Proper Fhl1 expression as Wnt signaling is required for chondrogenesis of ATDC5 cells

Wnt signaling is well known playing dual roles during chondrogenesis. The overexpression of Fhl1 (Four-and-a-half LIM domain 1) induces myotube formation as a downstream event of Wnt signaling. Because Fhl1 is widely expressed in other mesenchyme-derived tissues, including chondrocytes, we investigated the role of Fhl1 in chondrogenesis and its relationship with Wnt signaling. We found that the expression of Fhl1 can be enhanced by β-catenin and LiCl (Lithium chloride) in chondrogenic ATDC5 cells. Overexpression of Fhl1 as well as canonical Wnt signaling inhibits chondrogenesis of ATDC5 cells. Moreover, shRNA-mediated knockdown of Fhl1 expression also inhibited ATDC5 cell differentiation, and this result is resembled to the mutant phenotype caused by deletion of β-catenin as was described previously. Because the endogenous Fhl1 expression remains constant during the middle and late phases of chondrogenesis, we propose that proper Fhl1 expression is necessary for the chondrogenic differentiation of ATDC5 cells and altered Fhl1 expression serves as an aberrant Wnt signal that impedes chondrogenesis.

Based on their respective antitumor and thrombolytic activities, the superantigen staphylococcal enterotoxin C2 (SEC2) and staphylokinase (Sak) were chosen for the construction of the novel chimeric proteins Sak-linker- SEC2 and SEC2-linker-Sak using a linker composed of nine Ala residues. Both chimeric proteins possessed nearly the same PBMC proliferation stimulating activity and antitumor activity as SEC2 and thrombolytic activity as Sak. Neither the SEC2 or Sak component of each chimeric protein affected the activity of the other component. The results presented in this study provide a possible strategy to prevent and cure tumor thrombus.

This paper reports on a headspace gas chromatographic method (HS-GC) for the determination of residual formaldehyde in formaldehyde related polymer latexes. The method is based on the reaction between formaldehyde and borohydride in a sodium hydroxide solution (1 mol/L), in which formaldehyde is quantitatively converted to methanol within 30 min at 90 8C and then determined by HS-GC. The results showed that the repeatability of the method had a relative standard deviation of less than 5.0%; the limit of quantification (LOQ) was 17.3 mg, and the recovery ranged from 96.2–102%. The present method is simple, rapid, and accurate.
Foreign Currency Derivative and Firm Value: An Australian Study

In Australian context, only a single study done by Nguyen and Faff (2007) has addressed the question of whether the use of derivatives directly affects firm value. They found that the use of derivatives in general, and the use of interest rate derivatives in particular, are negatively related to firm value. However, using a sample of firms from 1999 and 2000, Nguyen and Faff (2007) may pick up noise from an event in March 1999, which saw the disclosure of derivative information become compulsory for the first time in Australia. Owing to the confounding nature of their results, this paper examines the use of foreign currency derivatives in a sample of 106 Australian nonfinancial firms between 2003 and 2008 and its potential impact on firm value. An insignificant relation between firm value (represented by Tobin’s Q) and using foreign currency derivatives is found. The findings of this study contradict the study done by Nguyen and Faff (2007).

Pectinase and naringinase were investigated forimproving the production of pummelo juice by increasingthe juice yield and eliminating juice bitterness. Comparedto a control, the enzymatic treatment significantly (p<0.05)increased the juice yield, soluble pectin and total solublesolid (TSS) contents, and the clarity, while decreasing theconcentrations of the bitter chemicals naringin, limonin,and nomilin. A combined processing treatment of peelingand enzymatic hydrolyses using 5U/g of pectinase and 0.4 U/gof naringinase at 50oC for 60 min resulted in a juice yieldof 42.3%, a TSS content of 11.4oBx, a titrable acidity (TA)of 0.96%, a vitamin C concentration of 38.4 mg/100 mL,and concentrations of naringin, limonin, and nomilin of42.4 μg/mL, 33.5 μg/mL, and 13.3 μg/mL, respectively. The enzymatic method was effective and practical forproduction of high quantity pummelo juice.

Purpose: This study aimed to investigate the prevalence, risk factors, and effects of primary nocturnal enuresis (PNE) on physical and mental health in young adults in mainland China. Methods: An anonymous questionnaire was used to collect information including the sociodemographic characteristics, history of PNE, family history, daytime voiding symptoms, Pittsburgh Sleep Quality Index (PSQI) scores, Self-Esteem Scale (SES), and Self-Rating Depression Scale (SDS). A total of 22,500 university students from 23 provinces and 368 cities in mainland China were included. Results: In total, 21,082 questionnaires were collected, and 20,345 of them qualified for statistical analysis. The PNE prevalence was 1.17%, and the distribution of monosymptomatic nocturnal enuresis (MNE) and nonmonosymptomatic nocturnal enuresis (NMNE) was 66.1% and 33.9%, respectively. In total, 28% of respondents with PNE reported bedwetting daily, 31.6% between 1 and 7 times weekly, and 40.4% between 1 and 4 times monthly; 80% of PNE cases had no history of treatment. The prevalence of PNE in patients with a family history, frequency, urgency, urinary incontinence, and recurrent urinary tract infections was significantly higher than in those without these conditions (P<0.001). PNE was significantly correlated with the PSQI total score (sleep quality) (P=0.011). The SES score was lower and the SDS was higher (P<0.001) in the PNE group than in those without PNE. Conclusions: In mainland China, the PNE prevalence among young adults was found to be high, and PNE had significant effects on physical and mental health. Risk factors included a family history, daytime voiding symptoms, and lack of treatment.
Biological characterization of mesenchymal stem cells from bovine umbilical cord

Mesenchymal stem cells (MSCs) are multi-potential cells that are able to proliferate and differentiate into othercell types. Much research has been done on the MSCs from the umbilical cord (UCMSCs) in human, mice, andavian, but little literature has been published about these cells in big livestock. Here, we choose Luxi cattle asthe experimental animal, we describe an external culture of the UCMSCs from it and summarize the biologicalcharacteristics of these cells, e.g., morphologic appearance, surface antigens, colony-forming ability, geneexpression, and differentiation potential were detected via using immunofluorescence and reverse transcriptionpolymerase chain reaction (RT-PCR). The induced cells, osteoblast, lipoblast, hepatocyte, islet cells, andneurocyte were identified by Alizarin red staining, Oil-red-O staining, Periodic acid-schiff staining, andDithizone staining and RT-PCR detection for specific genes. Results suggest that biological characteristics ofthe UCMSCs were similar to those of MSCs previously analyzed. The primary UCMSCs were sub-cultured topassage 32, the UCMSCs expressed gene CD29, CD44, CD73, CD90, and CD166, induced cells illustratedtypical staining, and expressed specific genes, which indicate that the UCMSCs could be a novel alternativesource of MSCs for experimental and clinical applications.
