New Report of Three Unrecorded Species in Trichoderma harzianum Species Complex in Korea

The genus Trichoderma (Hypocreaceae, Ascomycota) consists of globally distributed fungi. Among them, T. harzianum, one of the most commonly collected Trichoderma species, had been known as a polyphyletic or aggregate species. However, a total of 19 species were determined from the polyphyletic groups of T. harzianum. Thus, we explored Korean “T. harzianum” specimens that were collected in 2013-2014. These specimens were re-examined based on a recent study with translate elongation factor 1-alpha (EF1a) sequences to reveal cryptic Trichoderma species in Korea. As a result, four different species, T. afroharzianum, T. atrobruneum, T. pyramidale, and T. harzianum, were identified. Except T. harzianum, the other three species have not been reported in Korea. In this work, we describe these species and provide figures.
Wood Decay Fungi in South Korea: Polypores from Seoul

In Seoul, a majority of plant communities have undergone significant changes over the last few decades; however, how wood decay fungi have responded and adapted to the changes in vegetation remains unknown. Through an ongoing investigation of Korean indigenous fungi, ca. 300 specimens with poroid basidiocarp were collected in Seoul during 2008~2012. Morphological examination and molecular analysis using the internal transcribed spacer and nuclear large subunit ribosomal DNA region sequences helped identify 38 species belonging to 28 genera, 10 families, and 5 orders in this area. Among them, three polypores, Abundisporus pubertatis, Coriolopsis strumosa, and Perenniporia maackiae were found to be new to South Korea.

Despite the huge worldwide diversity of Trichoderma (Hypocreaceae, Ascomycota), only about 22 species have been reported in Korea. Thus, between 2013 and 2015, soil-derived Trichoderma spp. were isolated to reveal the diversity of Korean Trichoderma. Phylogenetic analysis of translation elongation factor 1 alpha gene was used for identification. Among the soilderived Trichoderma, Trichoderma albolutescens, T. asperelloides, T. orientale, T. spirale, and T. tomentosum have not been previously reported in Korea. Thus, we report the five Trichoderma species as new in Korea with morphological descriptions and images.
New Records of Two Agarics: Galerina sideroides and Gymnopus luxurians in South Korea

Studies on the diversity of indigenous fungi were conducted in the central districts of South Korea from 2008 to 2013. During the studies, potentially unrecorded agarics were collected. All specimens collected in this study were examined morphologically and phylogenetic analysis was also performed. They were identified as Galerina sideroides and Gymnopus luxurians. These fungi have never been reported in South Korea. We report them here with detailed descriptions and figures.

Gymnopus is a cosmopolitan genus of agaric fungi and consists of ~300 species. In Korea, Gymnopus represents common saprobic mushrooms, and 12 species have been reported in Korea. Several Gymnopus specimens were collected in Korea between 2008 and 2015. To identify them exactly, phylogenetic analysis was performed by means of the internal transcribed spacer region of ribosomal-DNA sequences from the collected Gymnopus specimens. Among them, G. iocephalus, G. polygrammus, and G. subnudus have not been reported in Korea. A phylogenetic tree and images are provided.

Arthrinium (Ascomycota, Apiosporaceae) is a major marine fungal genus. Three Arthrinium species were reported previously, but not fully described. We isolated the two species A. arundinis and A. saccharicola from Sargassum sp. brown algae in Jeju Island, Korea. These species have not been previously reported in Korea. We described these species in detail and performed a phylogenetic analysis based on the nucleotide sequences of the EF1-α and β-tubulin genes.

균류에 의한 부후는 목재의 구조적 강도에 큰 영향을 주지 않으면서 적절한 수준으로 목재의 밀도를 감소시켜서 목재의 음색을 개선시킬 수 있다. 따라서 본 연구에서는 기타 제조에 사용되는 5 종의 목재로 시편을 제작하였으며, 여기에 각각 10 종의 목재부후균을 접종하여 4 주간 부후실험을 진행하였다. 주파수 응답 함수 방법으로 목재 시편의 공진주파수를 측정하였다. 실험을 통해 얻은 수치를 이용하여 목재의 Acoustic constant (A), Acoustic conversion efficiency (ACE) 값을 계산하였다. 대체로 목재의 중량감소율과 A 값 증가율은 비례하는 것으로 나타났으며, 따라서 목재부후에 의한 음향특성 개선 효과를 확인할 수 있었다.
국내 목재부후균 유래의 개미산 탈수소화효소 탐색 및 기능 분석

최근 전지구적인 기후변화가 진행됨에 따라서, 그 원인 물질로 이산화탄소(Carbon dioxide, CO<sub>2</sub>)가 주목받고 있다. 따라서 대기 중의 이산화탄소 농도를 줄이기 위한 이산화탄소 저감 및 자원화 기술에 대 하여 다양한 연구가 진행되고 있다. 그 중에서 포름산 탈수소화효소(Formate dehydrogenase, FDH)를 이용하면 생물학적인 방법으로 이산화탄소를 유기산인 포름산 전환할 수 있는 가능성이 보고되어 있다. FDH는 자낭균 효모인 Candida boidinii의 효소가 잘 알려져 있고 연구가 많이 되어 있지만, C. boidinii FDH의 이산화탄소 환원 효율이 낮기 때문에 추가적인 효소 생산 미생물의 탐색이 이루어지고 있다. 본 연구에서는 진균 중에서도 산림유전자원인 목재부후균 유래 FDH 발굴 연구를 수행하고 있으며, 배양체에 대한 포름산 저항성 시험을 통해 Phanerochaete sordida와 Wolfiporia cocos를 잠재적인 우수균주로 선별하였다. 각 균주의 균사체를 배양하여 RNA를 추출하고, RT-PCR을 통해 각 균주 FDH mRNA의 cDNA를 확보하였다. 얻은 mRNA는 둘 다 1077 bp였으며, 예상되는 단백질 크기의 경우 P. sordida FDH (PsFDH)는 39.27 kDa, W. cocos FDH (WcFDH)는 39.24 kDa로 거의 같았다. 각 단백질의 과발현 실험을 위하여 QIAexpress Type IV Kit를 이용하였다. 각 cDNA와 pQE-30 벡터에 제한효소 BamHI, SmaI로 처리하고, T4 DNA 리가아제를 이용하여 접합시켰다. 그리고 재조합된 플라스미드를 M15[pREP4] 대장균에 삽입하였다. PCR로 FDH 유전자의 삽입 여부를 확인하여 형질전환 균주를 선별하고, 배양 후 IPTG를 투입하여 각 FDH를 과발현 시키고, Ni-NTA로 정제하여 SDS-PAGE를 통해 결과를 확 중인 중에 있다.
