        사이프러스 에센셜 오일의 흡입이 전임상 실험동물의 손상된 학습능력과 기억력에 미치는 영향

        Objectives : Increasing evidence supports the biological and pharmacological activities of essential oils on the central nervous system such as pain, anxiety, attention, arousal, relaxation, sedation and learning and memory. The purpose of present work is to investigate the protective effect and molecular mechanism of cypress essential oil (CEO) against scopolamine (SCO)-induced cognitive impairments in C57BL/6 mice. Methods : A series of behavior tests such as Morris water maze, passive avoidance, and fear conditioning tests were conducted to monitor learning and memory functions. Immunoblotting and RT-PCR were also performed in the hippocampal tissue to determine the underlying mechanism of CEO. Results : SCO induced cognitive impairments as assessed by decreased step-through latency in passive avoidance test, relatively low freezing time in fear conditioning test, and increased time spent to find the hidden platform in Morris water maze test. Conversely, CEO inhalation significantly reversed the SCO-induced cognitive impairments in C57BL/6 mice comparable to control levels. To elucidate the molecular mechanisms of memory enhancing effect of CEO we have examined the expression of brain-derived neurotrophic factor (BDNF) in the hippocampus. CEO effectively elevated the protein as well as mRNA expression of BDNF via activation of cAMP response element binding protein (CREB). Conclusions : Our findings suggest that CEO inhalation effectively restored the SCO-impaired cognitive functions in C56BL/6 mice. This learning and memory enhancing effect of CEO was partly mediated by up-regulation of BDNF via activation of CREB.

        Succinic Anhydride로부터 N-Hydroxysuccinimide의 친환경 생산공정 개발

        제약화학 및 정밀화학 산업에서 널리 사용되는 N-히드록시숙신이마이드(N-Hydroxysuccinimide, NHS)의 새로운 친환경적인 합성법을 개발하였다. 기존 합성법은 숙신산에 히드록시아민 반응 후 강산을 촉매로 약 70%의 수율로 NHS를 얻는다. 이 방법은 저가 숙신산을 사용하여 NHS를 얻을 수 있으나NHS를 정제하는 데에 많은 용제가 필요하고 후처리 과정이 복잡하여 수율이 낮은 문제점이 있다. 그리고 대량생산하기에는 고온반응에 따른 안전상의 위험성이 있으며 산성 촉매를 사용함에 따 른 많은 폐기물발생과 다양한 용제를 사용함에 따라 고비용의 생산비로 인해 경제적이지 않다. 이런 단점을 보완하기 위해서 반응성이 우수한 무수 숙신산을 사용하였으며 용제의 단일화 그리고 결정화 방법을 통해 고순도 및 고수율의 NHS를 제조하는 경제적 인 방법을 개발하였다. 특히 촉매를 사용하지 않는 무촉매 반응과 저온의 반응조건을 확보함으로써 80% 이상의 높은 수율로NHS를 제조하는 새로운 친환경적인 공업적인 합성법이다. 향후에는 이 결과를 바탕으로 스케일 업 연구를 통해 상용화 생산기술을 확립하여 국외 기술이전을 추진할 예정이다. A new eco-friendly synthetic method for N-hydroxysuccinimide (NHS), widely used in the pharmaceutical and fine chemical industries, is developed. Conventional synthesis method yields NHS of about 70% after its reaction with NH₂OH to succinic acid. In this method, NHS can be obtained using low-cost succinic acid, but a great deal of solvents are required as an extraction method to purify NHS, while the work-up process is complicated, resulting in low yield. In addition, there is a safety risk due to the high reaction temperature for commercial production, and it is not economical due to the high cost of production from the generation of much waste because of an acid catalyst and the use of various solvents. In order to make up for this shortcoming, this study used succinic anhydride as a raw material under low temperature reaction and developed a new eco-friendly industrial synthesis method using isobutyl alcohol for a single solvent and non-catalytic reaction. The economic evaluation confirms that there is a cost reduction effect of about 20%. In the future, based on this result, studies may establish a commercial production technology through scale-up research and proceed with foreign technology transfer.

        Autosomal-dominant polycystic kidney disease (ADPKD) is a progressive genetic disease in which multiple small and large fluid-filled sacs (cysts or cysts) are formed in one or both kidneys. Polycystic kidney disease in cats is classified as an autosomal dominant genetic disease and has been found to be caused by mutations in the PKD1 gene that is coding a unit of mechanosensitive calcium channel, polycystin 1. It shows that humans and cats share a genetic cause for polycystic kidney disease. In this paper, we try to understand the pathogenesis of polycystic kidney disease through the genetic mutations and protein functions of feline polycystic kidney syndrome reported so far, and discuss the direction of genetic research that needs to be performed in the future.

        섬백리향 잎과 줄기 추출물의 항염 활성에 관한 세포생물학적 연구

        Objectives : Thymus quinquecistatus var. japonica (H.Hara) is a member of the genus Thymus of perennial aromatic herb, and it’s designated as a natural monument of South Korea. It has traditionally been known to have protective or therapeutic effects on various human disease including cerebrovascular and neurological disease. Recently it was suggested that essential oil extracted from thyme has anti-fungal and anti-bacterial effect. The aim of this study is to investigate anti-inflammatory effect of Thymus quinquecistatus var. japonica in Raw 264.7 macrophage cell line. Methods : The cytotoxic effects of water and 70% ethanol extracts from Thymus quinquecistatus var. japonica , was tested using MTT assay. Inhibitory effects of the extracts to nitric oxide production and mRNA expression of inflammatory cytokines were examined by RT-PCR. Also, MitoSOX-red assay and JC-1 assay were performed to determine if the extracts can inhibit mitochondrial ROS accumulation and maintain mitochondrial membrane potential. Results : In LPS-induced inflammatory response, the extracts efficiently reduced nitric oxide NO production through inhibiting mRNA expression of iNOS enzyme. In addition, expression of the proinflammatory cytokines, IL-1β and IL-6, was also down-regulated by the extract treatments. Excessive accumulation of mitochondrial ROS induced by LPS was inhibited in the extract treated cells, which finally protected mitochondrial membrane potential. Conclusions : These results showed that water and 70% ethanol extracts from Thymus quinquecistatus var. japonica have anti-inflammatory effect through down regulation of IL-1β, IL-6, and iNOS, and also have antioxidative effect against mitochondrial ROS accumulation that promote inflammatory response.

        치의학 교육의 학습윤리에 관한 연구

        The topic of academic integrity is an important public concern that has emerged in higher education. Recent surveys at U.S. and Canadian dental schools revealed that cheating and plagiarism were significant problems in dental schools. In addition, some schools stated that cheating had increased compared to a decade ago. Various institutional rituals have been implemented to enhance the academic integrity environment of U. S. and Canadian dental schools. Furthermore, the application of honor code which is dealing with ethical issues has been reported to improve the attitudes and behaviors of students. Since there have been no reported studies regarding ethics curricula in Korean dental schools, further studies should be needed to assess academic integrity policies, violations, and the results of the measures in Korean dental schools. Additionally, the challenge to provide professional ethics curricula for dental students must be conducted with respect and humanity for our students and thus, students will be more likely to respond positively to expectations in terms of ethical behaviors. Therefore, the outcome is clearly and undoubtedly link to better care for patients.
