A 12-week repeated-dose oral toxicity study of water-soluble Orostachys japonicus extract (WOJ) was performed in Sprague-Dawley (SD) rats of both genders. Each group of ten rats was orally administered in doses of either 0 or 250 mg/day over a 12-week period. As a result, no WOJ-related changes were observed in terms of survival rate, clinical signs, body weight, or food intake. In addition, no difference in organ weight between the control and treated groups was detected. Furthermore, serum biochemistry parameters revealed some changes within normal ranges although significant decreases in total-bilirubin in the females. In spite of some alterations in serum biochemistry, the clinical signs, body weight changes from food intake, and autoptical remarks indicated that WOJ was not toxic. This study suggests that repeated treatment of O. japonicus very low toxicity and the NOAEL (no observed adverse effect dose) of WOJ exceeds 250 mg/kg in the SD rats.

<P><I>Context</I>: <I>Orostachys japonicus</I> (Crassulaceae) is referred to as Wa-song in Korea. It is used as an anti-inflammatory, antifebrile, hemostatic, and anti cancer agent, and as an antidote.</P><P><I>Objective</I>: The purpose of this study was to evaluate the acute toxicity of the ethyl acetate fraction of <I>O. japonicus</I> (OJE) after the oral administration in Balb/c mice of both sexes.</P><P><I>Materials and methods</I>: Mice were oral administered a single doses of 500, 1000, and 2000 mg/kg of body weight and were monitored for 14 d. Biochemical parameters [aspartate amino transferase (AST), alanine amino transferase (ALT), alkaline phosphatase (ALP), total protein (TP), globulin (GB), total cholesterol (TC), triglyceride (TG), blood urea nitrogen (BUN), and creatinine (CR)] and histopathological examination of liver were performed.</P><P><I>Results and conclusion</I>: No animals died and no toxic changes were observed in clinical signs, body weight, and organ weight. The LD<SUB>50</SUB> of orally administered OJE was higher than 2000 mg/kg/d in both sexes. No toxicological findings were found in biochemical parameters. In histophathological examination, neutrophilic infiltration was observed at a dose of 2000 mg/kg group in both sexes. These finding suggest that oral administration of OJE does not produce acute toxicity. Therefore, these results could provide satisfactory preclinical evidence of safety to launch clinical trials on standardized formulation of OJE to be a biohealth product.</P>

<P>We investigated the anticancer mechanisms of the ethylacetate (EtOAc) fraction from Orostachys japonicus in human gastric cancer (AGS) cells. Flow cytometric analysis revealed that the number of total apoptotic cells following treatment with the EtOAc fraction increased in a dose-dependent manner. In the cell cycle analyses, the EtOAc fraction increased the peak in the sub-G1, indicating apoptosis, and in the G2/M phases in a dose-dependent manner. In the RT-PCR analysis, the expression of cyclin-dependent kinase 1 (CDK 1) and cyclin B1 decreased in a dose- and time-dependent manner. The results of western blotting revealed that the protein levels of p53, cytochrome c, and cleaved caspase-3, -8 and -9 proteins increased and those of B cell lymphoma-2 (bcl-2) and pro-caspase-3, -8 and -9 proteins decreased in a dose- and time-dependent manner, whereas the levels of bcl-2-associated x protein (bax) remained unchanged. Furthermore, the changes in the levels of pro-caspase-3, -8 and -9 and cleaved caspase-3, -8 and -9 were abolished by the pan-caspase inhibitor Z-VAD-FMK. In addition, phosphorylation of p38 and JNK increased in a time-dependent manner. These results, for the first time, provide an understanding of the potential anticancer activity of the O. japonicus, which functions through the induction of apoptosis and cell cycle arrest.</P>

Compositionally stepped HfxAlyOz dielectric thin films were fabricated for application to DRAM capacitor dielectrics. Compositionally stepped HfxAlyOz dielectric thin film is composed of an Al rich bottom layer and a Hf rich top layer between top and bottom TiN electrodes. The leakage current of TIT (TiN/Insulator/TiN) capacitors with compositional stepped HfxAlyOz could be lowered by compositional stepping and accompanying post ozone treatment, respectively. The prepared TIT capacitor showed drastically reduced leakage current and highly improved break down voltage characteristics of 0.45~0.75 V, while maintaining the same equivalent oxide thickness (EOT). In the case of stepped film with Hf/Al=1/1, EOT showed a very small value of 12.8 Å while maintaining leakage current as low as 2×10(-16)A/cell at +1.0 V.

In this paper, the 1/f noise characteristics of n-channel MOSFET (NMOSFET) and p-channel MOSFET (PMOSFET) are analyzed in depth as a function of body bias. The normalized drain current noise, $S_{ID}/I_D{^2}$ showed strong dependence on the body bias in the sub-threshold region for both NMOSFET and PMOSFET, and NMOSFET showed stronger dependence than PMOSFET on the body bias. On the contrary, both of NMOSFET and PMOSFET do not exhibit the dependence of $S_{ID}/I_D{^2}$ on body bias in strong inversion region, although the noise mechanisms of two MOSFETs are different from each other.

This paper proposes a coordinated control for an independent multi-phase transverse flux linear synchronous motor (IM-TFLSM) based on magnetic levitation. The stator structures of the IM-TFLSM are composed of a two set, which has independent three-phase windings and a double-sided air-gap as opposed to the conventional Y-connected three-phase linear motors. A suitable control algorithm is necessary to operate the applied linear machine. This study proposes a coordinated control algorithm for adjusting the mover air-gap and thrust force of the IM-TFLSM in order to maintain air-gap and phase shifted current control of the independent 3-phase modules. In addition, the principle of operation and its special structures are described in detail and the validity and effectiveness of the control algorithm is verified through multiple experimental results.

The most promising treatment for stress urinary incontinence can be a cell therapy. We suggest human amniotic fluid stem cells (hAFSCs) as an alternative cell source. We established the optimum in vitro protocol for the differentiation from hAFSCs into muscle progenitors. These progenitors were transplanted into the injured urethral sphincter and their therapeutic effect was analyzed. For the development of an efficient differentiation system in vitro, we examined a commercial medium, co-culture and conditioned medium (CM) systems. After being treated with CM, hAFSCs were effectively developed into a muscle lineage. The progenitors were integrated into the host urethral sphincter and the host cell differentiation was stimulated in vivo. Urodynamic analysis showed significant increase of leak point pressure and closing pressure. Immunohistochemistry revealed the regeneration of circular muscle mass with normal appearance. Molecular analysis observed the expression of a larger number of target markers. In the immunogenicity analysis, the progenitor group had a scant CD8 lymphocyte. In tumorigenicity, the progenitors showed no teratoma formation. These results suggest that hAFSCs can effectively be differentiated into muscle progenitors in CM and that the hAFSC-derived muscle progenitors are an accessible cell source for the regeneration of injured urethral sphincter.

Objectives: This study was performed to report the effect of oriental medical treatment in the paraplegia and pain after epidural nerve block. Methods: A 39-year-old woman who underwent epidural nerve block at a local clinic was admitted with motor weakness of lower limbs, severe lower radiating pain and decreased sensation when voiding and defecating. We treated her by acupuncture, a herbal medicine, a bee venom injection, moxibustion and cupping treatment and physical theraphy from 11th July 2008 to 14th October 2008. Results: After treatment, most symptoms decreased, VAS(Visual Analog Scale) score changed from 9 to 2. and examinated muscle power changed from 2-3 to 4-5. Conclusions: Our study suggested that oriental medical treatments are significantly effective in the paralplegia and pain after epidural nerve block. And further studies will be aid to identify underlying mechanism of treatment.

Objectives: The aim was to study the effect of bee venom pharmacopuncture therapy with different concentration on rheumatoid arthritis rat model. Methods: We enforced a bee venom pharmacopuncture therapy with different concentration on rheumatoid arthritis rat model by the intradermal injection of chicken type Ⅱ collagen emulsified. 14 days after the onset of the rheumatoid arthritis rat model, a fixed volume of bee venom was daily injected to ST-35 acupoint in the rat`s knee joint for 2-3 weeks. The hind paw volume, arthritic index, arthritic flexion pain test, pain threshold, and serum analysis(CRP, PGE2, ALT, AST) were analyzed, and the expression profiles of COX-2, c-fos, and substance-P at the dorsal horn region of the spinal cord and subchondral bone of the knee joint were also analyzed by using the immunohistochemistry. Results: After the treatment of rheumatoid arthritis rats with bee venom pharmacopuncture, the paw volume of edema of arthritic rats were almost restored to the level of normal group, and behavior tests were very effective. Also the evaluation on the blood serum analysis was remarkable. COX-2, c-fos, and substance-P positive cells in the immunohistological section of dorsal horn region of the spinal cord and subchondral bone of the knee joints were significantly decreased. also the bee venom pharmacopuncture was effective to alleviate their rheumatoid arthritic inflammation cytokine inhibition as regards to the behavior tests and joint histological appearance. Conclusions: Based on the results in this study, bee venom pharmacopuncture with concentrated treatment condition was very effective in low fixed quantity and progressive low increased quantity.
