Expression and Detection of Retinol-Binding Protein-4 Gene of Pig in E. coli

Retinol-Binding Protein-4 (RBP-4) is a low molecular weight lipocalin, which mainly functions as a carrier for vitamin A. Though liver is the main machinery for synthesis of this protein, it is also detectable in other extrahepatic tissues, for example, ovary, uterus, and placenta. Recent evidences have shown that RBP-4 plays important roles in animal reproduction, for example, promoting the development of uterus and embryo. To the best of our knowledge, our laboratory firstly reported that high level of RBP-4 existed in follicular fluid from follicular cysts in sows. Moreover, we have also found that RBP-4 could be secreted by granulosa cells, and RBP-4 receptor was detected in granulosa cells. However, there is no any evidence on the role of RBP-4 in regulating the follicular development. Therefore, cloning and expression of RBP-4 and preparation of polyclonal antibody could help us to explore the role of RBP-4 in follicular development. The aim of this work was to construct prokaryotic expression system of swine RBP-4 gene. The total RNA was extracted from swine’s normal ovarian tissue. The sequence including the whole length of RBP-4 was amplified by RT-PCR and inserted into pEASY-E1.Then transformed into E. coli BL21(DE3)pLysS after gene sequencing. Three hours later, adding IPTG with the final concentration of 1mmol/L and inducing five hours. After centrifugation, the supernatant was discarded. By adding Glucose to Luria-Bertani broth, the expressions of protein were increased. SDS-PAGE showed that the RBP-4 gene expressed in the form of inclusion body with a molecular weight of 21KD. Western-Blot results showed that the target protein could be specifically recognized by mouse anti-human monoclonal antibody. Prokaryotic expression vector of RBP-4 gene was successfully established, and the gene was successfully expressed n E. coli, which is ready for purification and RBP-4 polyclone antibody. Meanwhile, these results were beneficial to investigate the function of RBP-4 in follicular development.

Yellow peach moth, Conogethes punctiferalis(Guenée) (Lepidoptera: Crambidae), is a very destructive fruit borer in Eastern Asia and Australia. A successful artificial diet and rearing protocol are urgently needed for its biological control and research. In this study, we developed and assessed four artificial diets: AD-1, AD-2, AD-3 and AD-4, for rearing yellow peach moth larvae. AD-1 is effective supplements, leading to the larval survival rate of 82.05%, percentage of pupation 79.48%, mean pupa weight of 76.53 mg, emergence rate of 95.25%, and an adult fecundity rate of 121 eggs/female. Using it to feed can shorten the whole generation developmental period to 33 days. Based on this diet AD-1, we also develop a suitable mass rearing method. During the larval stages providing three- to four-time fresh diet can keep a healthy larval colony.
Protection Relay and Fault Information Analyzing System for Smart Grid

For smart grid, the fault information from protection relays and fault recorders is useful for dispatchers to identify the grid fault and resume the power supply fleetly when a fault occurs. Traditionally, EMS/SCADA doesn't have the ability to collect the fault information. An application of protection relay and fault information analyzing system is proposed. In order to implement smarter analyzing functions, three approaches are adopted. First, the system unifies IEC61850 and IEC103 for modeling. Second, an analyzing method of relay characteristic is proposed to evaluate the action behavior of protection relays. Finally, the paper presents an approach to replay the actual action sequence with relay's logic diagram. The system has been carried out and the paper concludes that the system is effective and useful for dispatchers and relay operators in smart grid.

Niemann-Pick type C2 (NPC2) proteins in arthropods have been extensively differentiated and possibly dupli cated according to environmental conditions and are probable to have different functions. The participation of NPC2 proteins in chemical communication in arthropods brings new objectives in environmental-friendly stra tegies for pest population control. In this study, NPC2 gene in Macrocentrus cingulum (McinNPC2) was newly identified by rapid amplification cDNA ends (RACE) technology. McinNPC2 amino acid sequence alignment with other representative NPC2 annotates to evaluate the highly conserved consensus amino acids, but with odorant binding proteins in M. cingulum show that only one consensus amino acid. Primary six-cysteine structures that are same to odorant binding proteins in M. cingulum were observed in McinNPC2. Phylogenetic analysis of McinNPC2 indicated that the nearest monophyletic group forming one clade with high posterior probability values clusters as Cyphomyrmex costatus (CcosNPC2) whereas the nearest evolutionary relation group as some odorant binding proteins. Moreover, quantitative real-time PCR (qPCR) measurements show that the McinNPC2 gene expression level in various tissues of the female is significantly and ubiquitously higher than in male, whereas the highest expression level in female antennae. We further explore the binding characterization of recombinant McinNPC2 to candidate odor molecules and did the modeling and docking simulations. The results showed ligands binding specificity and docking tests results indicate that β-ionone, an aroma compound commonly found in essential oils, can strongly bind with McinNPC2. In conclusion, we proposed that McinNPC2 may be involved in chemical communication and play roles in perception of plant volatiles.
Multi-focus Image Fusion using Fully Convolutional Two-stream Network for Visual Sensors

We propose a deep learning method for multi-focus image fusion. Unlike most existing pixel-level fusion methods, either in spatial domain or in transform domain, our method directly learns an end-to-end fully convolutional two-stream network. The framework maps a pair of different focus images to a clean version, with a chain of convolutional layers, fusion layer and deconvolutional layers. Our deep fusion model has advantages of efficiency and robustness, yet demonstrates state-of-art fusion quality. We explore different parameter settings to achieve trade-offs between performance and speed. Moreover, the experiment results on our training dataset show that our network can achieve good performance with subjective visual perception and objective assessment metrics.
