Carboxymethylcellulose, which is water soluble at room temperature, was manufactured from the cellulose material in this study. Experimental parameter included reaction temperature, time, and concentration of NaOH and trichloroacetic acid. CMC was tested for solubility, degree of substitution, and tensile strength. CMC manufactured from viscose rayon was affected by the chemical concentration rather than the reaction time and temperature. Also, DS is closely related to the solubility of the CMC.

Copolymer consists of polyethylene oxide (PEO) and maleic acid anhydride (MA) was used to modify the lipase. By controlling the ratio of ethylene oxide to alkylene oxide (AO), hydrophilic property of copolymer was controlled and the MA functional group of copolymer covalently coupled with the amino groups of lipase. Activity of modified lipase increase more than native one by about 20~30% due to substrate effect of synthesized copolymers. Native lipase was absolutely specific that only reacts limited substrate. However, as the modification was progressed, lipase was changed to group specificity that widely reacts several substrates having similar characteristic.

Lipase was modified with hydrophilic and hydrophobic copolymers and, was reacted to the several substrates. In general, modified lipase was better than native one as it resolved the substrate. As lipase was modified with strongly hydrophobic copolymers, degree of modification decreased and, relative activity increased.

Reactive aqueous two phase system(ATPS) using hydrophilic polymer and dextran was applied to recover the Copolymer consists of polyethlene oxide(PEO) and maleic acid anhdride (MA) was used to modify the cellulase. BY controlling the ratio of ethylene oxide, hydrophilic property of copolymer was controlled, and the MA functional group of copolymer covalently coupled with the amino groups of cellulase. The two phase system was mixed with dextran and copolymers with molecular weight of 505,000. The modified moved to the down phase,dextran As a result, most enzymes, approximately over 70%, moved to copolymer Therefore, reactive aqueous two phase partition by modification of cellulase is very effective to separate enzyme from the fermentation broth.
