This study was carried out to compare the efficacy and selectivity of TOS and BS media for enumeration of bifidobacteria in commercial fermented milk products. First, bifidobacteria was isolated from 20 fermented milk products, and all isolated bifidobacteria were identified by genomic technology as Bifidobacterium lactis. The two media significantly differed from each other with regard to the recovery of B. lactis, that is, the recovery of this organism was as much as 6 logs lower on BS medium than on TOS. When the concentration of BS solution (mixture of paromomycin sulfate, neomycin, sodium propionate, and lithium chloride) used in BS medium was reduced to 50% (BS50), a relatively high percentage recovery of bifidobacteria from pure cultures was achieved. Susceptibility tests to antibiotics and tests for selective agents for the isolated bifidobacteria and lactic acid bacteria were conducted. The BS solution inhibited some lactic acid bacteria and Bifidobacterium species, while mupirocin (MU) suppressed the growth of all tested lactic acid bacteria but not Bifidobacterium. As compared with BS50 medium, TOS with or without MU showed good bifidobacteria recovery and readily distinguishable colonies; in particular, TOS supplemented with MU had a high selectivity for bifidobacteria. In conclusion, all results suggested that TOS medium with or without MU was found to be suitable for selective enumeration of bifidobacteria from mixed cultures in fermented milk, and better in that capacity than BS medium.

This study was conducted to compare the cellular fatty acid composition and genotypic analysis of Bifulobacteriwn longum MK-G7 originated from Koreans with other commercial type strains of bifidobacteria. The cellular fatty acid of Bif. longum MK-G7 was shown to be composed of C_(16:0 FAME), C_(18:1 cis DMA), C_(18:1 cis 9 FAME), C_(14:0 FAME), C_(19:0 cyc 9, l0DMA), Feature 7(C_(17:2 FAME)), and Feature l0(C_(18.1 c11/t9/t6 FAME)). Bif. longum MK-G7 showed 99.9% homology and the highest relatedness with Bif. longum ATCC 15707 type strain. Both Bif. longum MKG7 and Bif. longum ATCC 15707 showed 153bp products on RAPD (randomly amplified polymorphic DNA) analysis, however, they showed quite different band patterns on PFGE (pulsed-field gel electrophoresis) analysis. Consequently, our present study showed that Biff longum MK-G7 was different from any commercial type strains of Biff longum tested.
Antitumor Activity of Lactobacillus plantarum Cytoplasm on Teratocarcinoma- Bearing Mice

Potential antitumor activity of Lactobacillus plantarum cytoplasm was examined using F9 teratocarcinoma-bearing BALB/c mice. The cytoplasmic fraction of L. plantarum was separated by sonication followed by ultracentrifugation. The fraction at a dose of 100 or 200 mg/kg/day was orally administered for 7 consecutive days before or after tumor inoculation to 16 mice. As a control, heat-killed whole cell was used at a dose of 100 mg/kg/day. Upon oral administrations of both the cytoplasm and heat-killed whole cell, when performed after and before tumor inoculation, the survival of F9-bearing mice prolonged more effectively. Administration of the cytoplasm after tumor inoculaion extended the average survival days by 30 and 40% or daily dosages of 100 and 200 mg/kg/day, respectively. This result suggests that the cytoplasmic fraction of L. plantarum has string antitumor activity against mouse F9 teratocarcinoma in vivo.
