납 및 구리화합물이 Carassius auratus(goldfish)에 미치는 독성에 관한 연구

In order to investigate acute toxicity and bioconcentration of heavy metals for a freshwater fish, the fish used in this experiment was goldfish, Carassius auratus. Each ten goldfish was accommonidated in a water and was treated with different concentration of Pb and Cu compound. The 24 $hr-LC_{50}$ was obtained by plotting on the log-normal distribution graph. Furthermore, the combined effect of Pb and Cu was also investigated the fish was treated with Pb or Cu compound only, and Pb and Cu compound together, respectively. These results were summarized as follows: 1. The 24 $hr-LC_{50} s$ of Pb and Cu were 7.48 mg/l and 0.666 mg/l, respectively. 2. When single or/and combined treatment with Pb(7.0 mg/l) or/and Cu(0.6 mg/l) to Carassius auratus for 24 hours were performed, there was significant difference between the single or/and the combined treatment in their bioaccumulated Cu concentrations. Cu concentrations in goldfish were higher in the combined treatment than in the single treatment. 3. When Carassius auratus was exposed to 0.748 mg/l (1/10 of 24 $hr-LC_{50}$) and 1.496 mg/l of Pb (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 79.14 and 100.11 for Pb, respectively. The BCF of Pb was obtained as a linearity according to the concentration and exposure time as follows log BCF=1.014 log $P\cdot T$+1.011 ($r^2$=0.9041) where, P: pollutant concentration(mg/l) T: exposure time(day) 4. When Carassius auratus was pxposed to 0.0666 mg/l (1/10 of 24 $hr-LC_{50}$) and 0.1332 mg/l of Cu (1/5 of 24 $hr-LC_{50}$) for 7 days, the bioconcentration factors (BCF) were 55.42 and 63.24 for Cu respectively. This study evaluated the protective effect of Chungkookjang (CKJ) extract, a Korean traditional fermented soybean product made from Bacillus species in rice straw and boiled soybean, and one of its main flavonoids, genistein, against Trp-P-1 induced cytotoxicity and DNA damage in HepG2 cells. CKJ and genistein exhibited protective effect against Trp-P-1 induced cytotoxicity and Trp-P-1 induced DNA single strand breaks. CKJ and genistein inhibited Trp-P-1 induced CYP1A1 and CYP1A2 transcription in HepG2 cells. Our results indicated that CKJ and genistein have the protective effect against Trp-P-1 induced cytotoxicity and DNA damage. Via inhibiting expression of CYP1A1 and CYP1A2. CKJ can be used as a promising functional food material that prevents the genotoxicity induced by carcinogens produced by the heat treatment of foods such as heterocyclic amines (HCAs) that cause genomic instability.
여성 생식기에서 추출된 과량의 여성 DNA가 남성의 Y-STR DNA형 확보와 남성 DNA 정량값에 미치는 영향

In sexual crime cases, it is difficult to obtain a male short tandem repeat (STR) profile from samples with high concentration of female DNA. In the cases, it is important to obtain Y chromosome STR(Y-STR) profile from the samples in order to solve the cases. Occasionally, intact Y-STR profile is found in samples containing the male DNA less than the minimum detection limit in sexual crime cases. At this point, we wonder whether the quantitative value of male DNA was affected by the high concentration of female DNA. Using two Y-STR kits, Y-STR profiles were examined in the mixed DNA samples of female DNA isolated from vaginal fluid and male standard DNA. It was confirmed that high concentrations of female DNA did not affect the acquisition of Y-STR profile in the mixed DNA samples. However, the female DNA affected the quantitative values of male DNA in the samples. This study is expected to contribute to the reliability of obtaining intact Y-STR profile in mixed DNA samples with concentrations below the minimum detection limit for obtaining the Y-STR profile.
