The objective of the present study was to isolate and identify infectious bursal disease viruses (IBDVs) from broiler and layer chickens of outbreaks of infectious bursal disease (IBD) in three districts of Bangladesh. A total of 70 bursal samples were collected from dead broiler (n=40) and layer (n=30) chickens showing specific lesions of IBD from seven commercial poultry farms of three different districts (Mymensingh, Chittagong and Tangail) of Bangladesh during the year 2007. Five representative bursal samples from each farm were used for the isolation of IBDVs using 9-day-old embryonated eggs of seronegative flock of layer birds and for identification the samples were subjected to agar gel immunodiffusion test (AGIDT), immunohistochemistry (IHC) and reverse transcription-polymerase chain reaction (RT-PCR). Out of 35 bursal samples, IBDVs were successfully isolated from 28 (80%) samples. By AGIDT, 32 (91.4%) samples were found positive for IBDV antigen. Results of AGIDT clearly indicated that IBDVs detected in 29 bursal samples of six affected farms were identical to each other but not to IBDVs present in the remaining three samples of another farm. Indirect immunoperoxidase staining of the bursal sections revealed the presence of IBDV antigen in 32 (91.4%) samples and the IBDV antigen was detected mainly in the cortex of the lymphoid follicles of the bursal tissues. In histopathology, cell depletion, atrophy and necrosis were observed in many bursal follicles with severe edema of interfollicular septa. Of the 35 bursal samples, 34 (97.1%) samples generated 254 bp product by RT-PCR. In conclusion, the results of virus isolation and identification by AGIDT, IHC and the analysis of viral genome by RT-PCR confirmed the outbreaks of acute IBD in commercial poultry of Bangladesh. Moreover, histopathological findings and results of AGIDT gave a clear indication that the isolates from six outbreaks were different from classical strain and it seems to be of very virulent strain. On the other hand, the isolates from the other outbreak were similar to the classical strain.

Apparent digestibility coefficients (ADCs) of dry matter, crude protein, crude lipid, energy, essential amino acids, and fatty acids in extruded pellets containing various fish meals were determined for olive flounder (Paralichthys olivaceus). Eight extruded pellet diets were prepared to contain different fish meals (herring fish meal, anchovy fish meal, mackerel fish meal, sardine fish meal-A, sardine fish meal-B, tuna fish meal, pollock fish meal-A, and pollock fish meal-B) designated as HM, AM, MM, SM-A, SM-B, TM, PM-A, and PM-B, respectively. Chromic oxide ($Cr_2O_3$) was used as an inert indicator at a concentration of 0.5 % in the diet. Feces were collected from triplicate groups of fish ($151{\pm}4.0g$) using a fecal collection column attached to the fish rearing tank for 4 weeks. Dry matter ADCs of the MM, SM-A, SM-B, and PM-A diets were higher than those of all the other dietary groups, and the lowest digestibility of dry matter was observed in the PM-B diet. Fish fed the MM, SM-A, and PM-A diets showed significantly higher ADC of protein than those fed the AM, SM-B, TM, and PM-B diets. Lipid ADC of PM-B was significantly lower than that of the other diets. Energy ADCs of fish fed the MM, SM-A, and PM-A diets were significantly higher than those of the other diets. The availability of essential amino acids in the MM, SM-A, and PM-A diets were generally higher than that of the other fish meal diets, while TM showed the lowest values among all the experimental diets. ADCs of fatty acids in the AM, MM, SM-A, and PM-A diets were generally higher than those of fatty acids in the other diets, and the lowest values were recorded for the PM-B diet. These results provide information on the bioavailability of nutrients and energy in various fish meals which can be used to properly formulate practical extruded feeds for olive flounder.

Apparent digestibility coefficients (ADCs) of dry matter, crude protein, crude lipid, nitrogen-free extract, and energy in selected carbohydrate sources including wheat flour (WF), α-potato starch (PS), α-corn starch (CS), Na alginate (AL), dextrin (DEX), and carboxymethyl cellulose (CMC) were determined for olive flounder. The olive flounder averaging 150 ± 8.0 g were held in 300-L tanks at a density of 30 fish per tank. Chromic oxide was used as the inert marker. Feces were collected from the flounder by a fecal collector attached to a fish rearing tank. Apparent dry matter and energy digestibilities of flounder fed WF, PS, CS, and DEX diets were significantly higher than those of fish fed AL and CMC diets. Apparent crude protein digestibility coefficients of flounder fed PS and CS diets were significantly higher than those of fish fed AL, DEX, and CMC diets. Apparent crude lipid and nitrogen-free extract digestibility coefficients of flounder fed PS and DEX diets were significantly higher than those of fish fed WF, CS, AL, and CMC diets. The present findings indicate that PS and DEX could be effectively used as dietary carbohydrate energy compared to WF, CS, AL, and CMC for olive flounder.

Dietary Inclusion of Distillers Dried Grain for Growth of Juvenile Abalone Haliotis discus hannai

A feeding experiment was conducted to determine the effect of dietary distillers dried grain (DDG) on the growth and body composition of juvenile abalone, Haliotis discus hannai. Three diets were formulated to contain 0% (DDG0), 30% DDG (DDG1) replacing wheat flour and 30% DDG (DDG2) replacing fish meal and wheat flour. A commercial feed (CF) was also compared with experimental diets. Three replicate groups of abalone averaging $0.6{\pm}0.01g$ were fed one of the four diets for 14 weeks. The highest survival rate was observed in the abalone fed the DDG2 diet, which was higher than that of abalone fed the DDG0 diet. Weight gain, shell length, shell width and soft body weight of the abalone were not affected by dietary DDG and CF diet (P > 0.05). Proximate and amino acid composition of the soft whole body were not affected by dietary DDG and CF diet (P > 0.05). The result of this experiment suggests that DDG is a good ingredient to replace fish meal and wheat flour in the diet and could be used up to 30% in the diet without negative effects on the growth performance of juvenile abalone Haliotis discus hannai.

This study was designed to investigate the effects of dietary astaxanthin levels on growth performance, feed utilization, muscle pigmentation, and antioxidant capacity in juvenile rainbow trout. Four experimental diets were formulated to contain 0, 50, 75, and 100 mg/kg astaxanthin (designed as AX0, AX50, AX75, and AX100). Each diet was fed to triplicate groups of fish (18.5 g/fish) for 10 weeks. Growth performance and muscle composition of fish were not affected by dietary astaxanthin levels. Total carotenoid concentration in the muscle of fish fed the AX50 diet was higher than that of fish fed the AX0 diet, but no significant differences were observed between these fish and those fed the AX75 and AX100 diets. Muscle astaxanthin content increased with increased astaxanthin in the diet. Deposition of astaxanthin in the flesh resulted in a decrease in lightness and an increase in redness and yellowness. The fillets from trout fed the AX75 diet had significantly lower lightness than trout fed the AX50 and AX100 diets. Fish fed the AX50 and AX75 diets showed significantly lower catalase activity than those fed the control diet. Total antioxidant status increased significantly in all astaxanthin supplemented groups when compared to the control group. Superoxide dismutase activity was significantly decreased in fish fed the AX50 diet compared to fish fed the AX0 diet. These findings suggest that while fillet pigmentation increased with increasing dietary astaxanthin concentration, indices of fish antioxidant capacity may not be affected in a dose dependent manner.
