        Effect of Lactobacillus mucosae on In vitro Rumen Fermentation Characteristics of Dried Brewers Grain, Methane Production and Bacterial Diversity

        The effects of Lactobacillus mucosae (L. mucosae), a potential direct fed microbial previously isolated from the rumen of Korean native goat, on the rumen fermentation profile of brewers grain were evaluated. Fermentation was conducted in serum bottles each containing 1% dry matter (DM) of the test substrate and either no L. mucosae (control), 1% 24 h broth culture of L. mucosae (T1), or 1% inoculation with the cell-free culture supernatant (T2). Each serum bottle was filled anaerobically with 100 mL of buffered rumen fluid and sealed prior to incubation for 0, 6, 12, 24, and 48 h from which fermentation parameters were monitored and the microbial diversity was evaluated. The results revealed that T1 had higher total gas production (65.00 mL) than the control (61.33 mL) and T2 (62.00 mL) (p<0.05) at 48 h. Consequently, T1 had significantly lower pH values (p<0.05) than the other groups at 48 h. Ammonia nitrogen ($NH_3$-N), individual and total volatile fatty acids (VFA) concentration and acetate:propionate ratio were higher in T1 and T2 than the control, but T1 and T2 were comparable for these parameters. Total methane ($CH_4$) production and carbon dioxide ($CO_2$) were highest in T1. The percent DM and organic matter digestibilities were comparable between all groups at all times of incubation. The total bacterial population was significantly higher in T1 (p<0.05) at 24 h, but then decreased to levels comparable to the control and T2 at 48 h. The denaturing gradient gel electrophoresis profile of the total bacterial 16s rRNA showed higher similarity between T1 and T2 at 24 h and between the control and T1 at 48 h. Overall, these results suggest that addition of L. mucosae and cell-free supernatant during the in vitro fermentation of dried brewers grain increases the VFA production, but has no effect on digestibility. The addition of L. mucosae can also increase the total bacterial population, but has no significant effect on the total microbial diversity. However, inoculation of the bacterium may increase $CH_4$ and $CO_2$ in vitro.

        TOPOLOGICAL ANALYSIS OF POWERTRAINS FOR REFUSECOLLECTING VEHICLES BASED ON REAL ROUTES – PART I: HYBRID HYDRAULIC POWERTRAIN

        In this two-part paper, a topological analysis of powertrains for refuse-collecting vehicles (RCVs) based on the simulation of different architectures (internal combustion engine, hybrid electric, and hybrid hydraulic) on real routes is proposed. In this first part, a characterization of a standard route is performed, analyzing the average power consumption and the most frequent working points of an internal combustion engine (ICE) in real routes. This information is used to define alternative powertrain architectures. A hybrid hydraulic powertrain architecture is proposed and modelled. The proposed powertrain model is executed using two different control algorithms, with and without predictive strategies, with data obtained from real routes. A calculation engine (an algorithm which runs the vehicle models on real routes), is presented and used for simulations. This calculation engine has been specifically designed to analyze if the different alternative powertrain delivers the same performance of the original ICE. Finally, the overall performance of the different architectures and control strategies are summarized into a fuel and energy consumption table, which will be used in the second part of this paper to compare with the different architectures based on hybrid electric powertrain. The overall performance of the different architectures indicates that the use of a hybrid hydraulic powertrain with simple control laws can reduce the fuel consumption up to a 14 %.

In this second part, three different hybrid electric powertrain architectures are proposed and modeled. These architectures are based on the use of fuel cells, ultracapacitors, and batteries. A calculation engine, which is specifically designed to estimate energy consumption, respecting the original performance as the original internal combustion engine (ICE), is presented and used for simulations and component sizing. Finally, the overall performance of the different architectures (hybrid hydraulic, taken from the first paper part, and hybrid electric, estimated in this second part) and control strategies are summarized in a fuel and energy consumption table. Based on this table, an analysis of the different architecture performance results is carried out. From this analysis, a technological evolution of these vehicles in the medium- and long terms is proposed.

        Partial Implementation of the Q.F.D. Methodology for the Identification of the Most Important Characteristics and Features of Gymnastics Mats Design

        This research allows us to identify and arrange in order of importance the needs and problems gymnasts and coaches currently find with regard to gymnastic mats. It also helps us to analyze if the different mats standard (E.N. European Standard 12503 and F.I.G. International Gymnastics Federation, 2000) establish any criteria and tests for consider them. To achieve this aim, the methodology Quality Function Deployment (Q.F.D.) was partially implemented, together with a tool called ``User panel``. The results show that the most important features of gymnastic mats are cushioning, shock absorption (related to resilience), deflection (related to aeroelastic behaviour) and surface finish. They must also be easy to clean and portable. When analyzed the existing standards, it can see that several of these considered very important aspects are not tested.

        Evaluation of the relationship between lactacidemia and postoperative complications after surgery for peritoneal carcinomatosis

        BackgroundCytoreductive surgery was developed as a treatment for peritoneal carcinomatosis. However, this surgery is associated with important complications. The present study aimed to assess the relationship between lactacidemia and the rate of associated complications during the immediate postoperative period in the intensive care unit (ICU) in patients undergoing cytoreductive surgery.MethodsThis was a retrospective observational study. A total of 57 patients underwent cytoreductive surgery. All patients were admitted to the ICU immediately after the surgery. Data on lactic acid levels at the time of admission and discharge from the ICU were collected. Postsurgical complications that occurred during the ICU stay were recorded according to failure-to-rescue analysis and their severity stratified according to the Clavien-Dindo classification.ResultsThe lactic acid levels at admission to the ICU were significantly higher in patients who developed complications, with an almost tripled unadjusted relative risk (2.9, 95% CI: 1.6, 5.3), than in those who did not develop complications for the lactacidemia threshold established in the cumulative sum curve graphs. After adjustment for confounding effects, the relative risk became even higher (3.1, 95% CI: 1.8, 3.6). Lactic acid levels were still significantly higher in this group at the time of discharge from the ICU.ConclusionsSerum lactate level is a risk factor for postoperative complications in patients undergoing cytoreductive surgery for peritoneal carcinomatosis. This study suggests that the risk of developing severe complications almost triples with a lactic acid level of 2.5 mmol/L or higher at the time of admission in the ICU.

        Transcriptional Regulation of the AP-1 and Nrf2 Target Gene Sulfiredoxin

        “Two-cysteine” peroxiredoxins are antioxidant enzymes that exert a cytoprotective effect in many models of oxidative stress. However, under highly oxidizing conditions they can be inactivated through hyperoxidation of their peroxidatic active site cysteine residue. Sulfiredoxin can reverse this hyperoxidation, thus reactivating peroxiredoxins. Here we review recent investigations that have shed further light on sulfiredoxin’s role and regulation. Studies have revealed sulfiredoxin to be a dynamically regulated gene whose transcription is induced by a variety of signals and stimuli. Sulfiredoxin expression is regulated by the transcription factor AP-1, which mediates its up-regulation by synaptic activity in neurons, resulting in protection against oxidative stress. Furthermore, sulfiredoxin has been identified as a new member of the family of genes regulated by Nuclear factor erythroid 2-related factor (Nrf2) via a conserved cis-acting antioxidant response element (ARE). As such, sulfiredoxin is likely to contribute to the net antioxidative effect of small molecule activators of Nrf2. As discussed here, the proximal AP-1 site of the sulfiredoxin promoter is embedded within the ARE, as is common with Nrf2 target genes. Other recent studies have shown that sulfiredoxin induction via Nrf2 may form an important part of the protective response to oxidative stress in the lung, preventing peroxiredoxin hyperoxidation and, in certain cases, subsequent degradation. We illustrate here that sulfiredoxin can be rapidly induced in vivo by administration of CDDO-TFEA, a synthetic triterpenoid inducer of endogenous Nrf2, which may offer a way of reversing peroxiredoxin hyperoxidation in vivo following chronic or acute oxidative stress.

        Early Management of Spinal Cord Injury: WFNS Spine Committee Recommendations

        Scientific knowledge today is being generated more rapidly than we can assimilate thus requiring continuous review of gold-standards for diagnosis and treatment of specific pathologies. The aim of this paper is to provide an update on the best early management of spinal cord injury (SCI), in order to produce acceptable worldwide recommendations to standardize clinical practice as much as possible.The WFNS Spine Committee voted recommendations regarding management of SCI based on literature review of the last 10 years. The committee stated 9 recommendations on 3 main topics: (1) clinical assessment and classification of SCI; (2) emergency care and early management; (3) cardiopulmonary management. American Spinal Injury Association impairment scale, Spinal Cord Independence Measure, and International Spinal Cord Injury Basic Pain Data Set are considered the most useful and feasible in emergency evaluation and follow-up in case of SCI. Magnetic resonance imaging is the most indicated examination to evaluate patients with symptomatic SCI. In early phase, correction of hypotension (systolic blood pressure<90 mmHg), and bradycardia are strongly recommended. Surgical decompression should be performed as soon as possible with the ideal surgical time being within 8 hours for both complete and incomplete lesions.
