생체 조직에서의 estradiol receptor 양을 간편하고 정확하게 정량하기 위하여 종래의 $^3H$-estradiol을 사용한 dextran coated charcoal 방법을 개선한 결과는 다음과 같다. 새로운 방법으로 쥐 고환 조직내의 estradiol receptor 함량은 23.8 fmol/mg protein으로 비교적 높았고, 근육 조직에서는 0.57 fmol/mg protein으로 비교적 낮게 나타났다. 사람의 유암조직의 estradiol receptor 함량은 유암이 아닌 다른 종양성 환자의 유방 조직에서의 함량보다 높게 나타났다. Scatchard plot 상에서의 free form에 대 한 bound form의 비와 estradiol receptor 농도와의 linearity(r=0.925-0.977, P<0.01) 가 향상되었다. 따라서, 유방암 조직만의 estradiol receptor 양을 정량함으로써 유방암의 진단과 치료에 쉽게 이용할 수 있다. The correlaton between the concentraion of estradiol receptor and the ratio of bound to free form (B/F) showed excellent linearity (r=0.925-0.977, P<0.01) by Scatchard plot with the modified method, therefore these results strongly suggested that the quantitation of estradiol receptor In breast cancer tissue would be useful for the diagnosis and treatment of breast cancer.

조직(組織)에서의 Estradiol Receptor 분석방법(分析方法)에 관한 연구(硏究)

Several methods of estradiol receptor assays in tissue have been proposed; The method of dextran coated charcoal (DCC), sucrose density gradient centrifigation (sedimentation) and hydroxylapatite, etc. Among them, the method of DCC was simply modified in this experiment. The result are as follows; 1. It is required in this model to decrease the amount of (6.7(3)H) estradiol to around one twentieths and to shorten the time of solvent evaporation. Also it can performed on many specimen simultaneously. 2. True bound forms are abtained by use of DES (diethyl styl bestrol) as non specific bound forms in cytosol instead of the blank without cytosol. 3. The correlation between the concentration of estradiol receptor and Boun/Free ratio in Scatchard plot shows the excellent linearity (r=0.925-0.994) 4. Using such a simplified method, estradiol receptor assay in breast tissue can be performed easily and particulary it useful in the diagnosis and treatment of breast cancer by determination of the concentration of estradiol receptors.
당뇨병 환자 혈청에 있어서 Direct-potentiometry와 Flame-photometry에 의한 Na 측정비교

The percentage of flame photometric to direct potentiometric (Na values depends upon water content, hyperprotein, hyperlipid, bicarbonate-Na+ binding etc. We have studied on the relationship between direct potentiometric .and flame photometric measurement of sodium in serum of diabetic patients and normal. The results are as follows; 1. The correlation between direct potentiometric and flame photometric Na measurement in serum of diabetic patients, r is 0.5809. 2. The correlation between direct potentiometric and flame photometric Na value in normal serum, r is 0. 9370. In clinical conditions such as hyperlipemia (diabetic ketoacidosis), the water content of serum is decreased. Under such conditions, direct photentiometric value are more clinically approciate.
HPLC 와 EMIT 를 이용한 혈청에서의 Theophylline 분석에 관한 비교연구

The purpose of this article was to assay theophylline in serum by HPLC and EMIT. HPLC offered some advantage over EMIT: firstly, it was more economic, and secondly, it simultaneously could measure theophylline and its metabolites. The limit of detection of theophylline by EMIT was 2.5 ㎍/㎖, while HPLC could detect 100 ng/㎖ of theophylline. In comparison of the EMIT and HPLC assay of theophylline, EMIT values (2.6-16.1 ㎍/㎖) were higher than HPLC values (1.5-14.9 ㎍/㎖), and the two technique values in 30 samples generally were lower than effective range (10-20 ㎍/㎖).

혈액에서의 theophylline의 HPLC방법과 EMIT방법으로 측정하였다. HPLC방법이 비용이 경제적이고, theophylline과 그 대사물질을 동시에 측정할 수 있었고, 감도도 EMIT법이 $2.5{\mu}g/ml$인데 반해 100 ng/ml 까지 검출 가능하였다. HPLC와 EMIT에 의한 theophylline 측정값은 평균 $7.1{\pm}3.6{\mu}g/ml$과 $8.6{\pm}3.4{\mu}g/ml$로서 대체적으로 낮은 값을 나타냈다. The purpose of this article was to assay theophylline in serum by HPLC and EMIT. Reversed Phase Chromatography를 이용한 혈청에서의 Tricyclic amine 분석에 관한 연구

Tricyclic amine was measured by paired ion liquid chromatography with use of a reversed phase column and UV detection at 254nm. A high separation efficiency can be obtained with a mixture hexane/iso-amyl alcohol. The method proposed here can be used for determining the concentrations of Amitriptyline, Nortriptyline, Imipramine, Desipramine in human serum.
흰쥐 뇌 조직에서의 ^(3)H-imipramine Receptor측정에 관한 연구

In synaptic transmission the catecholamine and it's ³H-imipramine bindings stimulate the adenylate cyclase activity which reflect the change of biological response. In rat medicated with the agonist to activate metabolites, the change of catecholamines and it's ³H-imipramine binding sites in the nerve system were as follows. l. In rat brain tissues, norepinephrine level was 370±10.8 ng/g, epinephrine was 1107.2 ng/g±7.2 ng/g, dopamine was 810±0.26 ng/g, ³H-imipramine receptor was 23.7+3.2 fmol/rm of protein, affinity(Kd) was 3.8±0.261 nmol. 2. In administrated with imipramine to affect as agonist to activate adrenergic receptor in the rats and the depression patients, the plasma concentrations of circulating catecholamine were increased. Such results shows that the low plasma levels of the norepinephrine could be increased in the disease process of depression patients medicated with imipramine as agonist to activate metabolites, and so imipramine medication could be increased the serum levels of norepinephrine and it's 3H-imipramine receptor in the rat brain tissues.
