      • DYNAMIC DESIGN METHODS OF ROCK ENGINEERING

        The key features of an intelligent and dynamic design methodology for rock engineering projects has been introduced and summarized firstly, which include some new functions such as intelligent recognition of mechanical rockmass parameters, strategies to select modeling methods and codes, integrated feedback modeling and information, and technical auditing in rock engineering design process. Then typical examples of applications of the dynamic design methodology in some large slopes, underground powerhouses in China are summarized. The discussions are given for the future of the methodology.

        Evaluation of secondary cytoreduction surgery in platinum-resistant ovarian cancer patients within three-line recurrent: a multicenter, randomized controlled study

        Background: Epithelial ovarian cancer is the leading cause of death among gynecologicalmalignancies. Platinum resistance remains a dilemma and bottleneck in treatment, andsalvage chemotherapy has limited effectiveness. Recently, the role of secondar y cytoreductivesurger y (SCS) in patients with platinum-resistant recurrent ovarian cancer (ROC) has causedattention especially in patients with oligometastases. However, there is neither high-qualityevidence-based evidence nor standardized criteria for selecting SCS for patients withplatinum-resistant ROC until now. Methods: This multicenter, randomized, controlled clinical trial is to evaluate the valueof SCS and to clarif y reliable criteria of utilizing SCS in women with ROC, which is led byGynecologic Oncology Group, Women’s Hospital, Zhejiang University School of Medicine. Recruitment has started on Januar y 1st, 2023, and is scheduled to end in December 2026. One hundred and forty participants with platinum-resistant ROC who meet the “RSCScriteria” will be randomized assigned at a ratio of 1:1 to either the experimental arm or thestandard arm. Patients in the experimental arm will receive SCS followed by non-platinumsingle agent chemotherapy (paclitaxel, gemcitabine or liposomal adriamycin) for at least4 cycles while patients in the standard arm will be provided with only non-platinum singleagent chemotherapy. The primar y outcome is progression-free sur vival. The secondar youtcomes are overall sur vival, adverse events and health-related cancer-specific quality of life.         20(S)-Ginsenoside Rh2 displays efficacy against T-cell acute lymphoblastic leukemia through the PI3K/Akt/mTOR signal pathway

        Background: T-cell acute lymphoblastic leukemia (T-ALL) is a kind of aggressive hematological cancer, and the PI3K/Akt/mTOR signaling pathway is activated in most patients with T-ALL and responsible for poor prognosis. 20(S)-Ginsenoside Rh2 (20(S)-GRh2) is a major active compound extracted from ginseng, which exhibits anti-cancer effects. However, the underlying anticancer mechanisms of 20(S)-GRh2 targeting the PI3K/Akt/mTOR pathway in T-ALL have not been explored. Methods: Cell growth and cell cycle were determined to investigate the effect of 20(S)-GRh2 on ALL cells. PI3K/Akt/mTOR pathway-related proteins were detected in 20(S)-GRh2-treated Jurkat cells by immunoblotting. Antitumor effect of 20(S)-GRh2 against T-ALL was investigated in xenograft mice. The mechanisms of 20(S)-GRh2 against T-ALL were examined by cell proliferation, apoptosis, and autophagy. Results: In the present study, the results showed that 20(S)-GRh2 decreased cell growth and arrested cell cycle at the G1 phase in ALL cells. 20(S)-GRh2 induced apoptosis through enhancing reactive oxygen species generation and upregulating apoptosis-related proteins. 20(S)-GRh2 significantly elevated the levels of pEGFP-LC3 and autophagy-related proteins in Jurkat cells. Furthermore, the PI3K/Akt/mTOR signaling pathway was effectively blocked by 20(S)-GRh2. 20(S)-GRh2 suppressed cell proliferation and promoted apoptosis and autophagy by suppressing the PI3K/Akt/mTOR pathway in Jurkat cells. Finally, 20(S)-GRh2 alleviated symptoms of leukemia and reduced the number of white blood cells and CD3 staining in the spleen of xenograft mice, indicating antitumor effects against T-ALL in vivo. Conclusion: These findings indicate that 20(S)-GRh2 exhibits beneficial effects against T-ALL through the PI3K/Akt/mTOR pathway and could be a natural product of novel target for T-ALL therapy.

      • Mitochondrial DNA Levels in Blood and Tissue Samples from Breast Cancer Patients of Different Stages

        Aims: Alterations in mitochondrial DNA (mtDNA) have been implicated in carcinogenesis and tumor progression. We here evaluated the diagnostic and prognostic potential of mtDNA as a biomarker for breast cancer. Methods: Using multiplex real-time polymerase chain reaction, nuclear DNA (nDNA) and mtDNA levels in serum, buffy coat, tumor, and tumor-adjacent tissue samples from 50 breast cancer patients were determined and assessed for associations with clinicopathological features. To evaluate mtDNA as a biomarker for distinguishing between the four sample types, we created receiver operating characteristic (ROC) curves. Results: The mtDNA levels in buffy coat were significantly lower than in other sample types. Relative to tumor-adjacent tissue, reduced levels of mtDNA were identified in buffy coat and tumor tissue but not in serum. According to ROC curve analysis, mtDNA levels could be used to distinguish between buffy coat and tumor-adjacent tissue samples with good sensitivity (77%) and specificity (83%). Moreover, mtDNA levels in serum and tumor tissue were positively associated with cancer TMN stage. Conclusions: The mtDNA levels in blood samples may represent a promising, non-invasive biomarker in breast cancer patients. Additional, large-scale validation studies are required to establish the potential use of mtDNA levels in the early diagnosis and monitoring of breast cancer.

      • Long Non-coding RNAs are Differentially Expressed in Hepatocellular Carcinoma Cell Lines with Differing Metastatic Potential

        Background: Metastasis is a major reason for poor prognosis in patients with cancer, including hepatocellular carcinoma (HCC). A salient feature is the ability of cancer cells to colonize different organs. Long non-coding RNAs (lncRNAs) play important roles in numerous cellular processes, including metastasis. Materials and Methods: In this study, the lncRNA expression profiles of two HCC cell lines, one with high potential for metastasis to the lung (HCCLM3) and the other to lymph nodes (HCCLYM-H2) were assessed using the Arraystar Human LncRNA Array v2.0, which contains 33,045 lncRNAs and 30,215 mRNAs. Coding-non-coding gene co-expression (CNC) networks were constructed and gene set enrichment analysis (GSEA) was performed to identify lncRNAs with potential functions in organ-specific metastasis. Levels of two representative lncRNAs and one representative mRNA, RP5-1014O16.1, lincRNA-TSPAN8 and TSPAN8, were further detected in HCC cell lines with differing metastasis potential by qRT-PCR. Results: Using microarray data, we identified 1,482 lncRNAs and 1,629 mRNAs that were differentially expressed (${\geq}1.5$ fold-change) between the two HCC cell lines. The most upregulated lncRNAs in H2 were RP11-672F9.1, RP5-1014O16.1, and RP11-501G6.1, while the most downregulated ones were lincRNA-TSPAN8, lincRNA-CALCA, C14orf132, NCRNA00173, and CR613944. The most upregulated mRNAs in H2 were C15orf48, PSG2, and PSG8, while the most downregulated ones were CALCB, CD81, CD24, TSPAN8, and SOST. Among them, lincRNA-TSPAN8 and TSPAN8 were found highly expressed in high lung metastatic potential HCC cells, while lowly expressed in no or low lung metastatic potential HCC cells. RP5-1014O16.1 was highly expressed in high lymphatic metastatic potential HCC cell lines, while lowly expressed in no lymphatic metastatic potential HCC cell lines. Conclusions: We provide the first detailed description of lncRNA expression profiles related to organ-specific metastasis in HCC. We demonstrated that a large number of lncRNAs may play important roles in driving HCC cells to metastasize to different sites; these lncRNAs may provide novel molecular biomarkers and offer a new basis for combating metastasis in HCC cases.

Increase carbon constraints performance indicators: the amount of carbon emissions trading, Carbon assets net profit margin, carbon-income ratio, the ratio of carbon costs, carbon assets turnover, unit profit of carbon emissions, CO2 emissions of Yuan output value, in order to fully assess the financial performance and non-financial performance, enable the enterprise performance evaluation system better and more responsive to the requirements of sustainable development, and more convincing.

      • Prognostic Value of PLCE1 Expression in Upper Gastrointestinal Cancer: a Systematic Review and Meta-analysis

        Background: A number of studies have identified a shared susceptibility locus in phospholipase C epsilon 1 (PLCE1) for esophageal squamous cell carcinoma (ESCC) and gastric cardia adenocarcinomas (GCA). However, the results of PLCE1 expression in esophageal and gastric cancer remain inconsistent and controversial. Moreover, the effects on clinicopathological features remain undetermined. This study aimed to provide a precise quantification of the association between PLCE1 expression and the risk of ESCC and GCA through meta-analysis. Materials and Methods: Eligible studies were identified from PubMed, Wanfang Data, ISI Web of Science, and the Chinese National Knowledge Infrastructure databases. Using RevMan5.2 software, pooled odds ratios (ORs) with 95% confidence intervals (CIs) were employed to assess the association of PLCE1 expression with clinicopathological features relative to ESCC or GCA. Results: Seven articles were identified, including 761 esophageal and gastric cancer cases and 457 controls. Overall, we determined that PLCE1 expression was associated with tumor progression in both esophageal cancers (pooled OR=5.93; 95%CI=3.86 to 9.11) and gastric cancers (pooled OR=9.73; 95%CI=6.46 to 14.7). Moreover, invasion depth (pooled OR=3.62; 95%CI=2.30 to 5.70) and lymph node metastasis (pooled OR=4.21; 95%CI=2.69 to 6.59) were linked with PLCE1 expression in gastric cancer. However, no significant associations were determined between PLCE1 overexpression and the histologic grade, invasion depth, and lymph node metastasis in esophageal cancer. Conclusions: Our metaanalysis results indicated that upregulated PLCE1 is significantly associated with an increased risk of tumor progression in ESCC and GCA. Therefore, PLCE1 expression can be appropriately regarded as a promising biomarker for ESCC and GCA patients.

        A Load Identification Method for ICPT System Utilizing Harmonics

        Online identification of load parameters is the premise of establishing a stable and highly-efficient ICPT (Inductive Coupled Power Transfer) system. However, compared with pure resistive load, precise identification of composite load, such as resistor-inductance load and resistance-capacitance load, is more difficult. This paper proposes a method for detecting the composite load parameters of ICPT system utilizing harmonics. In this system, the fundamental and harmonic wave channel are connected to the high frequency inverter jointly. The load parameter values can be obtained by setting the load equation based on the induced voltage of secondary-side network, the fundamental wave current, as well as the third harmonic current effective value received by the secondary-side current via Fourier decomposition. This method can achieve precise identification of all kinds of load types without interfering the normal energy transmission and it can not only increase the output power, but also obtain higher efficiency compared with the fundamental wave channel alone. The experimental results with the full-bridge LCCL-S type voltage-fed ICPT system have shown that this method is accurate and reliable.

      • An Evolving-Graph-Based Finite State Machine Model for Protocol Conformance Testing in MANETs

        Dynamic network topology is one of the inherent features of Mobile Ad hoc Network (MANET). However, it may lead to unfeasible test sequences in the FSM-based active conformance testing, due to the topological distinction between testing sequences generation and its execution. In this paper, a formal model, named Evolving-Graph- based Finite State Machine (EGFSM), is proposed to help alleviate the impact of the dynamic network on conformance testing in MANETs. The evolving graph theory is first introduced to extend the Finite State Machine model to describe the dynamic behavior of the protocol with time-varying network topology. In the testing scenario with a predictable node movement pattern, test sequences adaptable to the dynamic topological changescan be generated on the basis of the EGFSM for the protocol under testing. A case study based on the AODV routing protocol is conducted, and promising experimental results show that it is a feasible way to test the protocols in MANETs using an EGFSM.
