유기폐수처리를 위한 Rhodospirillum rubrum P17 의 종균생산

A photosynthetic bacterium strain P17 having high growth rate and assimilating ability of organic acids was isolated from several soil samples, which was identified as Rhodospirillum rubrum. Cultural conditions of the strain P17 were examined for the production of starter culture used in the treatment of organic waste water. The addition of organic acids mixture as carbon source containing 0.2% Na-acetate, 0.1% Na-propionate and 0.2%r Na-lactate and 0.1% of yeast extract as growth factor stimulated the cell growth. The maximal cell production was obtained at 30℃, pH 7.0, 2,500 lux of illumination and 50∼100 rpm of agitation. Under the optimal conditions of batch and fed-batch culture systems in a Jar fermentor, 5.17 g/ℓ and 7.93 g/ℓ of cells were obtained after 5 days of cultivation, respectively. In continuous culture system, the cell productivity was 0.206 g/ℓ/h at a dilution rate of 0.21 h^(-1). When R. rubrum P17 was cultivated in a soybean curd waste water, initial COD level(3,240 ㎎/ℓ) of the waste water was reduced to 250 ㎎/ℓ after 4 days of cultivation.
두부공업폐수를 이용한 광합성세균 Rhodospirillum rubrum P17의 균체생산

광합성 세균 R. rubrum P17의 균체생산을 위한 두부공업폐수의 이용가능성과 폐수처리 효과를 동시에 검토하였다. 원폐수에서의 최적 생육 환경인자들은 pH 7.0, 온도 30℃, 조도 2,500 lux, 50 rpm의 교반 등이었다. 두부공업폐수의 초기 COD는 3,240 ㎎/ℓ로 나타났으며 회분배양 4일에 폐수의 COD는 150㎎/ℓ로 감소하여 95.4%의 COD 제거율을 보였다. R. rubrum P17의 균체생산성은 유가배양에서 폐수의 연속첨가에 의해 크게 증가하여 5.45 g/ℓ의 최대 생산량을 보였다. 건조균체의 조단백질 함량은 약 70% 였고, 필수아미노산의 함량 및 조성이 클로렐라, 효모균체에 비해 손색이 없었다. Rhodospirillum rubrum P17 was used to investigate the pontential for the treatment of soybean curd waste water and for the utilization of the biomass produced. The maximal biomass production and COD removal from the waste water were obtained at 30℃, pH 7.0 under 2,500 lux of illumination and 50 rpm of agitation. The initial COD level of the soybean curd waste water was 3,240 ㎎/ℓ, and after 4 days of cultivation in batch culture, 3.46 g/ℓ of cells was obtained and COD level of the waste water reduced to 150 ㎎/ℓ (COD removal rate 95.4%). The biomass production of R. rubrum P17 was enhanced to 5.45 g/ℓ by continuous feeding of the waste water. The cells of R. rubrum P17 grown on soybean curd waste water contained approximately 70% crude protein, and the protein consisted of appreciable amounts of essential amino acids comparable with those of chlorella and yeast protein.
