        The Influence of AH-26 and Zinc Oxide-Eugenol Root Canal Sealer on the Shear Bond Strength of Composite Resin to Dentin

        AH-26 근관실러가 상아질에 대한 복합레진의 전단결합강도에 미치는 영향을 평가하기 위하여 144개의 발거된 대구치를 실린더형의 몰드에 매식한 다음 치관부의 상아질 표면을 노출시킨 후 편평하게 연마하고 AH-26을 도포한 군, ZOE paste를 도포한 군 및 실러를 도포하지 않은 대조군으로 나누어 One-step 상아질 접착제를 처리한 후에 Charisma (Heraeus Kulzer, Germany) 복합레진을 적용하고 광중합시켰다. 시편을 분리하여 37℃ 항온조에 24시간 보관후 Instron test machine (Model 4202, Instron Corp., USA)을 이용하여 시편의 전단 결합 강도를 측정하고 one-way ANOVA 및 Tukey's studentized rank test로 통계 분석하였다. AH-26 근관실러로 처리한 군과 대조군은 ZOE 근관실러 처리군에 비해 유의하게 놓은 접착강도를 나타내었다 (p<0.05). AH-26 근관실러 처리군과 아무 처리하지 않은 대조군 사이에는 결합강도에 있어 유의한 차이를 나타내지 않았다 (p>0.05). 본 연구의 조건에서는 ZOE 근관실러는 상아질에 대한 복합레진의 전단결합강도를 저하시키는 반면 AH-26 근관실러는 유의한 영향을 미치지는 않는 것으로 보인다. 따라서 임상에서 근관실러로 AH-26을 사용한 근관충전 후에는 즉시 복합레진 코어를 해 주어도 결합력에 저해가 없을 것으로 여겨진다. The purpose of this study was to evaluate the influence of the AH-26 root canal sealer on the shear bond strength of composite resin to dentin. One hundred and forty four (144) extracted, sound human molars were used. After embedding in a cylindrical mold, the occlusal part of the anatomical crown was cut away and trimmed in order to create a flat dentin surface. The teeth were randomly divided into three groups; the AH-26 sealer was applied to the AH-26 group, and zinc-oxide eugenol (ZOE) paste was applied to the ZOE group. The dentin surface of the control group did not receive any sealer. A mount jig was placed against the surface of the teeth and the One-step dentin bonding agent was applied after acid etching. Charisma composite resin was packed into the mold and light cured. After polymerization, the alignment tube and mold were removed and the specimens were placed in distilled water at 37℃ for twenty four hours. The shear bond strength was measured by an Instron testing machine. The data for each group were subjected to one-way ANOVA and Tukey's studentized rank test so as to make comparisons between the groups. The AH-26 group and the control group showed significantly higher shear bond strength than the ZOE group (p<0.05). There were no significant differences between the AH-26 group and the control one (p>0.05). Under the conditions of this study, the AH-26 root canal sealer did not seem to affect the shear bond strength of the composite resin to dentin while the ZOE sealer did. Therefore, there may be no decrease in bond strength when the composite resin core is built up immediately after a canal filling with AH-26 as a root canal sealer.

        Efficient Interleukin-21 Production by Optimization of Codon and Signal Peptide in Chinese Hamster Ovarian Cells

        Interleukin-21 is a common γ-chain cytokine that controls the immune responses of B cells, T cells, and natural killer cells. Targeting IL-21 to strengthen the immune system is promising for the development of vaccines as well as anti-infection and anti-tumor therapies. However, the practical application of IL-21 is limited by the high production cost. In this study, we improved IL-21 production by codon optimization and selection of appropriate signal peptide in CHO-K1 cells. Codon-optimized or non-optimized human IL-21 was stably transfected into CHO-K1 cells. IL-21 expression was 10-fold higher for codon-optimized than non-optimized IL-21. We fused five different signal peptides to codon-optimized mature IL-21 and evaluated their effect on IL-21 production. The best result (a 3-fold increase) was obtained using a signal peptide derived from human azurocidin. Furthermore, codon-optimized IL-21 containing the azurocidin signal peptide promoted IFN-γ secretion and STAT3 phosphorylation in NK-92 cells similar to codon-optimized IL-21 containing original signal peptide. Collectively, these results indicate that codon optimization and azurocidin signal peptides provide an efficient approach for the high-level production of IL-21 as a biopharmaceutical.

        소아에서 간질발작의 임상양상과 뇌파검사의 관계

        목적:발작의 분류는 발작의 원인, 예후를 추정하거나 항경련제를 선택하는 데 있어 매우 중요하며 뇌파검사는 발작의 종류를 구분하고 적절한 치료를 하기 위해 시행하는 중요한 검사이다. 저자들은 소아에서 임상적 발작이 확실한 경우 간질발작의 임상양상과 뇌파검사 간의 일치 정도를 알아보고자 한다. 방법:2000년 1월 1일부터 2008년 9월 30일까지 1회 이상의 간질발작을 주소로 가천의대 길병원 소아청소년과에 방문한 환자 중 본원에서 처음으로 뇌파검사를 시행한 환자를 대상으로 하였다. 발작의 임상양상은 목격자와 본인의 설명을 기록한 의무기록지를 기준으로 하여 부분발작과 전신발작으로 분류하였고 뇌파검사는 발작간간질양방전을 기준으로 정상과 비정상으로 분류하였으며 각성과 수면상태 모두에서 시행하였다. 결과:총 461명의 환자의 뇌파검사 시의 평균 연령은 6.7세였고 남자는 247명, 여자는 214명이었다. 전체 환자에서 발작의 임상양상은 부분발작이 310명, 전신발작이 187명이었다. 각성뇌파검사에서 비정상은 158명으로 부분발작파가 118명, 전신발작파가 59명, 두 가지 발작파가 한 환자에서 나온 경우가 19명이었고, 수면뇌파검사에서는 비정상이 239명으로 부분발작파가 196명, 전신발작파가 77명, 두 가지 발작파가 한 환자에서 나온 경우가 34명이었으며 전체 환자에서는 비정상이 273명으로 부분발작파가 216명, 전신발작파가 97명, 두 가지 발작파가 한 환자에서 나온 경우가 40명이었다. 간질증후군은 전체 환자 중 90명에서 관찰되었다. 146명(41.5%)의 부분발작과 44명(23.4%)의 전신발작 및 79명(87.8 %)의 간질증후군이 뇌파소견과 일치하였다. 결론:영유아 및 소아에 있어 발작을 진단할 때에는 발작양상 분 아니라 정확한 뇌파소견을 세심하게 고려하여야 한다. Purpose:Electroencephalography(EEG) is an essential method carried out for classifying seizures and taking appropriate treatment. The aim of this study is to investigate the concordance between clinical findings of epileptic seizures and EEG in children. Methods:We enrolled 461 patients from those who visited Gil hospital from January 1, 2000 to September 30, 2008 with the chief complaint of epileptic seizure more than once and checked their first EEG at the same hospital. The clinical findings of seizure were based on the charts and interictal waking and sleep EEGs were done. Results:The mean age of the patients was 6.7 years old. 497 epileptic seizures occurred and its clinical finding included 310 of partial seizures and 187 of generalized seizures. In 315 waking EEG 158 were abnormal including 118 of partial seizures, 59 of generalized seizures and 19 of both seizures, in 431 sleep EEG 239 were abnormal including 196 of partial seizures, 77 of generalized seizures and 34 of both seizures, and in waking and/or sleep EEG 273 were abnormal including 216 of partial seizures, 97 of generalized seizures and 40 of both seizures. Epileptic syndromes were observed in 90 patients. 146(41.5%) of partial seizures, 44(23.4%) of generalized seizures, and 79(87.8%) of epileptic syndromes accorded with EEG. Conclusion:When diagnosing seizures in children, we must consider not only clinical findings but also accurate EEG findings.

      • Construction of Stably Transformed Bm 5 Cells by Using Autographa californica Nuclear Polyhedrosis Virus IE 0 Gene

        To construct transfurmed Bm5 cells, Autographa californica nuclear polyhedrosis virus(AcNPV) IE1 gene, an immediate early viral gene was firstly used in this study. AcNPV IE1 gene, which shares on 95.3% nucleotide sequence homology with Bombyx mori nuclear polyhedrosis virus (BmNPV) IE1 gene, was isolated and cloned into pBluescript. Neomycin gene from pKO-neo was inserted under the control of the IE1 promoter to yield pAcIE1-neo. The plasmid pAcIE1-neo was transfected into Bm5 or Sf9 cells, and neomycin-resistant cells were selected in TC100 medium containing 10% fetal bovine serum (FBS) and 1 mg/ml G4l8 for two weeks. Individual clones were picked and each was amplified for further characterization. The genomic DNA from neomycin-resistant cells was isolated and characterized by PCR using AcNPV IEI gene-specific primers and by Southern blot analysis using neomycin gene probe. We concluded that AcNPV IE1 gene was functional in B. mori-derived Bm5 cells as well as Spodoptera fugjprrda-derived Sf9 cells to produce stably-transformed insect cells

        A Study on the Characteristics of North Korea’s Normalization and Standardization System in the Kim Jong-un Era: Focusing on the Reports of the Rodong Sinmun

        In the 21<sup>st</sup> century, standards have become ‘the universal language of industries that transcend borders and races.’ This article seeks to investigate the development of normalization and standardization in North Korea’s industrial sector during the Kim Jong-un era by analyzing North Korean media sources. By comparing the policies pursued under Kim Ilsung, Kim Jong-il, and Kim Jong-un, it describes the gradual adoption of international standard systems in North Korea. It points out that it is not easy to achieve practical results by simply emphasizing the necessity of standardization and normalization if other industrial conditions are insufficient. Still, the North Korean authorities’ efforts to draw attention to the importance of normalization and standardization must be regarded as a positive trend.

        Trunk Impairment Scale for Predicting Lumbar Spine Bone Mineral Density in Young Male Patients With Subacute Stroke

        Objective To investigate the relationship between Trunk Impairment Scale (TIS) and lumbar spine bone mineral density (BMD) in subacute stroke patients. Methods Twenty-three subacute male stroke patients under the age of 65 were prospectively enrolled to exclude both postmenopausal and senile effects on BMD. The TIS, Berg Balance Scale, the Korean version of the Modified Barthel Index, and manual muscle test were measured at admission and 3 months after stroke onset. BMD of the bilateral lower extremities and lumbar vertebrae was measured by dual-energy X-ray absorptiometry 3 months after stroke onset. Results TIS at baseline (TIS_B) and TIS at 3 months after stroke (TIS_3m) showed significant correlations with lumbar BMD (TIS_B, r=0.522; TIS_3m, r=0.517). Through multiple regression analysis, the TIS_B was associated with lumbar BMD (adjusted R2=0.474). However, BMD of the bilateral lower extremities was not correlated with any clinical measurements except body mass index. Conclusion We found a relationship between TIS_B and lumbar BMD in subacute young male stroke patients. Stroke patients with poor trunk control in the early subacute stage would have low BMD of vertebral bones at 3 months. The TIS can be useful for estimating bone fragility in the lumbar vertebrae of subacute stroke patients.

      • Generation of immortalized marmoset fibroblast cell lines by a CRISPR-Cas9-mediated gene targeting

        Immortal cell lines are suitable for diverse in vitro experiments, which are prerequisite for effective animal testing. Callithrix jacchus, the common marmoset is an important non-human primate model used for establishing various human diseases. Several strategies generating immortal marmoset cell lines have been reported, but only a limited number of immortalized marmoset cells are now available. Previous studies using gene knockout mice have shown that p53- or Ink4a/Arf-deficient mouse embryonic fibroblasts (MEFs) are immortal and do not express any signs of replicative senescence. In this study, we utilized lentiviruses expressing CRISPR-Cas9 endonucleases and showed that their gene targeting can be convenient procedures for the immortalization of primary marmoset fibroblasts from skin biopsies. When we destroyed the sequence encoding the DNA-binding motif of p53 protein, in-frame mutations that possible lost the function of p53 as a transcription factor, were frequently detected. We designed an sgRNA to destroy both INK4A gene and the alternative ARF gene, and most mutations resulted in frameshift. Especially, as INK4a/ARF-targeted marmoset cells still expressed p53 proteins, the immortalized marmoset cells that we generated can be useful for studying p53-dependent/p53-independent genotoxic stresses of drugs. Therefore, our results indicate that Cas9-mediated p53 or INK4A/ARF targeting is a robust tool for establishing immortalized marmoset cell lines with defined genetic alterations.
