        Mitochondrial citrate accumulation drives alveolar epithelial cell necroptosis in lipopolysaccharide-induced acute lung injury

        Necroptosis is the major cause of death in alveolar epithelial cells (AECs) during acute lung injury (ALI). Here, we report a previously unrecognized mechanism for necroptosis. We found an accumulation of mitochondrial citrate (citratemt) in lipopolysaccharide (LPS)-treated AECs because of the downregulation of Idh3α and citrate carrier (CIC, also known as Slc25a1). shRNA- or inhibitor–mediated inhibition of Idh3α and Slc25a1 induced citratemt accumulation and necroptosis in vitro. Mice with AEC-specific Idh3α and Slc25a1 deficiency exhibited exacerbated lung injury and AEC necroptosis. Interestingly, the overexpression of Idh3α and Slc25a1 decreased citratemt levels and rescued AECs from necroptosis. Mechanistically, citratemt accumulation induced mitochondrial fission and excessive mitophagy in AECs. Furthermore, citratemt directly interacted with FUN14 domain-containing protein 1 (FUNDC1) and promoted the interaction of FUNDC1 with dynamin-related protein 1 (DRP1), leading to excessive mitophagy-mediated necroptosis and thereby initiating and promoting ALI. Importantly, necroptosis induced by citratemt accumulation was inhibited in FUNDC1-knockout AECs. We show that citratemt accumulation is a novel target for protection against ALI involving necroptosis.

        SNS 1인마켓에 대한 불만족 및 불평행동 연구 : 주관적 지식의 조절효과 검증

        최근 SNS 1인마켓이 빠른 속도로 성장하고 있으며, 이와 더불어 거래와 관련하여 불만족 및 피해가 증가하고 있으므로 불만족을 파악하고 이에 대한 불평행동을 분석하는 것은 의미가 있다. 따라서 본 연구는 SNS 1인마켓에서 구매 경험이 있는 20대, 30대 소비자를 대상으로 SNS 1인마켓에 대한 지각된 혜택, 지각된 위험, 부정적 기대불일치가 불만족에 미치는 영향을 분석하였다. 또한, 불만족이 불평행동에 미치는 영향과 불만족과 불평행동의 관계에서 주관적 지식의 조절효과를 분석하였다. 본 연구는 SNS 1인마켓의 불만족 감소를 위한 방안을 제시하여 소비자의 합리적 이용에 도움이 되고자 한다. 또한, 기업이나 개인 판매자들이 SNS 1인마켓의 각종 불만족과 불평행동을 파악하여 합리적인 서비스 제공과 SNS 1인마켓 운영을 위한 기초자료를 제공하고자 한다. 본 연구의 주요 결과는 첫째, SNS 1인마켓의 불만족에는 부정적 기대불일치가 가장 큰 영향을 보였으며, 다음으로 시간 위험, 환불/교환 위험, 경제적 혜택, 정보 혜택, 경제적 위험의 순이었다. 둘째, SNS 1인마켓의 불만족은 사적 불평행동과 공적 불평행동 모두에 영향을 미치고 있었다. 셋째, SNS 1인마켓에 대한 주관적 지식이 불만족과 사적불평행동 간의 관계에서 조절효과가 있음이 밝혀졌다. Recently, SNS market is growing rapidly, and in addition, dissatisfaction and damage related to transactions are increasing, so it is meaningful to grasp dissatisfaction and analyze complaints about it. Therefore, this study analyzed the impact of perceived benefits, perceived risks, and negative expectancy disconfirmation on the SNS market to identify complaints and behaviors of consumers in their 20s and 30s who have experience in using the SNS market. In addition, the effect of dissatisfaction on the complaints behavior and moderating effect of subjective knowledge on the relationship between dissatisfaction and dissatisfaction behavior were analyzed. This study aims to help consumers rationally use SNS by suggesting measures to reduce dissatisfaction among consumers of the SNS market. In addition, the company or individual sellers will identify various dissatisfaction and complaints behavior from consumers of the SNS market and provide them as basic data for providing reasonable services and operating the SNS market. The main results and suggestions of this study are as follows. First, the dissatisfaction of the SNS market was most affected by the negative expectancy disconfirmation, followed by time risk, refund/exchange risk, economic benefit, information benefit, and economic risk. Second, the dissatisfaction of the social media market was affecting both private and public complaints. Third, it was found that subjective knowledge of SNS market had a moderating effect in the relationship between dissatisfaction and personal dissatisfaction.

      • Inhibition of Metastasis and Invasion of Ovarian Cancer Cells by Crude Polysaccharides from Rosa Roxburghii Tratt in Vitro

        Background: Rosa Roxburghii Tratt is a promising wild fruit crop in Southwest China. Its extracts have been used as traditional Chinese medicine, which benefit immune responses and cure various health disorders. However, whether Rosa Roxburghii Tratt polysaccharides could inhibit metastasis and invasion of ovarian cancer cells remains unknown. Materials and Methods: Effects of crude polysaccharides from Rosa Roxburghii Tratt on the viability of ovarian cancer A2780 cells were detected by MTT assay. Ovarian carcinoma cell migration and invasion after exposure to Rosa Roxburghii Tratt polysaccharides were quantified by wound healing and Transwell assays, respectively. Western blotting was applied to assess protein levels of MMP-9. Results: The results indicated that Rosa Roxburghii Tratt polysaccharides significantly reduced wound closure rate of A2780 cells, inhibited their migration and invasion, and suppressed the expression of MMP-9. Conclusions: Our findings indicated that Rosa Roxburghii Tratt polysaccharides have potential for develop as anti-metastatic cancer drug preparations for ovarian cancer patients.

        Screening, Gene Cloning, and Characterizations of an Acid-Stable α-Amylase

        Based on its α-amylase activity at pH 5.0 and optimal pH of the crude enzyme, a strain (named B-5) with acid α-amylase production was screened. The B-5 strain was identified as Bacillus amyloliquefaciens through morphological, physiological, and biochemical characteristics analysis, as well as 16S rDNA phylogenetic analysis. Its α-amylase gene of GenBank Accession No. GU318401 was cloned and expressed in Escherichia coli. The purified recombinant α-amylase AMY-Ba showed the optimal pH of 5.0, and was stable at a pH range of 4.0-6.0. When hydrolyzing soluble starch, amylose, and amylopectin, AMY-Ba released glucose and maltose as major end products. The α-amylase AMY-Ba in this work was different from the well-investigated J01542-type α-amylase which also came from B. amyloliquefaciens. AMY-Ba exhibited notable adsorption and hydrolysis ability towards various raw starches. Structure analysis of AMY-Ba suggested the presence of a new starch-binding domain at its C-terminal region.

        The comparison of microbial communities in thyroid tissues from thyroid carcinoma patients

        Thyroid carcinoma is a common endocrine organ cancer associated with abnormal hormone secretion, leading to the disorder of metabolism. The intestinal microbiota is vital to maintain digestive and immunologic homeostasis. The relevant information of the microbial community in the gut and thyroid, including composition, structure, and relationship, is unclear in thyroid carcinoma patients. A total of 93 samples from 25 patients were included in this study. The results showed that microbial communities existed in thyroid tissue; gut and thyroid had high abundance of facultative anaerobes from the Proteobacteria phyla. The microbial metabolism from the thyroid and gut may be affected by the thyroid carcinoma cells. The cooccurrence network showed that the margins of different thyroid tissues were unique areas with more competition; the stabilization of microcommunities from tissue and stool may be maintained by several clusters of species that may execute different vital metabolism processes dominantly that are attributed to the microenvironment of cancer.

        Synthesis and Biological Activities of Novel Artemisinin Derivatives as Cysteine Protease Falcipain-2 Inhibitors

        A series of novel artemisinin derivatives were synthesized from artemisinin and different anilines. All compounds were obtained as β-isomers. The target compounds were evaluated for inhibition activity against Plasmodium falciparum falcipain-2 in vitro, and most of them exhibited potent inhibition in the low micromolar range and proved to be new types of falcipain-2 inhibitors.

        Optimal impulse and regular control strategies for proportionalreinsurance problem

        We formulate a stochastic control problem on proportional reinsurance that includes impulse and regular control strategies. For the first time we combine impulse control with regular control, and derive the expected total discount pay-out (return function) from present to bankruptcy. By relying on both stochastic calculus and the classical theory of impulse and regular controls, we state a set of sufficient conditions for its solution in terms of optimal return function. Moreover, we also derive its explicit form and corresponding impulse and regular control strategies.

        Cyclic Softening Properties of 30Cr2MoV Steel at Elevated Temperatures

        Low cycle fatigue tests for 30Cr2MoV steel are done at both room and elevated temperatures and their softening properties are investigated. Based on the experimental results, a quantitative relationship between hardness and remaining life is established, allowing prediction of the remaining life of metals. A modified Mason-Coffin equation including the aging effect is also derived. Both the experimental and theoretical results show that it is useful to be able to predict the remaining life of a steam turbine rotor through the measurement of hardness.

        During the network communication process, attacker carry on ARP spoofing by using the disadvantage of the ARP(Address Resolution Protocol) protocol, this phenomenon seriously threatens the LAN security. This paper will introduce the commonly used ARP spoofing methods such as internal/external network sniffing, interception, malicious attack and so on. It presents the Matching IP method, Data monitor method, Echo time method, ARP response analysis method, software tools detecting method as well as the new method of ARP cache updating, using switching equipment to control and other strategy and presents the algorithm to keep ARP spoofing away and maintain network security.

      • A Hybrid Model for Short-Term Load Forecasting Based on Non-Parametric Error Correction

        In this paper, we presented the performance of forecasting model and error correction will affect the accuracy of short-term load forecasting. Least squares support vector machines (LS-SVM) based on improved particle swarm optimization is selected as load forecasting model. Forecasting accuracy and generalization performance of LS-SVM depend on selection of its parameters greatly. Adaptive particle swarm optimization (APSO) based on fitness function was put forward to optimize the kernel parameter σ and regularization parameter γ of LS-SVM. Based on the optimized forecasting model, non-parametric error correction model is also presented by iterative method. The error forecasted by non-parametric model was used to update the forecasted load so as to improve the forecasting accuracy. Load data selected from some area in South China as training and forecasting data is used to analyze. Case study illustrates that the proposed forecasting model (NP-APSO-SVM) has more generalized performance and better forecasting accuracy compared with the method of standard SVM.
