebb tide in the contaminative sea area and flood tide in the clean sea area. The contamination grade of the ocean in B, C, D sea area was supernutritive area with 10³~10⁵/ml when it was flood tide among April and July and ebb tide April and October. A sea area become to known with unpolluted sea was eutrophical area with 10²~10⁴/ml. Among 214 strains isolated from investigative sea area, Pseudomonas, Moraxella, Enterobacteriaceae and Gram-positive bactera predominated with 69‰.

        In order to prolong the period of storage, corn syrup 25% was added in low salt fermented squid-jeotkal. During the storing periods of this squid-jeotkal at 5℃, 15℃ and 25℃, total viable bacteria, proteolytic bacteria and this genus according to the period of storage were investigated. The results were as follows. Viable bacteria in the adding section and control increased in number of 4.1×10⁴ and 8.1×10⁶ CFU/g in the period of 50 days of storage at 5℃, respectively. The adding section of 15℃ was 4.7×10⁶ CFU/g during storage of 55 days and control 1.2×10⁸ CFU/g in the course of 40 days. Viable bacteria at 25℃ was more increase than at 5℃ an 15℃. So viable bacteria of the adding section and control were each 3.6×10⁷ and 4.3×10⁸ CFU/g. Proteolytic bacteria increase slowly up to 50 days of storage at 5℃ and so viable bacteria was 3.6×10³ CFU/g in the adding section and 4.8×10⁴ CFU/g in the control. These were each 100 CFU/g in 40 days at 15℃ of control and in 30 day at 25℃ of the adding section. The isolated bacteria was only staphylococcus sp. in the adding section but bacillus sp. and pseudomonas sp. besides staphylococcus sp. appeared in control.

      • 駕莫灣內 細菌 Flora의 季節的 變動에 關한 硏究: 1. 病原 Vibrio와 Salmonella에 關하여

        都市下水 및 工場廢水 流入으로 인한 生物棲息環境의 惡化로 生態界에 莫大한 支障을 招來하고 있는 駕莫灣海域의 微生物群集 특히 病原 Vibrio 와 病原腸內 細菌을 調査하여 이들 細菌과 水域生態界와의 相互 關聯性을 解析하고저 下水流入海域과 淸淨海域을 中心으로 調査하여 다음과 같은 結果를 얻었다. 駕莫灣內 海水溫度는 8月에 29.5℃ 가장 높았고 冬季에는 10℃이하였다. 病原 Vibrio의 水系에 따른 生菌數는 淸淨海域(A)이 5月에 出現하여 9月에 3.2×10²/ml로 나타났고 汚染海域(C,D)에서는 4月에 出現하여 7~9月에 1.2×10²~1.7×10³/ml였고 病原腸內細菌은 淸淨海域에서 6月, 汚染海域에서 3月에 出現하였으며 各各 9月에 1.3×10²/ml, 6.2×10²/ml의 生菌數가 檢出되었다. 分離菌株에 對한 同定結果는 病原 Vibrio 屬 139 菌株中 V.parahaemolyticus 가 48菌株로 62.3%, V. fluvialis 가 24菌株로 17.3% 同定되어 이들 두 菌株가 病原 Vibrio 屬中 優点種이었고 病原腸內細菌은 分離菌 122菌株中 Salmonella 屬이 43菌株로 35.2% 檢出되었고 이 가운데서 Sal. typhi 가 24菌株로 55.8%, Sal. paratyphi-C가 10菌株로 23.3%를 차지하였다. Kamak bay is the very important sea area in which have been producing not only fish but shellfish. But recently this bay is showing the tendency contaminated by domestic sewage and industrial waste-water. Therefore in order to investigate for bacteria in the contaminated environment to give what kind of influence on an ecosystem of microorganism, the distribution and bacteriological characteristics of the pathogenic Vibrio and Enterobacteriaceae were investigated as follows : The temperature of sea water in Kamak bay was the highest temperature when it was 29.5℃ in August and it was less than 10℃ during winter season. The viable cell counts of pathogenic Vibrio in the clean and the contaminated sea area appears for the first time in May and April, respectively. The former was 3.2×10² per ml in September and the latter was 1.2×10² 1.7×10³ per ml during July and September. Those of pathogenic Enterobacteriaceae appears in June in the clean sea area and in March in contaminated sea area. Each of them in both areas were 1.3×10²and 6.2×10²per ml in September. Showing the indentification result of 139 strains isolated into genus Vibrio from each samples, V.parahaemolyticus and V.fluvialis were 62.3% and 17.3%, respectively. Among 122 stains isolated into Enterobacteriaceae by MacConky agar, genus Salmonella was 35.2% and among this genus Sal.typi(55.8%) and Sal.paratyphi-C(23.3%) were the predominant species.

        여수연안 및 동중국해의 세균상: 일반 세균, Vibro spp., 대장균군 균수에 따른 수평 분포

        1992년 8월 6일부터 8월 14일까지에 걸쳐 조사한 동중국해 및 여수연안의 세균 flora, Vibrio spp., 대장 균군에 대한 양적인 수평분포를 조사하여 미생물 생태계 해석을 위한 자료를 얻고자 실험한 결과를 요약하면 다음과 같다. 1. 동중국해의 연분농도는 24.64~33.78‰였고, 외양으로 갈수록 낮아졌다. 2. 수온은 22.53~29.18℃였고, 연안에 비해 외양으로 갈수록 높게 나타났다. 3. 여수 연안 해수중의 일반세균수는 1.0×10(2)~3.0×10(4)/ml, Vibrio spp.는 0.2×10~9.0×10(3)/ml, 대장균군은 0.3×10~3.0×10(3)ml로서 연안에서 멀어질수록 균량은 감소하였다. 4. 동중국해 해수중의 일반세균수는 0.4×10~2.0×10(3)/ml, Vibrio spp.는 0.8×10~3.0×10ml, 대장균군은 0.9×10~1.3×10ml였고, 외양으로 갈수록 일반세균은 적게 나타났으며, Vibrio spp.와 대장균군은 나타나지 않았다. 5. 동중국해에서 Vibrio spp.와 대장균군의 출현한 곳은 중국연안에 가까운 해역이였다. In order to understand microbial ecosystem in the East china sea and Yosu coastal sea horizontal distributions of bacterial flora, Vibrio spp., coliform group, temperature, and salinity in this sea area, were studied 42 sampling stations during 6th-14th August, 1992. From the results, salinity and temperature were 24.64~33.78‰ and 22.53~29.18℃, respectively. As the open sea far from land goes on, salinity became low while temperature became high. Viable cell counts of bacteral flora, Vibrio spp., and coliform group in Yosu coastal sea water were 1.0×10(2)~3.0×10(4)/ml, 0.2×10~9.0×10(3)/ml, and 0.3×10~3.0×10(3)/ml while those of these in the open sea water were 0.4×10~2.0×10(3)/ml, 0.8×10~3.0×10/ml, and 0.9×10~1.3×10/ml, respectively.
