      • Styrene-co-lithium methacrylate ionomer의 기계적인 성질에 미치는 LiCl의 영향

        The effect of LiCl salt added to styrene-co-lithium methacrylate ionomer on the mechanical properties of ionomers was studied dynamic rnschanically. When the LiCl salt was added to a 100% Li-neutralized ionomer, the amount of added salt. in the range of 100-300% to ionic groups of the ionomer, did not change the mechanical properties of the ionomers, This implies that the added salt may form isolated LiCl salt aggregates in the polymer matrix. However, when LiCl was added to a 200% neutralized ionomer, the cluster T_(g) shifted to a low temperature and then the sample started to flow. This result suggests that in this case the LiCl salts reside in and near the multiplets, resulting the weakening of ionic interactions between ionic groups in multiplets.

        The dynamic mechanical properties of poly(styrene-b-isobutylene-b-styrene) and its sulfonated ionomers were investigated. The triblock copolymer showed a tan δ peak at ca. -25℃, which is ascribed to the T_(g) of polyisobutylene blocks. The copolymer also showed a shoulder on a tan δ curve at ca. 50℃, which is probably associated with crystallization and/or melting of the crystalline polyisobutylene blocks. Above 100℃, the copolymer started to flow. In the cases of ionomers, well developed rubbery plateaus, extended to ca. 150-160℃, were seen in modulus-temperature plots. A tan δ peak for the T_(g) of the polyisobutylene blocks and a shoulder peak were also observed for the tetramethylammonia neutralized sulfonated ionomer. However, the shoulder peak became smaller and shifted to low temperatures, which implies that the ionic interactions in this system reduces the ability of crystallization or/and melting of the polyisobutylene blocks. For the K neutralized ionomer sample, only a tanδ peak for the T_(g) of the isobutylene blocks was seen. This different result in these two ionomers is believed to be due to the difference in the strength of ionic interactions.

        The styrene-co-methacrylate ionomers neutralized with Na +, Ba2 +, and mixed cation of both were prepared. The dynamic mechanical properties of two cation-containing ionomers were measured and compared to those of the ionomers neutralized with only one type of cations. We found that in the ionomers with one type of cations the difference in the type of cation does not change the glass transition temperatures. It was found, however, that the cluster Tg of the mixed cation ionomer is lower than that of the single cation ionomers. Thus it was postulated that in the two-cation system the two cations in a solution have sufficient dynamics to form new type of multiplet containing both cations, which may be slightly bigger than that found in the single cation multiplet. The mechanical properties also imply that the strength of ion interactions in the multiplets of the two cation ionomer is thought to be weaker than that of the one cation ionomers. These two effects are believed to contribute the lower in the cluster glass transition of mixed cation system.

        Background: Recent studies suggest that miR-137 functions as tumor suppressor in various tumors including colorectal cancer, glioblastoma multiforme, oral cancer, and squamous cell carcinoma of the head and neck. The silencing of miR-137 could be related with its abnormal promoter hypermethylation. The purpose of this study was to investigate the significance of MIR137 promoter methylation in lung cancer. Methods: Lung cancer cell lines were treated with DNA methyltransferase inhibitor (5’-Aza) and/or HDAC inhibitor (Trichostatin A) whether miR-137 could be reactivated. From paired tumor and adjacent non-tumor lung tissues (n=50), real-time RT-PCR for miR-137 expression and cyclin dependent kinase 6 (CDK6), quantitative methylation specific PCR for methylation analysis, and bisulfite modified DNA sequencing for validation were used for the study. Results: miR-137 was reactivated by treatment with 5’-Aza and/or Trichostatin A in lung cancer cell lines. miR-137 was significantly downregulated and CDK6 was significantly upregulated in lung tumor tissues compared with adjacent non-tumor tissues. Quantitative methylation specific PCR showed increased MIR137 promoter methylation in lung tumor tissues compared with adjacent non-tumor tissues, which was further validated with bisulfite sequencing. Conclusions: These results suggest that miR-137 has a role of tumor suppressor and its expression is regulated by promoter methylation. The decreased expression of miR-137 combined with increased expression of CDK6 could be associated with lung cancer carcinogenesis.

        <P>Interleukin (IL)-8 plays a central role in the initiation and maintenance of inflammatory responses in the inflammatory bowel disease. The proinflammatory cytokine-mediated production of IL-8 requires activation of various kinases, which leads to the I kappa B degradation and NF-kappa B activation. We investigated the role of 18 beta-glycyrrhetinic acid (GA), a saponin isolated from licorice roots, on TNF-alpha-induced IL-8 production in human colonic epithelial cells. HT29 cells were stimulated with TNF-alpha in the presence or absence of GA (1, 5 or 10 microM). IL-8 production was measured by enzyme-linked immunosorbent assay (ELISA) and reverse transcriptase-polymerase chain reaction analysis, and the mitogen-activated protein kinases (MAPKs) activation and I kappa B alpha degradation were determined by Western blot analysis. GA suppressed TNF-alpha-induced IL-8 production in a concentration-dependent manner. In addition, GA inhibited TNF-alpha-induced phosphorylation of p38 MAPK and extracellular-regulated kinases (ERK), I kappa B alpha degradation, and NF-kappa B activation. These results suggest that GA has the inhibitory effects on TNF-alpha-induced IL-8 production in the intestinal epithelial cells through blockade in the phosphorylation of MAPKs, following I kappa B alpha degradation and NF-kappa B activation.</P>

        Inhibition of Trypsin-Induced Mast Cell Activation by Water Fraction of Lonicera japonica

        Lonicera japonica Thunb.(Caprifoliaceae) has long been known as an anti-inflammatory. In the present study, the effect of water fraction of Lonicera japonica (LJ) on trypsin-induced mast cell activation was examined. HMC-1 cells were stimulated with trypsin (100 nM) in the presence or absence of LJ (10, 100, and 1000 $\mu$ g/mL). TNF-$\alpha$ and tryptase production were measured by enzyme-linked immunosorbent assay (ELISA) and reverse transcription-PCR. Extracellular signal-regulated kinase (ERK) phosphorylation was assessed by Western blot. Trypsin activity was measured by using Bz-DL-Arg-p-nitroanilide (BAPNA) as substrate. LJ (10, 100, and 1000 $\mu$g/mL) inhibited TNF-$\alpha$ secretion in a dose-dependent manner. LJ (10, 100, and 1000 $\mu$g/mL) also inhibited TNF-$\alpha$ and tryptase mRNA expression in trypsin-stimulated HMC-1. Furthermore, LJ inhibited trypsin-induced ERK phosphorylation. However, LJ did not affect the trypsin activity even 1000 $\mu$g/mL. These results indicate that LJ may inhibit trypsin-induced mast cell activation through the inhibition of ERK phosphorylation than the inhibition of trypsin activity.

        The effects of crown ether on the dynamic mechanical properties of potassium sulfonated polystyrene ionomer were investigated. As expected, it was found that when crown ether molecules were added to the ionomer, the crown ether molecules interacted with K^(+) cations and weakened the ionic interaction between K^(+)/sulfonate ion pairs of the ionomer significantly. Thus, no multiplets were present, and, in turn, no clustering was found. When the ether was added to interact with 50% of K^(+) cations, weak clustering was observed: the clustering was slightly stronger than that of a 50% neutralized ionomer. From this result, it was speculated that in the 50% neutralized sample H^(+) ⇔ K^(+) cation exchange occurs rapidly between -SO₃H and -S0₃K groups in multiplets. which weakens ionic interactions. In the 50% crown ether-added sample, however, -S0₃- group, instead of -S0₃H, is present, and thus no cation exchange is expected to occur.

        The dynamic mechanical properties of polyfstyrene-co-sodium methacrylate) ionomer (3.4 mol% of ions) blended with three low-molecular-weight carboxylate organic salts. i.e. sodium benzoate, sodium isophthalata, and sodium benzenetetrabenzoate, were studied. When organic salts were added to ionomers to make 1:1 and 1:2 ratio of mole of caboxylate groups of ionomer/organic salts, it was found that the ionic modulus increased, but that cluster Tg shifted to lower temperature. Thus, it was suggested that part of organic salts participate in the multiplets with ionic groups of the ionomer, resulting in lowering cluster Tg, and that the rest of salts reside in matrix and form ionic aggregates, which act as filler. However. in a 1:8 blend, very weak clustering was found. This means that the ionic groups of ionomer start to form ionic aggregates with any ionic groups in their proximity; thus the number density of chains surrounding multiplets drops drastically, esulting in a very small amount of clusters.

        결혼 이주 여성들의 행위자성과 평생교육의 지향점 모색

        본 연구는 ‘결혼 이주’여성들에게 월경(月境)에 대한 욕망을 형성하는 세계화 맥락과, 필리핀의 경제구조 및 이민정책으로 인한 욕망 추동의 지형, 그리고 결혼생활의 경험을 통해 발현되는 결혼 이주에서의 행위자성을 드러내고자 한다. 행위자성에 대한 강조는 결혼 이주 여성들을 위해 실시되는 다양한 평생교육적 기획이 지금까지 매스컴이나 문헌들이 드러낸 것처럼 그들을 수동적인 소수자로 인식함으로써 무엇을 어떻게 ‘가르쳐야’할지만을 고민하고 있는 것에대해 성찰적 지점을 모색하고자 하는 의도를 가진다. 평생교육은 그녀들의 잠재 능력을 함양하는 것은 물론 그녀들의 존재로 인하여 한국 사회가 어떤 방식의 발전과 성장을 ‘변화’라는 이름으로 이루게 될 것인가, 그것이 가능하기 위해서는 그녀들과 그녀들의 한국 가족들을 위해 어떠한 교육적 개입이 이루어져야 하는가에 대한 고민을 해야 한다. 이러한 맥락에서 본 논문은 한국으로 결혼 형태로 이주해 들어온 여성들의 행위자성을 드러내면서 그녀들을 위한 평생교육적 기획이 목표로 해야 하는 것은 무엇인지를 모색한다. This study reveals the ‘agentship’ of Philippine women who immigrated to Korea through marriage to Korean men. These women’s agentship has been exercising in the process of planning, practicing, and experiencing ‘crossing over the nation boards’ and their marriage to foreign men. Revealing theses women’s agentship is intended to reflect on existing educational projects targeted on them, which recognize these women just as ‘passive minorities’, make an effort to enlighten them, and help them adopt to Korean society. However, authors insist that lifelong education needs to work on marriage-immigrant women with a framework which takes in their ability to bring about ‘changes’ to Korean society. At a micro-level, inter-adaptation should be taking place between these women and her Korean family members, and marriage-immigrant women and Korean society at a macro level as well. Lifelong education is required to intervene with this process and help Korean society re-born into a space where people with diverse cultural backgrounds can lead ‘fair‘ lives among themselves, Philippine women who participated in this study as interviewees provide lifelong educators with insights on how to and what to do with people with diverse cultural backgrounds within Korean society.

      • Induction of apoptotic cell death by synthetic naringenin derivatives in human lung epithelial carcinoma A549 cells

        Although flavonoids, which are both qualitatively and quantitatively one of the largest groups of natural products, exhibit a variety of beneficial health effects, the exact molecular mechanism of the cellular activities is still not fully explained and there currently exists a lack of evidence for any relationship between the structure-activity relationship and apoptosis-inducing activity. In order to determine the importance of the OH group or substitution of the 5 or carbon-7 in the diphenylpropane skeleton of flavonoids, we originally synthesized several modified naringenin derivatives, including 7-O-benzyl naringenin (KUF-1) and 7-0-(MeO-L-Leu-D-Pro-carbonylmethyl) naringenin (KUF-7). Treatment with KUF-1 or KUF-7 resulted in significant apoptosisinducing effects concomitant with chromatin condensation, caspase activation, and intracellular ROS production. Our data indicate that originally synthesized naringenin derivatives, KUF-l and KUF-7 differentially regulate the apoptosis of A549 cells via intracellular ROS production coupled with the concomitant activation of the caspase cascade signaling pathway, thereby implying that hydroxylation or substitution at Carbon-7 is critical for the apoptosis-inducing activity of flavonoids.
