Experiments were conducted to find out the effect of several environmental and cultural factors on the growth of excised cotyledons of radish (Raphanus sativus L.). Among the 9 cultivars tested, 'Sinjinju' and 'Sachulyulmoo' were highly suitable for bioassay based mainly on the uniform seed germination, seedling vigor, and higher response to applied cytokinins. The growth of excised radish cotyledons was enhanced when the solution pH was increased from 4.0 to 6.0 but significantly suppressed in solutions of higher pH value. Incubation period of 72 hours, under the conditions of 24℃ and continuous illumination of fluorescence light at 1000 to 2000 lux, was proven to be most appropriate in terms of the duration and efficiency of the experiment. The cotyledons of dark-grown seedlings responded better to the applied cytokinins compared to the light-grown ones. Red fluorescence lighting was better than the white fluorescent or incandescent lightings when compared at the same intensity. Among the cytokinins compared, zeatin showed the best response followed closely by BA. Kinetin and 2-ip were less effective for increasing cotyledon growth as compared to zeatin and BA. Adenine sulfate suppressed the growth, though not effective at lower concentrations. Brassinolide significantly promoted the cotyledon expansion at low concentrations. At higher concentration (1 ppm), however, apparent toxicity symptoms, such as degreening and tissue softening, were noticed. The growth promoting effect of BA could be effectively countered by high concentrations of 2,4-D. The enhancing effect of BA was further maximized by the addition of brassinolide, thus showing the maximum growth promoting response at BA 1 ppm and brassinolide 10^(-3) ppm.
