        Differential Expression of HSP90β in MDA-MB-231 and MCF-7 Cell Lines after Treatment with Doxorubicin

        Background: Breast cancer is a complex, heterogeneous disease and one of the most common malignancies in women worldwide. The efficacy of chemotherapy as an important breast cancer treatment option has been severely limited because of the inherent or acquired resistance of cancer cells. The molecular chaperone heat shock protein 90 (HSP90) upregulated in response to cellular stress is required for functions such as conformational maturation, activation and stability in more than 200 client proteins, mostly of the signaling type. In this study, the expression of HSP90 isoforms including $HSP90{\alpha}$ and $HSP90{\beta}$ in breast cancer cell lines before and after treatment with doxorubicin (DOX) was assessed. Material and Methods: The cell cytotoxicity of DOX in MDA-MB-231 and MCF-7 cell lines was determined using the MTT assay. immunofluorescence and western blotting techniques were used to determine the expression of $HSP90{\beta}$ in the cell lines before and after DOX treatment. Immunofluorescence was also conducted to ascertain the expression of $HSP90{\alpha}$. Results: The MTT assay results showed that the MDA-MB-231 cells ($IC_{50}=14.521{\mu}M$) were more sensitive than the MCF-7 cells ($IC_{50}=16.3315{\mu}M$) to DOX. The immunofluorescence results indicated that the expression of $HSP90{\alpha}$ in both cell lines decreased after exposure to DOX. The western blot and immunofluorescence analyses showed that $HSP90{\beta}$ expression decreased in the MCF-7 cells but increased in the MDA-MB-231 cells after DOX treatment. Conclusion: The obtained results suggested that $HSP90{\alpha}$ and $HSP90{\beta}$ expression levels were reduced in the MCF-7 cells after exposure to DOX. In the MDA-MB-231 cells, $HSP90{\alpha}$ expression was reduced while $HSP90{\beta}$ was found to be overexpressed following DOX treatment.

        Preserving reservoir safety has recognized to be important for the public where a vast majority of dams are located upstream of greatly populated cities and industrialized areas. Buckling, floatation and cavitation have caused failure in the spillway gates and conveyance features during past catastrophic events; showed their vulnerability and need for regular inspection along with reviewing design calculations to ensure the spillway meet current design standards. This paper investigates the hydraulic and structural consideration of dam's spillway by evaluating the data of Karkheh Dam's. Discharge capacity, flood routings and cavitation damage risk were main features for hydraulic considerations where hydrostatic and hydrodynamic forces and stability conditions were considered in structural considerations.

        Model Determination of Delayed Causes of Analgesics Prescription in the Emergency Ward in Arak, Iran

        Background: According to the reports of the World Health Organization 20% of world population suffer from pain and 33% of them suffer to some extent that they cannot live independently. Methods: This is a cross-sectional study which was conducted in the emergency department (ED) of Valiasr Hospital of Arak, Iran, in order to determine the causes of delay in prescription of analgesics and to construct a model for prediction of circumstances that aggravate oligoanalgesia. Data were collected during a period of 7 days. Results: Totally, 952 patients participated in this study. In order to reduce their pain intensity, 392 patients (42%) were treated. Physicians and nurses recorded the intensity of pain for 66.3% and 41.37% of patients, respectively. The mean (SD) of pain intensity according to visual analogue scale (VAS) was 8.7 (1.5) which reached to 4.4 (2.3) thirty minutes after analgesics prescription. Median and mean (SD) of delay time in injection of analgesics after the physician's order were 60.0 and 45.6 (63.35) minutes, respectively. The linear regression model suggested that when the attending physician was male or intern and patient was from rural areas the delay was longer. Conclusions: We propose further studies about analgesics administration based on medical guidelines in the shortest possible time and also to train physicians and nurses about pain assessment methods and analgesic prescription.

        In the present work a novel multifunctional drug nanocarrier was prepared. Polyglycerol-gpolycaprolactonewas grafted on Mesoporous silica-coated magnetic graphene oxide. Doxorubicin, asanticancer drug, was loaded on this carrier. The in vitro drug release showed the controlled pHresponsive behavior. At the endosomal pH (pH 5.5) the amount of drug release enhanced. Thisnanocarrier would have potential application in the tumor therapy owing to its biodegradability,controlled release and pH responsive behavior

      • Well-Organized Mesoporous TiO<sub>2</sub> Photoanode by Using Amphiphilic Graft Copolymer for Efficient Perovskite Solar Cells

        <P>The control of the thickness and porosity of a mesoporous TiO2 layer is important to improve the photovoltaic performance of perovskite solar cells. We produced organized mesoporous TiO2 (om-TiO2) layers using a low-cost amphiphilic graft copolymer, poly(vinyl chloride)-graft-poly(oxyethylene methacrylate) (PVC-g-POEM), as a sacrificial template. This simple but effective synthetic approach generates highly mesoporous and well-organized TiO2 nanostructures with interconnected and size-tunable features. Specifically, the average pore size increased with the amount of hydrophobic PVC main chain in the graft copolymer, which acted as the pore forming agent. Perovskite layers were prepared on top of an om-TiO2 layer according to a two-step sequential deposition: after coating the PbI2, solution in dimethylformamide (DMF) on an om-TiO2 substrate, the substrate was prewetted in isopropyl alcohol (IPA) solvent before immersing into a CH3NH3I/IPA solution. This prewetting treatment not only improves the yields of conversion from PbI2, to CH3NH3PbI3, but also increases the size of perovskite crystals with cuboid morphology. On varying the pore size and the film thickness of the om-TiO2 layer, the device performance attained 11.9% of power conversion efficiency (PCE) at pore size 70 nm and film thickness 300 nm. We measured extracted charge densities and decays of transient photovoltage to understand the kinetics of charge recombination in relation to the corresponding device performance.</P>

        Radioimmunotherapy (RIT) in Brain Tumors

        Annually, the incidence of brain tumors has slightly increased and also the patient prognosis is still disappointing, especially for high-grade neoplasms. So, researchers seek methods to improve therapeutic index as a critical aim of treatment. One of these new challenging methods is radioimmunotherapy (RIT) that involves recruiting a coupling of radionuclide component with monoclonal antibody (mAb) which are targeted against cell surface tumor–related antigens or antigens of cells within the tumor microenvironment. In the context of cancer care, precision medicine is exemplified by RIT; precision medicine can offer a tailored treatment to meet the needs for treatment of brain tumors. This review aims to discuss the molecular targets used in radioimmunotherapy of brain tumors, available and future radioimmunopharmaceutics, clinical trials of radioimmunotherapy in brain neoplasms, and eventually, conclusion and future perspective of application of radioimmunotherapy in neurooncology cancer care.
