N/A Enhanced HLA-DR expression is a characteristic feature of inflamed rheumatoid synovium. Among the cytokines demonstrated within synovial fluid and tissue, gamma-interferon (IFN-γ) is known to be a potent inducer of HLA-DR antigens in vitro. Gold compounds have been used in rheumatoid arthritis treatment for mamy decades, but their mechanism of action, especially on immune response, remains unk- own. Thus we investigated the potential of gold sodium thiomalate (GST) to interact with INF-γ on rheumatoid synovial cells in culture. The results were as follows: 1) IFN-γ increased the proliferation and HLA-DR expression of RA synovial cells in a dose-dependent manner. 2) GST inhibited synovial cell proliferation. How-ever, GST alone in various concentrations was found to stimulate DR expression by synoveal cells. 3) Simultaneous GST treatment of IFN-γ stimulated synovial cells inhibited the increase of HLA-DR at the concentration of 0.1~10 mcg/ml. 4) A higher concentration of GST (25mcg/ml) in- duced DR antigen synergistically. Our data suggest that GST has an immunostimulatory effect on synovial cells which is related to enhanced HLA-DR antigen expression.
