        XGB-Northern Goshawk Optimization: Predicting the Compressive Strength of Self-Compacting Concrete

        The contributions of this study, which include enhanced predictive precision and the integration of supplementary admixtures, may be effectively used in real-world scenarios to optimize self-compacting concrete (SCC) mixes for particular applications. This technology exhibits the capacity to improve construction productivity, minimize material wastage, and foster the development of resilient and environmentally friendly infrastructures. This research examined radial basis function (RBF) network and extreme gradient boosting (XGB-based models for predicting the compressive strength (Cs) of SCC. A dataset was generated through the collection of experimental samples, which encompassed supplementary admixtures in addition to the conventional constituents of concrete comprised of lime powders, fly ash, granulated blast furnace slag, silica fume, steel slag powder, super-plasticizer, and viscosity-modifying admixtures. In the present study, two optimization algorithms named the northern Goshawk optimization algorithm (NGOA), and Henry gas solubility optimization (HGSO) were linked with RBF and XGB models (abbreviated as XGBNG , XGBHG , RBFNG , and RBFHG). The results indicate that each of the four models exhibits a notable degree of precision in their prediction methodologies for Cs. A considered comprehensive metric named OBJ showed that the XGBNG simulation received the slightest value at 0.8062, followed by XGBHG at 1.657, then RBFNG at 2.4891, and last RBFHG by 3.9131.

        Anti-dumping Laws and Oligopolistic Trade

        We study the effect of anti-dumping laws in a differentiated products quantity-setting oligopoly. Dumping may or may not occur in the model and may or may not be reciprocal. We show that the effect of adopting an anti-dumping policy on the welfare of the importing country is ambiguous. It can even lead to an increase in the consumers` surplus in the importing country. Hence the importing country may in some cases find strong reasons for the adoption of an anti-dumping policy. We then study the endogenous determination of the equilibrium anti-dumping policies in a two-stage game with reciprocal dumping in which the two governments simultaneously choose anti-dumping policies in a first stage. We provide a sufficient condition, relating the degree of substitutability in demand between the two goods and transport cost, in order for each country to have an incentive to deviate unilaterally from free trade in the presence of anti-dumping laws. We show that governments acting cooperatively to maximize world welfare should choose to endorse the institution of antidumping laws. (JEL Classification: F12, F13, L13)

      • Down-regulation of Protease-activated Receptor 4 in Lung Adenocarcinoma is Associated with a More Aggressive Phenotype

        The role of protease-activated receptors (PARs) in lung tumors is controversial. Although PAR4 is preferentially expressed in human lung tissues, its possible significance in lung cancer has not been defined. The studies reported herein used a combination of clinical observations and molecular methods. Surgically resected lung adenocarcinomas and associated adjacent normal lung tissues were collected and BEAS-2B and NCI-H157 cell lines were grown in tissue culture. PAR4 expression was evaluated by RT-PCR, RT-qPCR, Western blotting and immunohistochemistry analysis. The results showed that PAR4 mRNA expression was generally decreased in lung adenocarcinoma tissues as compared with matched noncancerous tissues (67.7%) and was associated with poor differentiation (p=0.017) and metastasis (p=0.04). Western blotting and immunohistochemical analysis also showed that PAR4 protein levels were mostly decreased in lung adenocarcinoma tissues (61.3%), and were also associated with poor differentiation (p=0.035) and clinical stage (p=0.027). Moreover, PAR4 expression was decreased in NCI-H157 cells as compared with BEAS-2B cells. In conclusion, PAR4 expression is significantly decreased in lung adenocarcinoma, and down-regulation of PAR4 is associated with a more clinically aggressive phenotype. PAR4 may acts as a tumor suppressor in lung adenocarcinoma.

      • Up-regulation of Thy-1 Promotes Invasion and Metastasis of Hepatocarcinomas

        Increasing evidence has revealed that thy-1 was a potential stem cell marker of liver cancer, but no data have been shown on how thy-1 regulates the pathophysiology of liver cancer, such as proliferation, apoptosis, invasion and migration. We previously demonstrated that thy-1 was expressed in about 1% of hepg2 cells, thy-1+hepg2 cells, but not thy-1-, demonstrating high tumorigenesis on inoculation $0.5{\times}10^5$ cells per BACA/LA mouse after 2 months. In the present study, our results showed that higher expression of thy-1 occurs in 72% (36/50 cases) of neoplastic hepatic tissues as compared to 40% (20/50 cases) of control tissues, and the expression of thy-1 is higher in poorly differentiated liver tumors than in the well-differentiated ones. In addition, thy-1 expression was detected in 85% of blood samples from liver cancer patients, but none in normal subjects or patients with cirrhosis or hepatitis. There was a significant negative correlation between thy-1expression and E-cadherin expression (a marker of invasion and migraton), but not between thy-1 expression and AFP expression in all the liver cancer and blood samples. We further investigated the relationship between thy-1 and E-cadherin in liver cancer hepg2 cell line which was transfected with pReceiver-M29/thy-1 eukaryotic expression vector followed by aspirin treatment. Lower expression of E-cadherin but higher expressions of thy-1 were detected in hepg2 cells transfected with pReceiver-M29/thy-1. Taken together, our study suggested that thy-1 probably regulates liver cancer invasion and migration.

        Acetate attenuates inflammasome activation through GPR43-mediated Ca2+-dependent NLRP3 ubiquitination

        Acetate has been indicated to be elevated and to regulate inflammation in inflammatory and metabolic diseases. The inflammasome serves as a key component of immune homeostasis, and its dysregulation can lead to various inflammatory disorders. However, little is known about the effects of acetate on inflammasome activation and the underlying mechanism. Here, we demonstrate that acetate attenuates inflammasome activation via GPR43 in a Ca2+-dependent manner. Through binding to GPR43, acetate activates the Gq/11 subunit and subsequent phospholipase C-IP3 signaling to decrease Ca2+ mobilization. In addition, acetate activates soluble adenylyl cyclase (sAC), promotes NLRP3 inflammasome ubiquitination by PKA, and ultimately induces NLRP3 degradation through autophagy. In vivo, acetate protects mice from NLRP3 inflammasome-dependent peritonitis and LPS-induced endotoxemia. Collectively, our research demonstrates that acetate regulates the NLRP3 inflammasome via GPR43 and Ca2+-dependent mechanisms, which reveals the mechanism of metabolite-mediated NLRP3 inflammasome attenuation and highlights acetate as a possible therapeutic strategy for NLRP3 inflammasome-related diseases.

        Mapping the Most Massive Overdensities through Hydrogen (MAMMOTH). II. Discovery of the Extremely Massive Overdensity BOSS1441 at<i>z</i>= 2.32

        <P>Cosmological simulations suggest a strong correlation between high optical-depth Lya absorbers, which arise from the intergalactic medium, and 3D mass overdensities on scales of 10-30 h(-1) comoving Mpc. By examining the absorption spectra of similar to 80,000 QSO sight lines over a volume of 0.1 Gpc(3) in the Sloan Digital Sky Survey III (SDSS-III), we have identified an extreme overdensity, BOSS1441, which contains a rare group of strong Lya absorbers at z = 2.32 +/- 0.02. This absorber group is associated with six QSOs at the same redshift on a 30 comoving Mpc scale. Using Mayall/MOSAIC narrowband and broadband imaging, we detect Lya emitters (LAEs) down to 0.7 x L-Ly alpha ' * a and reveal a large-scale structure of LAEs in this field. Our follow-up LBT observations have spectroscopically confirmed 20 galaxies in the density peak. We show that BOSS1441 has an LAE overdensity of 10.8 +/- 2.6 on a 15 comoving Mpc scale, which could collapse to a massive cluster with M greater than or similar to 10(15) M-circle dot at z similar to 0. This overdensity is among the most massive large-scale structures at z similar to 2 discovered to date.</P>

      • Amycolatopsis jiangsuensis sp. nov., a novel endophytic actinomycete isolated from a coastal plant in Jiangsu, China.

        <P>A novel actinomycete, designated strain KLBMP 1262(T), was isolated from a coastal plant Dendranthema indicum (Linn.) Des Moul collected from the coastal region of Nantong, Jiangsu Province, in east China and was studied in detail for its taxonomic position. Phylogenetic analysis based on the 16S rRNA gene sequence revealed that strain KLBMP 1262(T) is a member of the genus Amycolatopsis. The 16S rRNA gene sequence similarity indicated that strain KLBMP 1262(T) is most closely related to Amycolatopsis sulphurea DSM 46092(T) (97.96 %), Amycolatopsis ultiminotia RP-AC36(T) (97.50 %) and Amycolatopsis jejuensis N7-3(T) (97.44 %); similarity to other type strains of the genus Amycolatopsis was less than 97.0 %. The organism was determined to have chemical and morphological features consistent with its classification in the genus Amycolatopsis such as meso-diaminopimelic acid as the diagnostic diamino acid in the cell wall peptidoglycan and arabinose and galactose as the diagnostic sugars. The predominant menaquinone was determined to be MK-9 (H(4)). The polar lipids detected were phosphatidylmethylethanolamine, phosphatidylethanolamine, an unknown aminophospholipid, two unknown glycolipids and several unknown lipids. The major fatty acids were found to be C(16:0), iso-C(16:0) and iso-C(15:0). DNA-DNA relatedness data, together with phenotypic differences, clearly distinguished the isolate from its closest relatives. On the basis of these phenotypic and genotypic data, the isolate is considered to represent a novel species, for which the name A. jiangsuensis sp. nov. is proposed. The type strain is KLBMP 1262(T) (=KCTC 19885 (T) = NBRC 108679(T)).</P>

        Discovery of an Enormous Ly<i>α</i>Nebula in a Massive Galaxy Overdensity at<i>z</i>= 2.3

        <P>Enormous Ly alpha nebulae (ELANe), unique tracers of galaxy density peaks, are predicted to lie at the nodes and intersections of cosmic filamentary structures. Previous successful searches for ELANe have focused on wide-field narrowband surveys or have targeted known sources such as ultraluminous quasi-stellar objects (QSOs) or radio galaxies. Utilizing groups of coherently strong Ly alpha absorptions, we have developed a new method to identify high-redshift galaxy overdensities and have identified an extremely massive overdensity, BOSS1441, at z = 2-3. In its density peak, we discover an ELAN that is associated with a relatively faint continuum. To date, this object has the highest diffuse Ly alpha nebular luminosity of L-nebula = 5.1 +/- 0.1 x 10(44) erg s(-1). Above the 2 sigma surface brightness limit of SBLy alpha = 4.8 x 10(-18) erg s(-1) cm(-2) arcsec(-2), this nebula has an end-to-end spatial extent of 442 kpc. This radio-quiet source also has extended C IV lambda 1549 and He II lambda 1640 emission on greater than or similar to 30 kpc scales. Note that the Ly alpha, He II, and C IV emissions all have double-peaked line profiles. Each velocity component has an FWHM of approximate to 700-1000 km s(-1). We argue that this Lya nebula could be powered by shocks due to an active galactic nucleus-driven outflow or photoionization by a strongly obscured source.</P>

        Mechanical Properties and Damage Characteristics of Coal-Rock Combination with Different Dip Angles

        The effect of the dip angle of an interlayer coal seam on rock mass mechanical behavior and damage characteristics is seldom discussed. To address the lack of the existing research, a numerical simulation was conducted using the Particle Flow Code (PFC2D), and its damage to mechanical behavior, acoustic emission (AE) characteristics, and damage characteristics were analyzed. The outcomes exhibit the peak strength and elastic modulus of coal-rock combinations are between that of rock and coal. As the coal inclination angle rises, the elastic modulus and peak strength gradually decrease. AE model experiences the initial growth period, stable growth period, and rapid decline period. The value of AE hits and the generation of the AE signal is affected by the size of the inclination angle. The degree of damage to the coal-rock combination gradually rises as the angle rises. The damage process of the coal and rock composite sample encompasses an initial damage phase, damage stable evolution phase, accelerated damage phase, and a failure phase. The damage values of the stable evolution stage for varying dip angle models are also different. As the inclination angle increases, the damage value gradually increases, while the strain at the same damage value gradually decreases.
