        사염화탄소에 의해 유발된 생쥐의 간독성에 미치는 누에 추출물의 영향

        To evaluate the protective effects of extracts of silkworm on carbon tetrachloride-induced hepatotoxicity, serum glutamic-oxaloacetic transaminase, glutamic-pyruvic transaminase and lactic dehydrogenase activities, malondialdehydes values and glutathione-S-transferase activity were measured in ICR mice. Extracts of silkworm was administered orally at 30min after the administration of CCl4 Mice were sacrificed at 24h after the administration of extracts of silkworm. The activities of serum aminotransferase and the hepatic content of lipid peroxide after carbon tetrachloride treatment were markedly increased than normal control but those levels were decreased by the treatment of butanol soluble fraction of silkworm methanol extract. Glutathione S-transferase activity was decreased by carbontetrachloride than control, but also inhibited by the treatment of butanol soluble fraction of silkworm methanol extract.

        혐기처리한 뽕잎의 혈당강하 효과

        The hypoglycemic effect of mulberry leaves using alloxan-induced diabetic mice was investigated. The hypoglycemic effect of mulberry leaves was significantly affected with anaerobic incubation up to for 6 hrs. The body weight of mice treated with 20% anaerobic conditioned mulberry leaves was higher than that in the control group. Mulberry leaves which were treated with anaerobic condition significantly decreased blood glucose level in alloxan-induced type I diabetes mice. Furthermore, glucose tolerance was significantly recovered by feeding mulberry leaves anaerobically treated. Pathological analysis revealed that the disappearance of insulin-secreting beta-cells in the islets of Langerhans from the alloxan induced diabetic mice was strongly inhibited in the mulberry leaves feeding group. The content of 1-deoxynojirimycin increased up to 5% in detached mulberry leaves incubated under nitrogen gas for 6 hrs. Nevertheless, it is not clear whether the synthesis of 1-deoxynojirimycin is increased or the degradation of it is decreased. Overall, results obtained from this study indicate that the hypoglycemic effect was increased by the mulberry leaves with anaerobic treatment.

        배추흰나비 과립병바이러스 단백질의 생화학적 특성

        배추흰나비 P. rapae와 P. brassicae GV에 대한 봉입체 및 바이러스단백질의 물리화학적 성상을 비교 분석한 결과 전기영동법에 의한 봉입체단백질은 그 분자량이 P. rapae GV는 30kd, P. brassicae GV는 31 kb의 단일단백질이었으며, trypsin, chymotrypsin, papain 및 staphylococcus aureus V8 protease에 의한 peptide mapping에서는 두 바이러스 간에 큰 차이가 없었다. 바이러스입자단백지르이 전기영동법에 의한 분석결과, 두 바이러스 간에 큰 차이가 없었다. 바이러스입자단백질의 전기영동법에 의한 분석결과, 두 바이러스에서 각각 42개의 band가 나타났으며 고분자량 부분에서 P. rapae GV의 경우는 115, 110, 105 및 103 kd의 4개 band인데 반해 P. brassicae GV는 107 kd의 한 band만 관찰되었다. 그리고 봉입체단백질의 아미노산 분석비교에서 두 바이러스 간의 큰 차이는 없었으며 일반적으로 곤충에 많이 존재하는 산성아미노산인 aspartic acid와 glutamic acid의 함량이 많았고 염기성 아미노산에서 lysine의 함량이 특히 많았으며 봉입체단백질의 면역학적인 시험결과에서는 P. rapae GV 항혈청에 대해 두 바이러스는 공통 침강선을 형성하였다. This study was carried out to acquire some basic biochemical informations on the granulosis virus(GV) of Pieris rapae and Pieris brassicae. The capsule protein was composed of a single polypeptide with a molecular weight of 30,000 dalton for P. rapae GV and 31,000 dalton for P. brassicae GV. The major amino acids of capsule protein were glutamic acid, aspartic acid and lysine. When the capsule protein was partially digested with trypsin, chymotrypsin, papain or Staphylococcus aureus V8 protease, the digested products of the two viruses showed no difference in electrophoretic mobility. The patterns of the polypeptides of the two virus particle on SOS-polyacrylamide gel showed a little difference in high molecular weight region(over MW 100 kd).

        배추흰나비 과립병바이러스 DNA의 생화학적 특성

        배추흰나비 P. rapae와 P. brassicae GV의 DNA성상과 이를 기초로한 상동성을 검정한 결과 Tm값에서는 P. rapae GV DNA는 $83.7^{\circ}C$이고, P. brassicae GV DNA의 경우는 $84.0^{\circ}C$로 G+C 함량은 전자가 35.5%, 후자가 35.9%였다. 그리고 제한산소처리에 의한 DNA의 분석에서는 P. Rapae GV의 genome의 크기는 103 kb, P. brassicae GV DNA는 108kb로 나타났으며, 두바이러스 간의 상동성은 97.0%였다. This study was carried out to acquire some basic biochemical informations on the granulosis virus (GV) DNA of Pieris rapae and Pieris brassicae. The thermal denaturation temperature (Tm) and G+C content of the DNA of the viruses were $83.7^{\circ}C$ and 35.5% for P. rapae GV, $84.0^{\circ}C$ and 35.9% for P. brassicae GV, respectively. There were some differences in the DNA fragmentation patterns of the two GV's produced by digestion with restriction endonucleases such as EcoR I , BamH I and Hind m . The homololgy between the two DNAs was caculated to be 97.0%. The size of the genome was estimated to be 103 kbp for P. rapae GV and 108 kbp for P. brassicae GV.

      • 누에분말 제조 조건에 따른 혈당강하효과

        Bombyx mori, silkworm powder has been studied for blood-glucose lowering activity by National Sericulture & Entomology Research Institute(NSERI) and Kyung Hee University group in 1995. To obtain the optimum preparative condition of silkworm powder for lowering blood-glucose levels in postprandial time, blood-glucose lowering activity of mulberry & silkworm varieties were examined. In mulberry varieties, Yongcheonppong showed the highest activity for lowering blood-glucose levels. In case of silkworm varieties, Samkwangjam showed the highest activity. Activity for lowering blood-glucose levels of autumn rearing silkworm was higher than that of spring silkworm and hybrid silkworm showed higher activity than that of parent silkworm. In the silkworm lines, Chinese lines showed higher activity than that of Japanese lines. In experiments to see the difference in blood-glucose lowering activity between either male and female or larvae and pupae, activity of larvae was higher than that of pupae and the male was higher than female. The heating dry and artificial diet showed lower than mulberry diet and normal freeze dry of the 5th instar 3rd day. Among the sericultural products, larvae showed the highest activity.

        1-Deoxynojirimycin Content and Blood Glucose-Lowering Effect of Silkworm (Bombyx mori) Extract Powder

        We investigated the 1-deoxynojirimycin (1-DNJ) content of extracts from silkworm larvae ateach developmental stage within three silkworm varieties. We also compared the contentof the following polyhydroxylated alkaloids in the silkworm extracts: 1-DNJ, fagomine, and1,4-dideoxy-1,4-imino-d-arabinitol (DAB). In addition, we evaluated the glucose-loweringeffects of silkworm extract powder in db/db mice. The 1-DNJ content was the highest inYeonnokjam 5th instar 3rd d larvae and Hansaengjam 5th instar 3rd d larvae, which contained18.4 mg/100 g dry weight and 18.3 mg/100 g dry weight, respectively. The 5th instar 3rdd larvae exhibited a higher content of 1-DNJ than that of 5th instar 5th d larvae among allvarieties. The glucose-lowering effects of silkworm extracts and Yeonnokjam powder weretested on db/db mice, and the blood glucose levels were found to decrease significantly inthe YR70 group. Silkworm extracts (180 mg/kg, 90 mg/kg, 45 mg/kg, and 22.5 mg/kg) andacarbose (50 mg/kg) were administered orally for 4 wk. Changes in water intake were notstatistically significant between control and silkworm extract-treated groups. Compared to thecontrol group, blood glucose levels in the silkworm extract powder-treated group decreasedin the 22.5 mg/kg/d group after being administered for 4 wk. This decrease was statisticallysignificant. Furthermore, biochemical changes in the AST(Aspartate aminotransferase),ALT(Alanine aminotransferase), TCHO(Total Cholesterol), TG(Triglyceride), LDL(Low densitylipoprotein), and HDL(High density lipoprotein) levels in blood were not observed. However,statistically significant decreases in blood GLU in the 22.5 mg/kg/d group compared to thatof the control group occurred. In addition, the epididymal fat weight of the silkworm extractpowder-treated group decreased significantly in both the 22.5 mg/kg/d group and 180 mg/kg/dgroup compared to that of the control group, but there were no statistically significant changesin perirenal fat weight. These results demonstrate that silkworm extracts inhibit changes inblood glucose levels in model diabetic mice.
