        Development of PCR Assay for Identification of Buffalo Meat

        A polymerase chain reaction (PCR) assay was developed to differentiate buffalo meat from the meat of Ceylon spotted deer (Axis axis ceylonensis), Ceylon sambhur (Cervus unicolor unicolor), cattle (Bovine), goat (Caprine), pig (Porcine), and sheep (Ovine). A set of primers were designed according to the sequence of the mitochondrial cytochrome b gene of bubalus bubalis and by PCR amplification a band of approximately 242 bp band was observed with buffalo DNA. These primers did not cross-react with DNA of other animal species tested in the study under the specified reaction conditions. A band of 649 bp was observed for all animal species tested when DNA was amplified with the universal primers indicating the presence of mitochondrial DNA in the samples. The technique was sensitive enough to identify rotten (10 days post slaughter), dried and cooked buffalo meat. The absence of a cross reaction with human DNA using the buffalo specific primers eliminates possible false positive reactions.

        Synthesis, Characterization and Antimicrobial Activity of Garcinol Capped Silver Nanoparticles

        Garcinol, a well-known medicinal phytochemical, was extracted and isolated from the dried fruit rinds of Garcinia quaesita Pierre. In this study, garcinol has successfully used to reduce silver ions to silver in order to synthesize garcinol-capped silver nanoparticles (G-AgNPs). The formation and the structure of G-AgNPs were confirmed by UV-visible spectroscopy, transmission electron microscopy and Fourier transform infrared spectroscopy. The antimicrobial activity of garcinol and G-AgNPs were investigated by well diffusion assays, broth micro-dilution assays and time-kill kinetics studies against five microbial species, including Staphylococcus aureus (ATCC 25923), Pseudomonas aeruginosa (ATCC 27853), Escherichia coli (ATCC 25922), Candida albicans (ATCC 10231) and clinically isolated methicillin-resistant Staphylococcus aureus (MRSA). The formation of G-AgNPs is a promising novel approach to enhancing the biological activeness of silver nanoparticles, and to increase the water solubility of garcinol which creates a broad range of therapeutic applications.

      • Analysis of electrochemical double-layer capacitors using a Natural Rubber-Zn based polymer electrolyte

        Electrochemical double-layer capacitors (EDLCs) based on solid polymer electrolytes (SPEs) have gained an immense recognition in the present world due to their unique properties. This study is about preparing and characterizing EDLCs using a natural rubber (NR) based SPE with natural graphite (NG) electrodes. NR electrolyte was consisted with 49% methyl grafted natural rubber (MG49) and zinc trifluoromethanesulfonate ((Zn(CF<sub>3</sub>SO<sub>3</sub>)<sub>2</sub>-ZnTF). It was characterized using electrochemical impedance spectroscopy (EIS) test, dc polarization test and linear sweep voltammetry (LSV) test. NG electrodes were made using a slurry of NG and acetone. EIS test, cyclic voltammetry (CV) test and galvanostatic charge discharge (GCD) test have been done to characterize the EDLC. Optimized electrolyte composition with NR: 0.6 ZnTF (weight basis) exhibited a conductivity of 0.6 x 10<sup>-4</sup> Scm<sup>-1</sup> at room temperature. Conductivity was predominantly due to ions. The electrochemical stability window was found to be from 0.25 V to 2.500 V. Electrolyte was sandwiched between two identical NG electrodes to fabricate an EDLC. Single electrode specific capacitance was about 2.26 Fg<sup>-1</sup> whereas the single electrode discharge capacitance was about 1.17 Fg<sup>-1</sup>. The EDLC with this novel NR-ZnTF based SPE evidences its suitability to be used for different applications with further improvement.

        In silico detection and characterization of novel virulence proteins of the emerging poultry pathogen Gallibacterium anatis

        The pathogen Gallibacterium anatis has caused heavy economic losses for commercial poultry farms around the world. However, despite its importance, the functions of its hypothetical proteins (HPs) have been poorly characterized. The present study analyzed the functions and structures of HPs obtained from Gallibacterium anatis (NCTC11413) using various bioinformatics tools. Initially, all the functions of HPs were predicted using the VICMpred tool, and the physicochemical properties of the identified virulence proteins were then analyzed using Expasy's ProtParam server. A virulence protein (WP_013745346.1) that can act as a potential drug target was further analyzed for its secondary structure, followed by homology modeling and three-dimensional (3D) structure determination using the Swiss-Model and Phyre2 servers. The quality assessment and validation of the 3D model were conducted using ERRAT, Verify3D, and PROCHECK programs. The functional and phylogenetic analysis was conducted using ProFunc, STRING, KEGG servers, and MEGA software. The bioinformatics analysis revealed 201 HPs related to cellular processes (n = 119), metabolism (n = 61), virulence (n = 11), and information/storage molecules (n = 10). Among the virulence proteins, three were detected as drug targets and six as vaccine targets. The characterized virulence protein WP_013745346.1 is proven to be stable, a drug target, and an enzyme related to the citrate cycle in the present pathogen. This enzyme was also found to facilitate other metabolic pathways, the biosynthesis of secondary metabolites, and the biosynthesis of amino acids.

      • Design of a Track-Leg Hybrid Locomotive Mobile Robot

        The use of robotic platforms for operations in harsh and high risk environments such as mining sites has become more popular with the advancements within the field of mobile robotics. In this paper, the designing of a hybrid locomotive mobile platform that utilizes both tracked locomotion and legged-like movements to achieve mobility is presented. The proposed track-leg mechanism and power transmission methodology are discussed in detail along with the overall design framework. The functionality validation and analysis of the legged locomotion was done in a simulation environment using MSC ADAMS with open-loop track-leg velocity control. The designed mobile robot demonstrated the ability to traverse a flat terrain and overcome a semi-cylindrical obstacle without tipping over in the simulations for the selected gait. The mobile robot also consists of 3 degrees of freedom (DOF) robotic arm which facilitates total human replacement by enabling the ability to perform any sample collecting, inspecting, pick and place tasks. The design of the robotic arm is also presented within this paper.
