본 연구는 세포자멸사에서 중요한 역할을 하는 p53 단백질 발현에 대한 웅담(bear bile)의 영향을 알아보기 위하여 Western blot 분석과 RT-PCR 분석 방법을 이용하여 p53 유전자 특성이 서로 다른 암세포주를 대상으로 p53 단백질 발현의 변화를 조사하였다. 그 결과 p53 유전자에 돌연변이가 있는 HaCaT, KUMA3, KUMA4 및 KUMA6 세포주에서는 웅담처리에 의해 p53 단백질 발현이 증가하였으나, 돌연변이가 없는 HCT116과 KUMA5 세포주에서는 p53 단백질 발현의 변화가 없었다. 또한, 모든 세포주에서 웅담처리에도 불구하고 p53 mRNA의 양적 변화는 관찰할 수 없었다. 이러한 결과들은 웅담처리에 의한 p53 단백질의 증가가 단백질 합성의 증가가 아니라 단백질 안정성의 증가를 유도하였음을 추측할 수 있다. 결론적으로 웅담에 대한 종양세포의 감수성은 p53 유전자의 특성에 따라 다르다는 것을 알 수 있었다. 즉, 웅담처리는 p53 유전자에 돌연변이가 있는 세포주의 변이형 p53 단백질에만 영향을 주었다. The effects of bear bile on the expression of the p53 protein in cancer cell lines were investigated by Western blot and RT-PCR methods. The p53 protein expression was increased after addition of bear bile in HaCaT, KUMA3, KUMA4 and KUMA6 cell lines that carried mutations in the p53 gene, but it is not increased in HCT116 and KUMA5 cell lines with the normal p53 gene as determined by Western blot method. The level of p53 mRNA was not altered in all cell lines after the treatment of bear bile as determined RT-PCR. These results indicate that the increment of the p53 protein after addition of bear bile was caused by increase in the protein stability but not by increase in protein synthesis. It can be concluded that bear bile affects positively only in the cell lines which have genetic variation of the p53 gene, and does not affect in the cell lines which have wild-type p53 gene, on the other hand.
