        A Two-stage State Transition Algorithm for Constrained Engineering Optimization Problems

        In this study, a state transition algorithm (STA) is investigated into constrained engineering design optimization problems. After an analysis of the advantages and disadvantages of two well-known constraint-handling techniques, penalty function method and feasibility preference method, a two-stage strategy is incorporated into STA, in which, the feasibility preference method is adopted in the early stage of an iteration process whilst it is changed to the penalty function method in the later stage. Then, the proposed STA is used to solve three benchmark problems in engineering design and an optimization problem in power-dispatching control system for the electrochemical process of zinc. The experimental results have shown that the optimal solutions obtained by the proposed method are all superior to those by typical approaches in the literature in terms of both convergency and precision.

        LCL Resonant Compensation of Movable ICPT Systems with a Multi-load

        Compared to LC resonance, LCL resonance has distinct advantages such as a large resonant capability, low voltage and current stresses of the power device, constant voltage or current output characteristics, and fault-tolerance capability. Thus, LCL resonant compensation is employed for a movable Inductive Contactless Power Transfer (ICPT) system with a multi-load in this paper, which achieves constant current output characteristics. Peculiarly, the primary side adopts a much larger compensation inductor than the primary leakage inductor to lower the reactive power, reduce the input current ripple, generate a large current in the primary side, and realize soft-switching. Furthermore, this paper proposes an approximate resonant point for large inductor-ratio LCL resonant compensation through fundamental wave analysis. In addition, the PWM control strategy is used for this system to achieve constant current output characteristics. Finally, an experimental platform is built, whose secondary E-Type coils can ride and move on a primary rail. Simulations and experiments are conducted to verify the effectiveness and accuracy of both the theory and the design method.

        Screening of Anti-Biofilm Compounds from Marine-Derived Fungi and the Effects of Secalonic Acid D on Staphylococcus aureus Biofilm

        Biofilm formation of Staphylococcus aureus is one of its mechanisms of drug resistance. Antibiofilm screening of 106 compounds from marine-derived fungi displayed that 12 compounds inhibited S. aureus biofilm formation by >50% at the concentration of 100 μg/ml, and only secalonic acid D (SAD) and B inhibited by >90% at 6.25 μg/ml without inhibiting cell growth after 24-h incubation. Meanwhile, it was found that the double bond between C-1 and C-10 of citrinin derivatives and the C-C connection position of two chromone monomers may be important for their anti-biofilm activities. Moreover, SAD slightly facilitated biofilm eradication and influenced its architecture. Furthermore, SAD slowed the cell growth rate in the preceding 18-h incubation and differentially regulated transcriptional expression of several genes, such as agr, isaA, icaA, and icaD, associated with biofilm formation in planktonic and biofilm cells, which may be the reason for the anti-biofilm activity of SAD. Finally, SAD acted synergistically against S. aureus growth and biofilm formation with other antibiotics. These findings indicated that various natural products from marine-derived fungi, such as SAD, could be used as a potential biofilm inhibitor against S. aureus.

        Separation of arenols from a low-temperature coal tar by liquid-liquid extraction

        Low-temperature coal tar (LTCT) is a sticky liquid mixture produced mainly from coal pyrolysis, which contains various value-added chemicals (VACs). Liquid-liquid extraction is considered as one of the green and effective ways to explore the organic composition and separate the VACs from LTCT. Herein, petroleum ether, methanol, and carbon disulfide were used to extract arenols from a LTCT. As a result, the relative content and absolute content of arenols extracted from the LTCT are ca. 96.3% and 85.9%, respectively. Among them, p-cresol is predominant, accounting for 22.2%. The isolated contents of arenols are up to 84.6%. Moreover, a kilogram-scale operation was carried out under the same conditions, which offers a potential application in industrial production.

        PREPARATION OF CINOBUFAGIN-LOADED BOVINE SERUM ALBUMIN NANOPARTICLES FOR HEPATOCARCINOMA THERAPY

        Cinobufagin-loaded bovine serum albumin nanoparticles were prepared for treating hepatocellular carcinoma. In this report, cinobufagin-bovine serum albumin-nanoparticles (Cino-BSA-NP) were prepared by an aqueous desolvation process. The physicochemical properties, toxicity, and cancer-related applications of Cino-BSA-NP were investigated. Cino-BSA-NP had a uniform spherical morphology with a particle size in the range of 50–240 nm and an average size of 86.3 nm. The zeta potential of the nanoparticles was -49 mV. The overall embedding ratio was 79.5% and the drug loading was 24.1%. Cino-BSA-NP gave cinobufagin release of up to 53.5% within 3 h, followed by slower controlled release. Cino-BSA-NP inhibited growth of hepatocarcinoma cells in vitro to a similar extent as free cinobufagin, but with a much higher median lethal dose (LD50). Hepatic histomorphological changes indicated that hepatic damage was much less severe with Cino-BSA-NP than with free cinobufagin (2.19 mg/kg). The survival time of nude mice with orthotopic transplantation tumors treated with Cino-BSA-NP was prolonged significantly. The results confirm that Cino-BSA-NP renders cinobufagin completely dispersible in aqueous media, meeting the key requirements for intravenous injection, and show controlled release, thus significantly improving cinobufagin's antitumor activity while reducing its side effects.

      • Involvement of Cdc25c in Cell Cycle Alteration of a Radioresistant Lung Cancer Cell Line Established with Fractionated Ionizing Radiation

        Cancer patients often suffer from local tumor recurrence after radiation therapy. Cell cycling, an intricate sequence of events which guarantees high genomic fidelity, has been suggested to affect DNA damage responses and eventual radioresistant characteristics of cancer cells. Here, we established a radioresistant lung cancer cell line, A549R, by exposing the parental A549 cells to repeated ${\gamma}$-ray irradiation with a total dose of 60 Gy. The radiosensitivity of A549 and A549R was confirmed using colony formation assays. We then focused on examination of the cell cycle distribution between A549 and A549R and found that the proportion of cells in the radioresistant S phase increased, whereas that in the radiosensitive G1 phase decreased. When A549 and A549R cells were exposed to 4 Gy irradiation the total differences in cell cycle redistribution suggested that G2-M cell cycle arrest plays a predominant role in mediating radioresistance. In order to further explore the possible mechanisms behind the cell cycle related radioresistance, we examined the expression of Cdc25 proteins which orchestrate cell cycle transitions. The results showed that expression of Cdc25c increased accompanied by the decrease of Cdc25a and we proposed that the quantity of Cdc25c, rather than activated Cdc25c or Cdc25a, determines the radioresistance of cells.

      • Mini-Array of Multiple Tumor-associated Antigens (TAAs) in the Immunodiagnosis of Esophageal Cancer

        Sera of cancer patients may contain antibodies that react with a unique group of autologous cellular antigens called tumor-associated antigens (TAAs). The present study aimed to determine whether a mini-array of multiple TAAs would enhance antibody detection and be a useful approach in esophageal cancer detection and diagnosis. Our mini-array of multiple TAAs consisted of eleven antigens, p53, pl6, Impl, CyclinB1, C-myc, RalA, p62, Survivin, Koc, CyclinD1 and CyclinE full-length recombinant proteins. Enzyme-linked immunosorbent assays (ELISA) were used to detect autoantibodies against eleven selected TAAs in 174 sera from patients with esophageal cancer, as well as 242 sera from normal individuals. In addition, positive results of ELISA were confirmed by Western blotting. In a parallel screening trial, with the successive addition of antigen to a final total of eleven TAAs, there was a stepwise increase in positive antibody reactions. The eleven TAAs were the best parallel combination, and the sensitivity and specificity in diagnosing esophageal cancer was 75.3% and 81.0%, respectively. The positive and negative predictive values were 74.0% and 82.0%, respectively, indicating that the parallel assay of eleven TAAs raised the diagnostic precision significantly. In addition, the levels of antibodies to seven antigens, comprising p53, Impl, C-myc, RalA, p62, Survivin, and CyclinD1, were significantly different in various stages of esophageal cancer, which showed that autoantibodies may be involved in the pathogenesis and progression of esophageal cancer. All in all, this study further supports our previous hypothesis that a combination of antibodies might acquire higher sensitivity for the diagnosis of certain types of cancer. A customized mini-array of multiple carefully-selected TAAs is able to enhance autoantibody detection in the immunodiagnosis of esophageal cancer and autoantibodies to TAAs might be reference indicators of clinical stage.

      • Three-Port Laparoscopic Exploration is not Sufficient for Patients with T4 Gastric Cancer

        Gastric cancer continues to be a leading cause of cancer death. The majority of patients with gastric adenocarcinoma in China present with advanced disease. Ruling out unresectable cancers from an unnecessary "open" exploration is very important. The aim of this study was to assess the value of five-port anatomical laparoscopic exploration in T4 gastric cancer in comparison with three-port laparoscopic exploration and laparotomy exploration. We conducted a retrospective study on 126 patients with T4 stage scheduled for D2 curative gastrectomy based on computed tomography (CT) staging at Department of Gastric Cancer and Soft Tissue Sarcoma, Fudan University Shanghai Cancer Center, from Apr. 2011 to Apr. 2013. Laparotomy exploration (Group I), three-port laparoscopic exploration (Group II) or five-port anatomical laparoscopic exploration (Group III) were performed prior to radical gastrectomy. Accuracy rate for feasibility of D2 curative gastrectomy in laparotomy exploration and five-port anatomical laparoscopic exploration groups was higher than that in the three-port laparoscopic exploration group. Five-port anatomical laparoscopic exploration group had the highest accuracy resection rate (Group I vs Group II vs Group III,92.6% vs78.6% vs 97.7%; p<0.05) and shorter length of hospitalization (Group I vs Group II vs Group III, $9.58{\pm}4.17$ vs $6.13{\pm}2.85$ vs $5.00{\pm}1.81$; p<0.001). Three-port laparoscopic exploration has low accuracy rate for assessing feasibility of D2 curative gastrectomy and five-port anatomical laparoscopic exploration should be performed on patients with T4 gastric cancer.
