Solubility and Conformation of Silk Fibroin Membrane

Transparency and insolubility of eardrum patch against exudates are important for otolaryngological surgery. The author prepared silk fibroin (SF) films with various concentrations and temperature and then examined solubility and conformation of SF films. SF films were transparent regardless of the various preparation conditions. Although most SF films are soluble in 1X PBS solution at 37˚C for 1 h, the SF film with 3.4% with 60˚C was insoluble. Scanning electron microscopy (SEM) showed that the SF films have solid and smooth surface. Fourier transform infrared spectroscopy (FT-IR) and differential scanning calorimetry (DSC) results showed that the conformation of SF films was influenced by the preparation conditions including SF concentration and casting temperature. In conclusion, SF membrane with transparence and insolubility against exudates could be considered as eardrum patch resources.
Silk Fibroin Membrane as Guided Bone Regeneration in Rat Calvarial Defects

Silk fibroin membrane was prepared and examined to know the feasibility of SF membrane as guided bone regeneration. The morphology of silk membrane was flat and smooth surface. The conformation of silk fibroin was β-sheet structure. When the silk membrane was applied on the rat calvarial defect model, it showed significantly higher new bone formation than uncovered control in histomorphometric analysis. The silk membrane was covered by thin fibrotic tissue and there was not observed any inflammatory cells infiltration. In conclusion, silk fibroin membrane could be useful materials for guided bone regeneration.
Degumming Characteristics and Color Stability of GoldenSilk Cocoon

A newly developed silkworm variety, GoldenSilk makes bright yellow cocoon. In this article, the basic characteristics of GoldenSilk cocoon were examined through color analysis, degumming characteristics and UV spectrophotometer. The colorant of Golden- Silk cocoon was not distributed evenly throughout the cocoon layer but existed in the outer layer of the cocoon. The colorant has not good resistance against sunlight. The degumming loss percentage of Golden- Silk cocoon was 29.2%, which is relatively higher than that of the authorized one. UV spectrophotometer showed specific absorption band around 400 to 500 nm, which is the specific yellow colorant of the GoldenSilk cocoon extracted to the degumming solution during the degumming process.
Effects of silk fibroin hydrolysate on bone metabolism in ovariectomized rats

This study aimed to investigate the effects of silk fibroin on bone metabolism in ovariectomized rats. A total of 30 Sprague-Dawley rats were randomized into sham-operated (SHAM), ovariectomized control (OVX), alendronate (OVX+ALEN, 10 mg/kg body weight/d), low silk fibroin (OVX+SF100, 100 mg/kg body weight/d), and high silk fibroin (OVX+SF300, 300 mg/kg body weight/d) groups. All the rats were fed by gavage for 12 wk. At the end of 12 wk, blood and urine were collected for analysis of bone turnover markers, and bone mineral density (BMD) was measured by micro-computed tomography. The results show that the OVX group (p < 0.05) displayed the highest mean body weight gain. Among the five groups, serum levels of bone alkaline phosphatase (ALP) and urine levels of deoxypyridinoline (DPD) were highest in the OVX group (p < 0.05). Bone ALP levels in the ALEN group were significantly lower than that of the silk-treated groups. On the other hand, DPD levels were not significantly different between the ALEN and silk-fibroin-treated groups (p < 0.05). The trabecular BMD was significantly higher in the ALEN and silk-treated groups compared to the OVX group (p < 0.05). In conclusion, this study showed that silk fibroin has similar effects as alendronate, which is used in osteoporosis medication. Therefore silk fibroin might be a new candidate for the prevention and treatment of osteoporosis in patients.

Silkworm pupae, a by-product of the silk industry are known to be valuable resource of nutrients for humans as well as animals besides encompassing diverse bioactive constituents. However, there is a paucity of knowledge on their role in amelioration of oxidative stress and anti-skin aging properties. In the present study, we evaluated the inhibitory effect of aqueous as well as ethanolic (30% and 70%) extracts from distinct stages of male and female silkworm pupae belonging to two silkworm varieties on skin aging-related enzymes. The activities of collagenase, elastase and tyrosinase were effectively inhibited by 70% ethanolic silkworm pupal extracts (SPE), followed by 30% with aqueous extracts exhibiting meager inhibitory potential. SPE were also investigated for their antioxidant activity in oxidative-stressed murine fibroblasts (L929). The intracellular ROS and lipid peroxidation induced by tert-butyl hydroperoxide (t-BHP) in fibroblasts was better attenuated by pre treatment with ethanolic (30%) and aqueous extracts, respectively. The safety of the extracts was determined by studying their effect on fibroblast cell viability and it was found that none of the extracts were cytotoxic. Our findings indicate the potential utility of SPE as anti-aging components in cosmeceuticals.
Antibacterial property of 4-hexylresorcinol incorporated in silk mat after sterilization

When 4-hexylresorcinol (4HR) incorporated silk mat is used for clinical application, sterilization process is essential. In this study, 4HR incorporated silk mat was sterilized with ethylene oxide (EO) gas or autoclave, and then antibacterial property was evaluated. For the examination of micro-structure after the sterilization, scanning electron microscopic (SEM) exam and Fourier Transform Infrared (FT-IR) spectroscopy were also done. There was no gross difference after sterilization in SEM exam and FT-IR spectra. Antibacterial property of 4HR was also maintained after the sterilization. In conclusion, EO gas or autoclave did not damage the antibacterial property of 4HR in the silk mat.
Optimal Condition of Natural Silk 3D Matrix Production by Silkworm Spinning

Silk is appealing materials for many biomedical applications involving tissue engineering and implantable devices, because of its biocompatibility, environmental stability, controlled proteolytic biodegradability and morphologic flexibility. Silk matrix is required for the treatment of a wide wound area, but the present silk matrix is made by the second processing, and thus, the labor and the cost are high. In this work, we investigated the optimal production condition of natural silk 3D matrix using the silkworms and invented Automatic Silk Matrix Making Machine (ASMMM) for natural silk 3D matrix production. As a result, we determined that optimal production condition for making A4 paper size natural silk 3D matrix was used Rough aquarelle paper on surface at $25^{\circ}C$ and 30 silkworm larvae. These results are expected to provide basic data for the efficient production of the natural silk 3D matrix, and it is suggested that the produced natural silk 3D matrix is useful as a medical biomaterials.

Mulberry was dried with low temperature vacuum dryer using microwave. The nutritional compositions of microwave-dried mulberry including proximate composition, sugar content, mineral content, total phenolic, flavonoids, and anthocyanin, beta-carotene, vitamin C, and amino acid composition were measured. Sugar contents of mulberry were 42.6 mg/100g (Cheongilppong) and 43.27 mg/100g dw (Gwasang No. 2). The main components of mulberry sugars were fructose and glucose. Mineral analysis showed that K, P, Ca, and Mg were abundant regardless of mulberry cultivars.
