Cimetidine , Ranitidine 이 Theophylline 의 약리역동학에 미치는 영향

N/A Maintenance of Theophylline concentration 10-20 ㎍/ml is important due to narrow therapeutic index of theophylline. To evaluate the effect of Cimetidine and Ranitidine on Theophylline pharmacokinetics, we measured the theophylline concentration at 1, 1.5, 2, 3, 6, 12hr after oral administration of theophylline 200㎎ six hourly for 3 days (Control group, 7 nonsmokers), oral administration of cimetidine 200㎎ six hourly for 7 days with theophylline 200㎎ six-hourly for last 3 days (Cimetidine group, 7 nonsmokers) and oral administration of ranitidine 150㎎ twelve-hourly for 7 days with theophylline 200㎎ six hourly for last 3 days (Ranitidine group, 7 nonsmokers) using a homogenous-enzyme immunoassay. The results were as follows; 1. Total Body Clearance of Theophylline was reduced average 10% (range 0-25%) in Cimetidine treatment group. (Control group; 29.05±1.52 ml/hr/㎏, Cimetidine group; 26.10±1.60 ml/hr/㎏, p<0.01). And reduced average 8% (range 0-19%) in Ranitidine treatment group (Control group; 29.05±1.52 ml/hr/㎏, Ranitidine group; 26.89±2.57 ml/hr/㎏, p<0.01). 2. Half life of Theophylline was increased average 16% (range 0-43%) in Cimetidine treatment group (Control group; 8.8±1.3 hr, Cimetidine group; 10.4±1.9 hr, p<0.05). And increased average 26% (range 0-53%) in Ranitidine treatment group (Control group; 8.8±1.3 hr, Ranitidine group; 11.7±2.8 hr, p<0.05). 3. There was no significant change in the Volume of Distribution.
