        B3GNT3 acts as a carcinogenic factor in endometrial cancer via facilitating cell growth, invasion and migration through regulating RhoA/RAC1 pathway‑associated markers

        Background Aberrant expression of beta-1,3-N-acetylglucosaminyltransferase-3 (B3GNT3) has been frequently clarifed in various cancers, however, its role in endometrial cancer (EC) has not been assessed in detail. Purpose This study aimed to investigate the biological role of B3GNT3 in EC and simply explored the detailed mechanism. Methods The EC RNA-Seq dataset from TCGA database was applied to evaluate the expression of B3GNT3 and assess its role on prognostic value. HEC-1-A and KLE cell lines of EC were used to perform loss- and gain-of-function B3GNT3 assays respectively. Quantitative real-time PCR (qRT-PCR) and western blot were used to measure the mRNA and protein levels of indicated molecules respectively. Cell counting kit-8, clone formation tests, and Transwell assay served to determine the changes of proliferative, invasive and migratory abilities of EC cells after altering the expression of B3GNT3. Results B3GNT3 was found to be highly expressed in EC tissues compared to normal tissues according to the online public databases, which confrmed by the following qRT-PCR in 3 EC cell lines. Besides, high B3GNT3 expression presented a worse overall survival in EC patients as compared with low B3GNT3 expression group. Furthermore, functional experiments in vitro indicated that B3GNT3 could facilitate the cell growth, invasion and migration. Moreover, we found that downregulation of B3GNT3 signifcantly reduced the expression level of GTP-RhoA and GTP-RAC1, whereas upregulation of B3GNT3 presented the opposite results. Conclusion The results of current study demonstrate that B3GNT3 acts as an oncogene that promotes EC cells growth, invasion and migration possibly through regulating the RhoA/RAC1 signaling pathway-related markers, suggesting that B3GNT3 may be a candidate biomarker for EC therapeutic intervention.

      • Preventive Effect of Hydrazinocurcumin on Carcinogenesis of Diethylnitrosamine-induced Hepatocarcinoma in Male SD Rats

        The purpose of the present study was to evaluate the preventive effects of hydrazinocurcumin (HZC) on diethylnitrosamine (DEN)-induced hepatocarcinogenesis in a male Sprague Dawley (SD) rat model. One hundred and twenty male SD rats used in this study were divided into six groups. Those receiving DEN with curcumin (CUR) or HZC were studied compared with the DEN-alone group. The study demonstrated that DEN induced severe histological and immunohistochemical changes in liver tissues, significantly increasing the levels of liver marker enzymes (alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), ${\gamma}$-glutamyltransferase (GGT) and total bilirubin level (TBL)). The hepatocarcinoma incidences were 100.0%, 36.7% and 20.0% in the DEN-alone, DEN-CUR and DEN-HZC groups, respectively. Although macroscopic and microscopic features suggested that both CUR and HZC were effective in inhibiting DEN-induced hepatocarcinogenesis, HZC was exerted a stronger influence. Immunohistochemical analysis with PCNA demonstrated significantly differences among the groups (all P < 0.05). Taken together, the results suggested application of CUR and HZC could prevent the occurrence of carcinogenesis and HZC may be a more potent compound for prevention of DEN-induced hepatocarcinogenesis in rats than CUR.

        This topic aims to clarify the method of optimizing tooth surface modification (TSM) fitting under thermoelastic lubrication (TEL) conditions to reveal the most popular and concerning mechanism issues in mechanical engineering. A traditional TSM numerical method has played an increasingly important role in our basic understanding of thermoelastic properties (TEPs) under TEL conditions, which is undoubtedly affirmative and effective. This subject involves the TSM fitting of the theoretical tooth surface superimposed structure, and performs 3D and diagonal modification optimization design, obtains the modified surface position and normal vectors, a variety of optimized modification models are established and complex curved surfaces are analyzed to fit the actual Interface thermoelastic contact numerical simulation, which has been further demonstrated and expressed in actual key working conditions and multiple influencing parameters, thereby reducing transmission error and vibration noise in order to improve gear meshing thermoelastic anti-scuffing load-bearing performance (TEASLBP).

        오모궐 잎과 뿌리 메탄올 추출물의 항산화 및 미백 효능

        Blechnum orientale, also known as the centipede fern, is an important medicinal fern in Malaysia, India, and Chinese. The present study was designed to evaluate the antioxidant, anti-melanogenesis activities of an 80% aqueous methanolic extract of Blechnum orientale rhizome(BOR) and leaf (BOL). The antioxidant assays were perforemed using 2,2-diphenyl-1-picrylhydrazyl(DPPH) and 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt(ABTS). The DPPH and ABTS radical scavenging activities of BOR extract were increased in a dose-dependent manner. Both BOR and BOL extracts did not affect human keratinocytes, HaCaT cell viability. Anti-melanogenesis activity was evaluated by inhibiting the mushroom tyrosinase enzymes, as this causes melanin content in murine melanoma, B16F10 cell line. The BOR extract significantly reduced tyrosinase activity and the cellular melanin synthesis in B16F10 cells without cytotoxic effects. Additionally, The BOR extract also significantly decreased the protein level of melanogenesis-related genes such as microphthalmia-associated transcription factor(MITF), tyrosinase-related protein(TRP)-1, and tyrosinase-related protein(TRP)-2. Collectively, these results indicated that cosmetics containing extract of B. orientale were safe and this plant extract could be used as a functional cosmetic material.

        The Study on Respiratory Function, Spirometric Lung Pattern and Fatigue of Elderly in a Facility

        The purpose of this study was to investigate spirometric lung pattern, respiratory function and degree of fatigue by lung function tests and fatigue tests of 39 elderly people in a care facility aged 65 and over. The respiratory function tests were used to the Spirovit SP-1 and fatigue tests were used modified Piper fatigue scale(mPFS). Regarding the respiratory function, the FVC was 1.41±0.36ℓ, the FVC % predicted was 69.10±14.98%, the FEV1 was 1.02±0.31ℓ, the FEV1 % predicted was 63.27±16.05%, the FEV1/FVC was 72.77±13.40%, and the fatigue score was 5.83±1.09. As for the spirometric lung pattern, 19 patients had a restrictive pattern(48.7%), followed by 11 with a mixed pattern(28.2%), 5 with an obstructive pattern( 12.8%), and 4 with a normal pattern(10.3%). Regarding the respiratory function and fatigue by spirometric lung pattern, the FVC and the FVC % predicted of patients with a normal pattern or an obstructive pattern were greater than other groups at a statistically significant level. As for the FEV1, that of patients with a normal pattern was significantly higher than others, and for the FEV1 % predicted, that of patients with a normal pattern or a restrictive pattern was significantly higher(p<.001). Fatigue score by patients with a normal pattern was significantly less than patients of other patterns(p<.001). Therefore, pulmonary physical therapy is considered necessary to improve respiratory function and fatigue degradation of elderly in a facility.

        <P><B>Abstract</B></P> <P>Prompt removal of misfolded membrane proteins and misassembled membrane protein complexes is essential for membrane homeostasis. However, the elimination of these toxic proteins from the hydrophobic membrane environment has high energetic barriers. The transmembrane protein, FtsH, is the only known ATP-dependent protease responsible for this task. The mechanisms by which FtsH recognizes, unfolds, translocates, and proteolyzes its substrates remain unclear. The structure and function of the ATPase and protease domains of FtsH have been previously characterized while the role of the FtsH periplasmic domain has not clearly identified.         Graphene quantum dots as a highly efficient solution-processed charge trapping medium for organic nano-floating gate memory

        <P>A highly efficient solution-processible charge trapping medium is a prerequisite to developing high-performance organic nano-floating gate memory (NFGM) devices. Although several candidates for the charge trapping layer have been proposed for organic memory, a method for significantly increasing the density of stored charges in nanoscale layers remains a considerable challenge. Here, solution-processible graphene quantum dots (GQDs) were prepared by a modified thermal plasma jet method; the GQDs were mostly composed of carbon without any serious oxidation, which was confirmed by x-ray photoelectron spectroscopy. These GQDs have multiple energy levels because of their size distribution, and they can be effectively utilized as charge trapping media for organic NFGM applications. The NFGM device exhibited excellent reversible switching characteristics, with an on/off current ratio greater than 10<SUP>6</SUP>, a stable retention time of 10<SUP>4</SUP> s and reliable cycling endurance over 100 cycles. In particular, we estimated that the GQDs layer trapped ∼7.2?×?10<SUP>12</SUP> cm<SUP>−2</SUP> charges per unit area, which is a much higher density than those of other solution-processible nanomaterials, suggesting that the GQDs layer holds promise as a highly efficient nanoscale charge trapping material.</P>

        Effects of a Self-Stretching Exercise Program on Musculoskeletal Symptom for Care Workers

        This study was carried out to identify how a self-stretching exercise program affects pain for each body area, pain relief and job satisfaction for care workers. 20 of 40 care workers with musculoskeletal symptom were randomly selected and participated a self-stretching exercise program consisting of 15 motions. The intervention was done five times or more per weeks for 8 weeks and 1 session lasted within 15 minutes. 'Musculoskeletal symptom survey table' of the Korea Occupational Safety and Health Agency(KOSHA) and JDI(Job Descriptive Index) was used for pain on the musculoskeletal symptom and job satisfaction. Survey were done twice before and after the program. The result of this study showed that self-stretching exercise program group(SSPG) relieved from pain significantly in the shoulders(p<.01) and lumbar(p<.05), comparing to the non self-stretching exercise program group(NSPG). Although no significant difference on variations in the JDI appeared in SSPG, the significant reduction appeared from the colleague relationship and organization in NSPG(p<.05). SSPG showed the significant increase on variations in JDI from the job and organization comparing to NSPG. Especially, the improvement on satisfaction for the organization was shown(p<.05). Accordingly, the self-stretching exercise program for care workers can be said to positively affect the overall pain relief and increase on the JDI.

      • Disorders of Small and Large Intestine : Colorectal Cancer In Guangdong Province: A Demographic And Anatomic Survey

        Background/Aims: Colorectal cancer is the third leading incidence of malignant tumour in the world and the incidence of colorectal cancer has steadily been increasing in Asia in recently years. The aim of our study is to determine the basic demographic features of patients with colorectal cancer and the anatomic distribution and characteristics of the tumour in Guangdong population. Methods: A review of patients from 1990 to 2004 at five hospitals was conducted, including Peal Triangle Area in Guangdong (Nanfang Hospital and Huizhou Central People`s Hospital), North area of Guangdong (Shaoguan North-Guangdong People`s Hospital), West area of Guangdong (Affiliated Hospital Guangdong Medical Institute) and East area of Guangdong (Meizhou People`s Hospital). Results: Analysis was carried out on 6,501 patents, only 6,488 cases provided age. The age ranged from 5 years old to 91 years old and the mean age of 6,488 cases is 59. 5.2% (340/6,488) of the patients was young CRC patients. The peak incidence was between the ages 61 and 70 years old (29.5%). The mean age increases from 55 years old (1990-1992) to 61 years old (2002-2004) and the proportion of young CRC patients descends from 7.1% to 3.5%. The make to female ratio is 1.5:1 and the ratio increased with age increasing. Of 6,501 lesions, 3,423 (52.7%) were located in rectum and 3,078 (47.3%) in colon, the ratio of rectum cancer to colon cancer is 1.1:1. The proportion of rectum cancer decreased significantly from 74.5% (1990-1992) to 64.9% (2002-2004) and that of the right sides colon cancer increased from 25.5% to 35.1%. In four different areas, the mean age of CRC increasing and East area of Guangdong ranked the fist. There was no significant difference in the ratio of male to female. Conclusions: The demography of colorectal cancer in Guangdong is different from before and further study should be pursued to find the reason.

        Fermentation properties of beer produced from Korean two-row barley or malt (Gwangmaek) supplemented with Korean red ginseng extracts and Bokbunja (Rubus coreanus Miquel) juice

        This study involved the production of specialty lager beers supplemented with Korean red ginseng extracts or Bokbunja (Korean black raspberry, Rubus coreanus Miquel) juice. The effects of the Korean red ginseng extracts or Bokbunja juice on the specific gravity, pH, yeast viability, free amino nitrogen content, reducing sugar content, color, alcohol content, turbidity, and sensory evaluation were evaluated. The alcohol content of the beers containing the extracts or juice were within the standard alcohol amounts (3.63-4.0%, v/v). The pH values of the three samples containing Bokbunja juice were lower than that of the control values. The sensory evaluation showed that the addition of Bokbunja juice was superior to the ginseng extracts, and the optimal addition time was before or after the secondary fermentation. These data indicate that the flavor and odor of the Bokbunja juice are more persistent than that of the ginseng extracts.
