      • Clinical Application of Transcatheter Arterial Chemoembolization Combined with Synchronous C-arm Cone-Beam CT Guided Radiofrequency Ablation in treatment of Large Hepatocellular Carcinoma

        Objective: This work aimed to evaluate the safety and clinical efficacy of transcatheter arterial chemoembolization (TACE) combined with c-arm cone-beam CT guided synchronous radiofrequency ablation (RFA) in treatment of large hepatocellular carcinoma (HCC). Methods: 21 patients with large HCC were studied from January 2010 to March 2012. TACE combined with synchronous C-arm cone-beam CT guided RFA were performed on a total of 25 lesions. Conventional imaging examination (CEUS, enhanced CT or MRI) and AFP detection were regularly conducted to evaluate the technical success rate of combined treatment, complications, treatment response, time without disease recurrence and survival rate. Results: The technical success rate of combined treatment was 100%, without any significant complication. After 1 month, there were 19 cases with complete response and 2 cases with partial response, with an complete response rate of 90.4% (19/21) and a clinical effective rate of 100% (21/21). The complete response rates of single nodular lesions (100%, 17/17) was significantly higher than that of multiple nodular lesions (50%, 2/4) (P<0. 05). During 2 to 28 months of follow-up, in 19 cases with complete response, the average time without disease recurrence was $10.8{\pm}6$ months. The total survival rates of 6, 12 and 18 months in 21 patients were 100%, respectively. Conclusion: TACE combined with synchronous C-arm CT guided RFA is safe and effective for treatment of large HCC. The treatment efficacy for single nodular lesion is better than that for multiple nodular lesions.

        Rice is the major food for half of the world population. The nutrition component in rice is critical for improvement of people’s health. Vitamin E serves as important antioxidant by quenching the free radical intermediates and thus protects the cell membrane. Because of the high nutritional value and the benefits of vitamin E in human health, increasing the tocochromanol content of major agricultural crops has long been in the focus of breeding programs and genetic engineering approaches. The key genes involved in tocopherol biosynthesis have been elucidated in Arabidopsis and other model organisms. Quantitative trait locus (QTL) study performed in Arabidopsis suggested that some of these key genes and a few additional loci contribute to natural tocopherol variations. Identifying such genetic variations in rice, enrich our understanding of the genetic mechanisms controlling tocopherol variation, which can be directly applied to rice breeding programs.       • Evaluation of Genetic Diversity in Rice Collections Using SSR Markers

        Rice(Oryza sativa L.) feeds more than 50% of the world’s population and is one of the most important crops in the world. To evaluate the variation between different rice classfications, genetic diversity amoung a diverse set of rice collection including 59 breedlines, 23 landraces, 18 weedy rices and 35 introductions were analysed using 134 SSR markers located on the 12 chromosomes. In total, 1269 alleles were identified with an average of 9.47 per locus. Of the 1269 alleles, 460 (36.2%) were common, with a frequency of 0.05–0.5; 741 (58.4%) were rare (frequency < 0.05) and 68 (5.4%) were abundant (frequency > 0.5). A relatively high Polymophism information content (PIC) value was detected in landraces with smaller number of accessions than that of breedlines. Model-based structure analysis revealed the presence of six subpopulations, which was essentially consistent with the clustering based on genetic distance. One hundred and eight accessions (80.0%) showed a clear relation to each cluster based on their inferred ancestry value (>70%), while the remaining 27 accessions (15.4%) of which nine from landraces and fifteen from introductions were categorized as admixtures. Landrace and introductions distributed to almost all the six subpopulations whereas most of breedlines distributed to two distinct subpopulations. In conculusion, landraces in the present study showed critical importance in preservation of genetic diversity and rice breeding programs.

      • A chloroplast variation map generated using whole genome re-sequencing of Korean landrace rice reveals phylogenetic relationships among Oryza sativa subspecies

        Although the overall structure of the chloroplast genome is generally conserved, a number of sequence variations have been identified, which are valuable for plant population and evolutionary studies. Here, we constructed a chloroplast variation map of 30 landrace rice strains of Korean origin, using the Oryza rufipogon chloroplast genome (Genbank: NC_017835) as a reference. Differential distribution of single nucleotide polymorphisms (SNPs) and indels across the rice chloroplast genome is suggestive of a region-specific variation. Population structure clustering revealed the existence of two clear subgroups (indica and japonica) and an admixture group (aus). Phylogenetic analysis of the 30 landrace rice strains and six rice chloroplast references suggested and supported independent evolution of O. sativa indica and japonica. Interestingly, two “aus” type accessions, which were thought to be indica type, shared a closer relationship with the japonica type. One hypothesis is that “Korean aus” was intentionally introduced and may have obtained japonica chloroplasts during cultivation. We also calculated the nucleotide diversity of 30 accessions and compared to six rice chloroplast references, which shown a higher diversity in the indica and aus groups than in the japonica group in lower level substitution diversity.

      • Galectin-9 Acts as a Prognostic Factor with Antimetastatic Potential in Hepatocellular Carcinoma

        Considerable research has been conducted concerning galectin-9 and carcinomas, but little information is available about any relation with the hepatocellular carcinoma. In this study, we employed a small interfering RNA (siRNA) targeting galectin-9 to down-regulate the expression in HepG2 cells. As a result, after galectin-9 expression was reduced, cell aggregation was suppressed, while other behaviour such as the proliferation, adhesion and invasion to ECM, cell-endothelial adhesion and transendothelial invasion of the cells were markedly enhanced. When tumors of 200 patients with hepatocellular carcinoma were tested for galectin-9 expression by immunohistochemistry, binding levels demonstrated intimate correlations with the histopathologic grade, lymph node metastasis, vascular invasion and intrahepatic metastasis (P<0.05). Moreover, survival analysis indicated that patients with galectin-9 expression had much longer survival time than those with negative lesions, and the Log-rank test indicated that this difference was statistical significant (P<0.0001). The Cox proportional hazards model suggested that negative galectin-9 expression in hepatocellular carcinoma represented a significant risk factor for patient survival. We propose that galectin-9 might be a new prognostic factor with antimetastatic potential in patients with hepatocellular carcinoma.

        Stationary Distribution for the Mobilities in Catastrophe Rescue Scenario

        Mobility Model has drawn more and more attentions since its critical role in Mobile Wireless Networks performance evaluation. This paper analyzes the mobility patterns in the catastrophe rescue scenario, and proposes the Random Waypoint with Base Point mobility model to model these characteristics. We mathematically analyze the speed and spatial stationary distributions of the nodes and derive explicit expressions for the one dimensional case. In order to keep the stationary distribution through the entire simulation procedure, we provide strategies to initialize the speed, location and destination of the nodes at the beginning of the simulation. The simulation results verify the derivations and the proposed methods in this paper. This work gives a deep understanding of the properties of the Random Waypoint with Base Point mobility model and such understanding is necessary to avoid misinterpretation of the simulation results. The conclusions are of practical value for performance analysis of mobile wireless networks, especially for the catastrophe rescue scenario.

        Optimized Transformation of Streptomyces sp. ATCC 39366 Producing Leptomycin by Electroporation

        Streptomyces sp. ATCC 39366 produces leptomycin derivatives. Leptomycin B, a potent and specific inhibitor against the export of nuclear proteins, is the main product; however, the introduction of DNA into this strain is almost impossible, which has impeded its further use. We developed a Streptomyces sp. ATCC 39366 transformation protocol to introduce foreign DNA via electroporation. Various conditions were examined, including treatments of the cell wall with weakening agents, electroporation parameters, and DNA content. We found that only plasmid DNA isolated from a dam- ET12567 strain resulted in successful transformation. The mycelium growing in a yeast-peptone-dextrose medium supplemented with 1% glycine at 28°C on a rotary shaker (220 rpm) was more dispersed than those without supplementation and prone to electroporation. The maximum transformation efficiency of 8×102 CFU/μg plasmid DNA was obtained at a field strength of 13 kV/cm with a time constant of 13 ms (25-μF capacitor; parallel resistance, 600 Ω) using 1-mm electrocuvettes. The results of the transformations of two other Streptomyces species indicated that the optimized conditions established in this study might only be applicable to Streptomyces sp. ATCC 39366. However, this is the first report of successful transformation of Streptomyces sp. ATCC 39366, and will facilitate the construction of a gene knockout mutant in Streptomyces sp. ATCC 39366 to produce series of new leptomycin derivatives.

        Molecular characterization and expression analysis of putative odorant carrier proteins in Adelphocoris lineolatus

        The alfalfa plant bug Adelphocoris lineolatus (Goeze) (Hemiptera: Miridae), a serious pest of several agricultural crops, is extremely attracted by flowering plants to perform season host switching. Generally, chemoreception of A. lineolatus plays a crucial role in detecting food sources, locating mates and oviposition sites. In addition, odorant carrier proteins are believed to participate in the initial chemical communication and perception in insects. In this work, we identified 31 putative odorant carrier protein genes from the antennal transcriptomes of A. lineolatus, including 17 new odorant binding proteins (OBPs), 10 new chemosensory proteins (CSPs) and four Niemann-Pick type C2 proteins (NPC2s). Phylogenetic analysis demonstrated that the OBP and CSP genes from four mirid bug species (A. lineolatus, Adelphocoris suturalis, Apolygus lucorum and Lygus lineolaris) had a relatively close evolutionary relationship. Regular patterns and key conserved motifs of OBPs and CSPs in Hemiptera insects are also identified by using Multiple EM for Motif Elicitation (MEME) tool. Tissue expression profiles analysis showed that nine of the 17 OBPs, 10 CSPs and two of the four NPC2s were primarily or uniquely expressed in the adult antennae suggesting their putative roles in chemoreception. Our study provides the first insight into the roles of odorant carrier proteins in chemoreception of A. lineolatus, which will help to facilitate biological functions of odorant carrier proteins and develop novel insect behavioral regulation strategy

        Mesenchymal Stem Cell Therapy in Acute Liver Failure

        Acute liver failure (ALF) is a severe liver disease syndrome with rapid deterioration and high mortality. Liver transplantation is the most effective treatment, but the lack of donor livers and the high cost of transplantation limit its broad application. In recent years, there has been no breakthrough in the treatment of ALF, and the application of stem cells in the treatment of ALF is a crucial research field. Mesenchymal stem cells (MSCs) are widely used in disease treatment research due to their abundant sources, low immunogenicity, and no ethical restrictions. Although MSCs are effective for treating ALF, the application of MSCs to ALF needs to be further studied and optimized. In this review, we discuss the potential mechanisms of MSCs therapy for ALF, summarize some methods to enhance the efficacy of MSCs, and explore optimal approaches for MSC transplantation.

      • Fire Resistance Studies on High Strength Steel Structures

        High strength steels have been widely applied in recent years due to high strength and good working performance. When subjected to fire conditions, the strength and elastic modulus of high strength steels deteriorate significantly and hence the load bearing capacity of structures reduces at elevated temperatures. The reduction factors of mechanical properties of high strength steels are quite different from mild steels. Therefore, the fire design methods deduced from mild steel structures are not applicable to high strength steel structures. In recent ten years, the first author of this paper has carried out a lot of fundamental research on fire behavior of high strength steels and structures. Summary of these research is presented in this paper, including mechanical properties of high strength steels at elevated temperature and after fire exposure, creep response of high strength steels at elevated temperature, residual stresses of welded high strength steel member after fire exposure, fire resistance of high strength steel columns, fire resistance of high strength steel beams, local buckling of high strength steel members, and residual strength of high strength steel columns after fire exposure. The results show that the mechanical properties of high strength steel in fire condition and the corresponding fire resistance of high strength steel structures are different from those of mild steel and structures, and the fire design methods recommended in current design codes are not applicable to high strength steel structures.
